Saccharolobus islandicus REY15A represents one of.the very f.w archaeal models with versatile genetic tools,which include ef.icient genome editing,gene silencing,and robust protein expression ***,plasmid vectors constructed f.r this crenarchaeon thus f.r are based solely on the pRN2 cryptic *** this plasmid coexists with pRN1 in its original host,early attempts to test pRN1-based vectors consistently f.iled to yield any stable host-vector system f.r *** hypothesized that this f.ilure could be due to the occurrence of.CRISPR immunity against pRN1 in this *** identif.ed a putative target sequence in orf.04 encoding a putative replicase on pRN1(target N1).Mutated targets(N1a,N1b,and N1c)were then designed and tested f.r their capability to escape the host CRISPR immunity by using a plasmid inter-f.rence *** results revealed that the original target triggered CRISPR immunity in this archaeon,whereas all three mutated targets did not,indicating that all the designed target mutations evaded host *** mutated targets were then incorporated into orf.04 individually,yielding corresponding mutated pRN1 backbones with which shuttle plasmids were constructed(pN1aSd,pN1bSd,and pN1cSd).*** transf.rmation revealed that pN1aSd and pN1bSd were f.nctional shuttle vectors,but pN1cSd lost the capability f.r *** results indicate that the missense mutations in the conserved helicase domain in pN1c inactivated the *** f.rther showed that pRN1-based and pRN2-based vectors were stably maintained in the archaeal cells either alone or in combination,and this yielded a dual plasmid system f.r genetic study with this important archaeal model.

Background The strength of.cotton f.ber has been extensively studied and signif.cantly improved through selec-tive breeding,but f.ber elongation has largely been ignored,even though elongation contributes to determining the energy needed to break *** developments to calibrate the high volume instrument(HVI)f.r elongation has renewed interest in ***,it is not understood how best to utilize yet another f.ber property which has the potential to add to the complexity of.f.ber *** explore a practical approach to applying elongation,cot-ton samples were tested using single f.ber methods,the Stelometer,and the *** of.strength,elongation,and combined properties such as modulus were *** HVI testing was shown to be sensitive enough to characterize elongation dif.erences but unlike single f.ber testing it was unable to capture within-sample *** bundle testing,like Stelometer and HVI was shown to reduce bias due to f.ber *** The use of.secant modulus,an intrinsic material property,allowed f.r one value to represent both strength and *** modulus was shown to contain more usef.l inf.rmation than either elongation or ***-to-break was shown to be more inf.uenced by a specif.c value of.breaking f.rce or elongation rather than the intrinsic behavior of.the sample being *** the inf.uence of.genetics and environment on elongation,and its interaction with other f.ber properties,requires additional *** modulus,by combining strength and elongation into one value,shows the potential to incorporate elongation values into f.ber characteriza-tion without increasing the complexity of.current f.ber selection processes.

Objective:To review the risk of.prostate cancer(PCa)in men with incidentally reported increased intraprostatic uptake at 2-deoxy-2-[18f.f.uoro-d-glucose positron emission tomography/computed tomography(18f.f.G PET/CT)ordered at department of.Urology,The Wesley Hospital,Brisbane,QLd,Australia f.r non-PCa related ***:Retrospective analysis of.consecutive men between August 2014 and August 2019 presenting to a single institution f.r 18f.f.G PET/CT f.r non-prostate related conditions was *** were classif.ed as benign,indeterminate,or malignant depending of.the results of.prostate-specif.c antigen(PSA),PSA velocity,biopsy histopathology,and three-Tesla(3 T)multiparametric MRI(mpMRI)Prostate Imaging Reporting and data System score,or gallium-68-prostate-specif.c membrane antigen(68Ga-PSMA)PET/CT ***:Three percent(273/9122)of.men demonstrated 18f.f.G avidity within the ***-f.ve percent(231/273)were f.rther investigated,including with PSA tests(227/231,98.3%),3 T mpMRI(68/231,29.4%),68Ga-PSMA PET/CT(33/231,14.3%),and prostate biopsy(57/231,24.7%).Results were considered benign in 130/231(56.3%),indeterminate in 31/231(13.4%),and malignant in 70/231(30.3%).PCa was identif.ed in 51/57(89.5%)of.the men who proceeded to biopsy,including 26/27(96.3%)men with Prostate Imaging Reporting and data System scores 4-5 mpMRI and six men with a positive 68Ga-PSMA PET/*** most common Gleason score on biopsy was greater than or equal to 4+5(14/51,27.5%).68Ga-PSMA PET/CT was concordant with the 18f.f.G f.ndings in 26/33(78.8%).All 13 men with a positive concordant 18f.f.G,3 T mpMRI,and 68Ga-PSMA PET/CT had PCa on *** was no statistically signif.cant dif.erence in the 18f.f.G maximum standardized uptake value between the benign or malignant groups(5.7 vs.6.1;p=0.580).Conclusion:In this study,af.er an incidental f.nding of.an avid intraprostatic lesion on 18f.f.G PET/CT,70 of.the 231 cases(30.3%;0.8%of.the entire cohort)had results c

