Lack of investment for magnetic resonance(MR)fusion systems is an obstacle to deliver targeted prostate biopsies within the prostate cancer diagnostic *** developed a coordinate-based method to support cognitive targeted prostate biopsies and then performed an audit on cancer detection and the location of *** each patient,the prostate is considered as two separate hemiprostates,and each hemiprostate is divided into 4×4×4 *** unit is therefore defined by a three-dimensional *** prospectively applied our coordinates approach to target 106 prostatic lesions in 93 *** 45(of 106;42.5%)lesions positive for cancer,27 lesions(60.0%)harbored clinically significant *** density was significantly higher in patients with proven cancer(median:0.264 ng ml^(-2))when compared to the noncancer group(median:0.145 ng ml-2;P=0.003,Wilcoxon ranksum test).Lesions with Prostate Imaging-Reporting and Data System(PIRADS)score of 5 were found to have a cancer incidence of 65.2%,while PI RADS 4 and 3 lesions have a lower risk of cancer detection,as expected,at 37.3%and 31.3%,*** probability of a lesion being cancerous in our series significantly decreases as we go from the“apex-to-base”dimension(odds ratio[OR]:2.62,95%confidence interval[Cl]:1.55-4.44,P=0.00034).Our analysis also indicates that the probability of cancer decreases as the prostate volume increases(OR:1.03,95%Cl:1.01-1.05,P=0.00327).Based on this feasibility study,the use of coordinates to guide cognitive targeted prostate biopsies warrants future validation study in additional centers.

BACKGROUND Trauma is a major cause of morbidity globally and the sixth leading cause of death,accounting for 10%of all *** genitourinary trauma is estimated for approximately 10%of all patients presenting with trauma,and the kidney is the most injured genitourinary organ ***,there is a paucity of data on genitourinary injury from the Sub-Saharan,and there may be variations from common genitourinary organs injured in developed *** To provide insight on the epidemiology and management of genitourinary trauma in Sub-Saharan Africa with recommendations based on international *** A thorough literature search of genitourinary trauma was conducted using PubMed,Google Scholar and African Journal *** A total of 30 studies from the Sub-Saharan region were eligible for the study and reviewed for epidemiology,biodata,types of injury,mechanisms of injury,treatment and *** evaluating 21904 patients presenting with urological emergencies,approximately 6.6%of cases were due to genitourinary *** commonest injury was urethral 42.9%(22.2-62.2%)followed by injury to the external genitalia(penis,scrotum,testes)25.1%(8.8-67.7%).CONCLUSION Genitourinary injury in Sub-Saharan Africa is underreported,and the presence of more trauma registries,trained urologists and trauma facilities could improve the overall standard of care as well as providing data for research and development in the field.

Urbanization affects vegetation within city administrative boundary and nearby rural *** primary production(GPP)of vegetation in global urban areas is one of important metrics for assessing the impacts of urbanization on terrestrial *** date,very limited data and information on the spatial-temporal dynamics of GPP in the global urban areas are *** this study,we reported the spatial distribution and temporal dynamics of annual GPP during 2000–2016 from 8,182 gridcells(0.5°by 0.5°latitude and longitude)that have various proportion of urban *** 79.3%of these urban gridcells had increasing trends of annual GPP during *** urban area proportion(%)within individual urban gridcells increased,the means of annual GPP trends also *** results suggested that for those urban gridcells,the negative effect of urban expansion(often measured by impervious surfaces)on GPP was to large degree compensated by increased vegetation within the gridcells,mostly driven by urban management and local climate and *** findings on the continued increases of annual GPP in most of urban gridcells shed new insight on the importance of urban areas on terrestrial carbon cycle and the potential of urban management and local climate and environment on improving vegetation in urban areas.

