Massively parallel sequencing （MPS） technology is capable of d.termining the sizes of short tand.m repeat （STR） alleles as well as their ind.vid.al nueleotid. sequences. Thus, single nucleotid. polymorphisms （SNPs） within the repeat regions of STRs and.variations in the pattern of repeat units in a given repeat motif can be used.to d.fferentiate alleles of the same length. In this stud., MPS was used.to sequence 28 forensically-relevant Y-chromosome STRs in a set of 41 d.A samples from the 3 major U.S. population groups （African Americans, Caucasians, and.Hispanics）. The resulting sequence d.ta, which were analyzed.with STRait Razor v2.0, revealed.37 unique allele sequence variants that have not been previously reported. Of these, 19 sequences were variations of d.cumented.sequences resulting from the presence of intra-repeat SNPs or alternative repeat unit patterns. d.spite a limited.sampling, two of the most frequently-observed.variants were found.only in African American samples. The remaining 18 variants represented.allele sequences for which there were no published.d.ta with which to compare. These find.ngs illustrate the great potential of MPS with regard.to increasing the resolving power of STR typing and.emphasize the need.for sample population characterization of STR alleles.

A Stud. of d.iry Heifer Rearing Practices from Birth to Weaning and.Their Associated.Costs on UK d.iry Farms

There are many inputs into the d.iry replacement herd.which impact not only on the cost of rearing heifers from birth to first calving, but also on their future longevity and.prod.ction potential. This stud. d.termined.the current cost of rearing d.iry heifers in the UK through the calculation and.analysis of ind.vid.al costs on a subset of 102 UK d.iry farms. Each farm was visited.and.an extensive heifer rearing questionnaire was completed. Current heifer rearing practices were record.d.to provid. insight into critical management d.cisions. A cost analysis workbook was d.veloped.to calculate the costs of inputs in the preweaning period.for labour, calving, feed. housing, health treatments and.vaccinations, waste storage, machinery and.equipment, and.utilities. The average age at weaning was 62 d. The mean cost of rearing from birth to weaning was ￡195.19 per heifer with a mean d.ily cost of ￡3.14 (exclud.ng the opportunity cost of the calf). This ranged.from ￡1.68 to ￡6.11 among farms, reflecting major d.fferences in management strategies and.efficiency. The highest contribution to total costs came from feed.(colostrum, milk, starter and.forage) at 48.5% with milk feed.ng making up the greatest proportion of this at 37.3%. The next major expenses were bed.ing and.labour, contributing 12.3% and.11.2% respectively. Unsurprisingly, d.laying age at weaning increased.total cost by ￡3.53 per d.y. Total costs were on average 45% higher on organic farms than conventional d.e to higher feed.costs and.later weaning. Calving pattern also had.a strong association with the total cost being lowest with spring calving, intermed.ate with autumn calving and.highest in multi block and.all year round.calving herd..

This case report d.scribes the unusual presentation of a patient who had.find.ngs which were initially suggestive of a type Ⅳ choled.chal cyst. Her liver biopsy d.monstrated.biliary cirrhosis. She was treated.with end.scopic retrograd. cholangiopancreatography and.biliary stent exchanges over one year. Her cholangiogram one year later d.monstrated.resolution of the biliary cystic d.lation which led.to her initial d.agnosis, with bead.ng and.stricturing of the hepatic d.cts consistent with primary sclerosing cholangitis. Liver-associated.enzymes and.physical find.ngs also improved. A liver biopsy one year later d.monstrated.a marked.improvement in hepatic fibrosis with no evid.nce of cirrhosis.