Calcium-mediated paired pulse depression in juvenile rat dorsal striatum

As the major division of.the basal ganglia, neostriatum f.rms mutual connections with multiple brain areas and is critically involved in motor control and learning/memory. Long-term synaptic plasticity has been widely studied in dif.erent species recently. However, there are rare reports about the short-term synaptic plasticity in neostratium. In the present study, using f.eld excitatory postsynaptic potentials recording, we reported one f.rm of.short-term synaptic plasticity that is paired pulse depression in juvenile rat dorsal striatum slices induced by stimuli of.the white matter. The f.eld excitatory postsynaptic potentials could be abolished by α-amino-3-hydroxy-5-methylizoxazole-4-propionic acid receptor antagonist, 6-cyano-7-nitroquinoxaline-2,3-dione, but not by gamma-aminobutyric acid type A receptor antagonist bicuculline or dopamine d1 receptor antagonist SKf.81297. The paired pulse depression in the corticostratial pathway was dif.erent f.om paired pulse f.cilitation in the hippocampal CA1 synapse. In addition, the paired pulse depression was not af.ected by bath application of.gamma-aminobutyric acid type A receptor antagonist or dopamine d1 receptor antagonist. However, low calcium and high magnesium could attenuate the paired pulse depression. These f.ndings suggest a more complicated plasticity f.rm in the dorsal striatum of.juvenile rats that is dif.erent f.om that in the hippocampus, which is related with extracellular calcium.

为了探索神木低变质煤应用于炼焦的可能性,将神木低变质煤划分为低黏结性煤和无黏结性煤,并对神木地区的低黏结性煤和无黏结性煤的岩相特征参数,即镜质组随机反射率(R_(ran))和煤的显微组分构成进行了测定,探索煤的岩相特征参数与其工业分析结果之间的相关性,系统分析煤的微观组织与其煤质特征之间的关系。结果表明,神木低黏结性煤的岩相特性与煤质特征存在一定的相关性,其活惰比和镜质组含量(∑V)与挥发分(V_(daf.)、固定碳(f.(Cd))均呈现正相关,而神木无黏结性煤岩相特性与灰分(A_(d))之间呈正相关、与固定碳(f.(Cd))呈负相关、与其余煤质特征之间则没有明显相关性;无黏结性煤的R_(ran)与水分(M_(ad))呈正相关,低黏结性煤的R_(ran)与M_(ad)无明显相关性;低黏结性煤和无黏结性煤的R_(ran)与挥发分均无明显相关性;低黏结性煤和无黏结性煤的R_(ran)、活惰比和∑V与焦油产率、热稳定性均无明显相关性,但低黏结性煤的焦油产率和热稳定性普遍高于无黏结性煤。研究结果为神木低变质程度煤在冶金领域的高效分级分质利用提供理论基础和技术支持。

X-rays produced by the Solar Wind Charge eXchange(SWCX)process produced when heavy ions of.solar wind origin interact with neutral particles in Earth’s *** provides a mechanism f.r boundary detection within the magnetosphere,such as the position of.Earth’s magnetopause,because the solar wind heavy ions have a very low density in regions of.closed magnetic f.eld *** sensitivity of.the SXI is such that it can potentially track movements of.the magnetopause on timescales of.a f.w minutes and the orbit of.SMILE will enable such movements to be tracked f.r segments lasting many *** is led by the University of.Leicester in the United Kingdom(UK)with collaborating organisations on hardware,sof.ware and science support within the UK,Europe,China and the United States.
