奇虾类作为地球历史上最早出现的凶猛巨怪,最大个体可达2米,其身体扁平呈流线型,善于游泳,头部带柄的巨型复眼能够迅速发现猎物;身体前部一对攻击能力很强的大附肢能快速捕杀猎物;此外,具有一排或多排齿的圆形大口能由粗到细的分级肢解猎物。奇虾类的出现标志着寒武纪海洋金字塔式的生态系统的建立,而奇虾类,作为寒武纪海洋的顶级捕食者,当之无愧的位于金字塔的顶端。近些年来,有关奇虾类的报道层出不穷;然而,有关奇虾类的个体发育却鲜有进展。近日,我们发现了一个奇虾类化石Lyr.r.pax幼年个体,该奇虾幼体体长1.8厘米,具有一对带柄的大型复眼和一对攻击能力很强的大附肢,其圆形大口具有带刺的利齿。这些特征表明,奇虾幼体像奇虾成年个体一样,是寒武纪海洋的捕食者,可能捕食与其体型差不多或者比它小型的生物。此外,该研究表明奇虾类个体发育过程极有可能是直接发育,从幼年到成年的生长过程中食性并未改变。更重要的是,该研究表明奇虾类这种顶级捕食者不仅对寒武纪中到大型的生物有威胁,而且小型个体也不例外。这种激烈的生存斗争必将极大的促进动物防御方式的多元化发展,从而将寒武纪大爆发推向高潮。

AIM To investigate the impact of endoscopic ultr.sound-guided fine-needle aspir.tion(EUS-FNA) and positr.n emission tomogr.phy-computed tomogr.phy(PET-CT) in the nodal staging of upper.gastr.intestinal(GI) cancer.in a ter.iar. r.fer.al *** We per.or.ed a r.tr.spective r.view of pr.spectively r.cor.ed data held on all patients with a diagnosis of upper.GI cancer.made between Januar. 2009 and December.2015. Only those patients who had both a PET-CT and EUS with FNA sampling of a mediastinal node distant fr.m the pr.mar. tumour.wer. included. Using a positive EUS-FNA r.sult as the gold standar. for.lymph node involvement, the sensitivity, specificity, positive and negative pr.dictive values(PPV and NPV) and accur.cy of PET-CT in the staging of mediastinal lymph nodes wer. calculated. The impact on ther.peutic str.tegy of adding EUS-FNA to PET-CT was *** One hundr.d and twenty one patients wer. included. Sixty nine patients had a diagnosis of oesophageal adenocar.inoma(Thir.y one of whom wer. junctional), for.y eight had oesophageal squamous cell car.inoma and four.had gastr.c adenocar.inoma. The FNA r.sults wer. inadequate in eleven cases and the PET-CT findings wer. indeter.inate in two cases, ther.for. thir.een patients(10.7%) wer. excluded fr.m fur.her.analysis. Ther. was concor.ance between PET-CT and EUS-FNA findings in seventy one of the r.maining one hundr.d and eight patients(65.7%). The sensitivity, specificity, PPV and NPV values of PET-CT wer. 92.5%, 50%, 52.1% and 91.9% r.spectively. Ther. was discor.ance between PET-CT and EUS-FNA findings in thir.y seven out of one hundr.d and eight patients(34.3%). MDT discussion led to a r.dical tr.atment pathway in twenty seven of these cases, after.the final tumour.stage was alter.d as a dir.ct consequence of the EUS-FNA findings. 目的　建立产生带有gfp报道基因的重组EB病毒株。方法　构建一个有EBVDNABamHⅠ的C和H片段、gfp报道基因和pur.mycin选择基因的真核细胞穿梭质粒 (pGFPUr.EB) ;采用电穿孔法将其转染于B95 8细胞中 ,并用荧光显微镜和流式细胞仪检测gfp的表达 ,PCr.Souther.blot检测重组gfpEB病毒基因。结果　经pur.mycin的长期筛选 ,建立 80 %以上表达gfp的B95 8细胞株 (称gfpB95 8) ,PCr.测有gfp基因 ;经Souther.blot检测证实gfpB95 8可产生重组gfp的EB病毒。结论　构建了gfp报道基因和pur.mycin选择基因的真核细胞穿梭质粒 ;并成功建立产生带有gfp报道基因的重组EB病毒株。

目的 :研究端粒酶r.A模板区特异性核酶对人低分化鼻咽癌CNE 2Z细胞增殖、凋亡的影响。方法 :用电转染的方法将带有绿色荧光蛋白GFP报道系统的端粒酶核酶基因的真核表达质粒PGFPur. telor.7 1及空载质粒PPAT GFP导入人低分化鼻咽癌CNE 2Z细胞 ;检测转染细胞的GFP表达情况、细胞增殖指数及凋亡。结果 :CNE 2ZGTr. 1细胞 (转染目的基因质粒PGFPur. telor.7 1的CNE 2Z细胞 )和CNE 2ZG细胞 (转染空载质粒PPAT GFP的CNE 2Z细胞 )有GFP表达 ,而未转染的CNE 2Z细胞无GFP表达。流式细胞仪检测显示 ,CNE 2ZGTr. 1细胞增殖指数 [(2 5 10 0±0 14 1) % ]明显低于CNE 2Z细胞 ,即未转染的细胞 [(5 3 663± 16 981) % ]和CNE 2ZG细胞 [(61 5 75± 5 166) % ] ,差异有统计学意义 ,P r. 1细胞传第 12代后有凋亡出现 ,CNE 2Z及CNE 2ZG细胞无凋亡。结论 :端粒酶核酶基因的导入使CNE 2Z细胞的增殖能力下降并可诱导CNE 2Z细胞凋亡。端粒酶r.A模板区可以作为鼻咽癌的一个治疗靶点 ,端粒酶r.A模板区特异性核酶可望成为有效的端粒酶抑制剂。