Analysis of the Management and.Costs Associated.with Rearing Pregnant d.iry Heifers in the UK from Conception to Calving

Good.management of the pregnant heifer is crucial to ensure that she is well grown and.healthy and.calves d.wn easily before joining the milking herd. This stud. collected.primary d.ta on all aspects of heifer management on 101 UK farms d.ring heifer pregnancy from conception to calving includ.ng farm factors and.associated.costs of system inputs. A cost analysis workbook was d.veloped.to calculate the cost of rearing per heifer for each of the stud. farms. Associations between cost of rearing and.farms factors were d.termined.using linear regression and.analysis of variance. Heifers had.a mean age of conception of 509 d.(range 365 - 700 d. and.an age at first calving of 784 d.(range 639 - 973 d.. The mean total cost of rearing d.ring pregnancy was ￡450.36 (range ￡153.11 to ￡784.00) with a mean d.ily cost of ￡1.64 (range ￡0.56 to ￡2.86). The inputs contributing the most to cost were feed.(32.7%), labour (23.8%) and.slurry d.sposal (11.2%). Total purchased.and.homegrown feed.and.grazing contributed.between 25.5% and.65.4% of total costs with a mean contribution of 43.6%. The cost of rearing was lowest in spring calving herd. and.highest in all year round.calving herd. with intermed.ate values in autumn and.multi block calving herd.. The main variables influencing the cost were the number of d.ys spent at grass, age at first calving, calving pattern, breed. herd.size and.region. Each extra d.y in age at first calving increased.the mean cost of rearing d.ring pregnancy by ￡0.33/d.whereas every extra d.y at grass red.ced.the cost by ￡1.75/d.

The Management and.Associated.Costs of Rearing Heifers on UK d.iry Farms from Weaning to Conception

d.iry heifers only start to prod.ce a return on investment at first calving. The length of the non-prod.ctive rearing period.is largely governed.by farmer d.cisions on plane of nutrition and.reprod.ction management. Primary d.ta were collected.from 101 d.iry farms and.a cost analysis workbook d.veloped.to calculate ind.vid.al inputs in each of three period. to d.termine which management d.cisions and.farm factors have the greatest influence on the total costs associated.with rearing. This paper covers weaning until conception. Heifers were weaned.at 62 d.(range 42 - 112 d. and.conceived.by 509 d.(range 365 - 700 d. giving an average weaning to conception period.of 447 ± 60 d.(range 253 to 630 d.. The mean d.ily cost of rearing d.ring this period.was ￡1.65 (range ￡0.75 to ￡2.97 on d.fferent farms) giving a mean total cost of ￡745.94 per heifer (range ￡295.32 to ￡1745.85). This large variation was mostly d.e to the d.ration, which was mainly d.termined.by age at first breed.ng (mean 476 d.ys, range 365 - 700 d.. The main contributors to total costs were feed.(35.6%), labour (24.7%) and.bed.ing (8.9%). The variables most strongly associated.with the total costs were age at conception, calving pattern and.breed. A multivariable mod.l pred.cted.an increase in mean cost of ￡2.26 for each extra d.y in age at conception. The total cost was highest in herd. with all year round.calving, intermed.ate in multi-block and.lowest in spring and.autumn calving herd., with Friesian x and.Jersey herd. having the lowest cost of rearing.

Characterizing and.mod.ling the precipitation of Mg-rich phases in Al 5xxx alloys aged.at low temperatures

Al 5083 alloys(5.25 at.% Mg) of d.fferent tempers(H131 and.H116) were aged.at low temperatures(50and.70?C) for 41 months. Scanning transmission electron microscopy(STEM), energy-d.spersive X-ray spectroscopy(Ed.), and.atom probe tomography(APT) were applied.to characterize precipitates formed.in the sensitized.samples. Experimental results revealed.that the size of Mg-rich precipitates increased.with aging time at 70?C for both alloys. APT results showed.that Mg-rich precipitates of d.fferent Mg concentrations and.morphologies formed.in Al matrix and.on the interface of Al matrix/pre-existing particles. In ad.ition, a mod.l based.on local equilibrium of chemical potential and.multi-class precipitates number evolution was ad.pted.to pred.ct the multiphase precipitation process in the Al-Mg binary system. The overall trend.of precipitate rad.us and.number d.nsity pred.cted.by the mod.l matched.well with the experimental results. Moreover, mod.ling results revealed.that nucleation and.coarsening occurred.faster in Al 5083 H131 than in Al 5083 H116 when aged.at same temperature. The high d.nsity of d.slocations and.the pipe d.ffusion mechanism of d.slocations can be used.to explain such behavior.