OBJECTIVE The goal of the study was to determine the potential for the KeratinoSens assay to identify electrophile allergens within a botanical extract *** In the KeratinoSens assay,the induction of a luciferase gene under the control of the antioxidant response element(ARE) derived from the human gene AKR1C2 gene is *** parallel,cytotoxicity is assessed by both Neutral Red Uptake(NRU)and MTT *** samples included gluteraldehyde(GA),dimethyl maleate(DM)and cinnamic aldehyde(CA)spiked into four different botanical extracts(including different excipient solvent systems).Test concentrations ranged up to 1 g·L(of complete extract)and a test concentration was considered positive if the relative viability was≥70%and the fold induction of luciferase was 1.5x relative to the solvent *** was measured relative to the EC1.5 of the neat sensitizer as a function of sensitizer concentration and extract *** independent trials were performed on each test *** No appreciable cytotoxicity was observed with any of the *** spiked at 1%to 2%were sufficient to induce a positive up regulation of the ARE below the maximum test material concentration of up to 1 g·*** responses occurred at cell survivals well in excess of 70%.The botanical matrix impacted recovery for both gluteraldehyde and dimethyl maleate *** recovery of the GA spike required at least a -3 fold increase in concentration relative to the chemical alone and botanical extract #3 reduced the activity below *** DM and CA showed activity at about the same effective concentrations as the neat chemical although the DM showed reduced activity in botanical extract #3 as *** These data suggest that the KeratinoSens assay has the potential to identify electrophile allergens within a botanical extract matrix.

Measuring the similarity between two arbitrary crystal structures is a common challenge in crystallography and materials *** there are an infinite number of ways to mathematically relate two crystal structures,only a few are physically *** we introduce both a geometry-based and a symmetry-adapted similarity metric to compare crystal *** crystal symmetry and combinatorial optimization we describe an algorithm to arrive at the structural relationship that minimizes these similarity metrics across all possible maps between any pair of crystal *** approach makes it possible to(i)identify pairs of crystal structures that are identical,(ii)quantitatively measure the similarity between crystal structures,and(iii)find and rank structural transformation pathways between any pair of crystal *** discuss the advantages of using the symmetryadapted cost metric over the geometric ***,we show that all known structural transformation pathways between common crystal structures are recovered with the mapping *** methodology presented in this study will be of value to efforts that seek to catalogue crystal structures,identify structural transformation pathways or prune large first-principles datasets used to parameterize on-lattice Hamiltonians.

Alkaptonuria (AKU) is an inherited disorder of tyrosine metabolism caused by lack of active enzyme homogentisate 1,2-dioxygenase (HGD). The primary consequence of HGD deficiency is increased circulating homogentisic acid (HGA), the main agent in the pathology of AKU disease. Here we report the first metabolomic analysis of AKU homozygous Hgd knockout (Hgd−/−) mice to model the wider metabolic effects of Hgd deletion and the implication for AKU in humans. Untargeted metabolic profiling was performed on urine from Hgd−/− AKU (n = 15) and Hgd+/− non-AKU control (n = 14) mice by liquid chromatography high-resolution time-of-flight mass spectrometry (Experiment 1). The metabolites showing alteration in Hgd−/− were further investigated in AKU mice (n = 18) and patients from the UK National AKU Centre (n = 25) at baseline and after treatment with the HGA-lowering agent nitisinone (Experiment 2). A metabolic flux experiment was carried out after administration of 13C-labelled HGA to Hgd−/−(n = 4) and Hgd+/−(n = 4) mice (Experiment 3) to confirm direct association with HGA. Hgd−/− mice showed the expected increase in HGA, together with unexpected alterations in tyrosine, purine and TCA-cycle pathways. Metabolites with the greatest abundance increases in Hgd−/− were HGA and previously unreported sulfate and glucuronide HGA conjugates, these were decreased in mice and patients on nitisinone and shown to be products from HGA by the 13C-labelled HGA tracer. Our findings reveal that increased HGA in AKU undergoes further metabolism by mainly phase II biotransformations. The data advance our understanding of overall tyrosine metabolism, demonstrating how specific metabolic conditions can elucidate hitherto undiscovered pathways in biochemistry and metabolism.
