The multi-step phosphorelay （MSP） system defines a key signal transduction pathway in plants and many eukaryotes. In this system, external stimuli first lead to the activation of a histidine kinase, followed by transfer of a phosphoryl group from the receiver domain of the kinase （HKRD） to downstream, cytosolic phosphotransfer proteins （HPs）. In order to establish the determinants of specificity for this signaling relay system, we have solved the first crystal structure of a plant HKRD, AHK5RD, in complex with one of its cognate HPs, AHP1. AHP1 binds AHK5RD via a prominent hydrogen bond docking ridge and a hydrophobic patch. These features are conserved among all AHP proteins, but differ significantly from other structurally characterized prokaryotic and eukaryotic HPs. Surface plasmon resonance experiments show that AHK5RD binds to AHP1-3 with similar, micromolar affinity, consistent with the transient nature of this signaling complex. Our correlation of structural and functional data provide the first insight, at the atomic level as well as with quantitative affinity data, into the molecular recognition events governing the MSP in plants.

AIM： To study the role of advanced glycation end products （AGE） and their specific receptor （RAGE） in the pathogenesis of liver fibrogenesis. METHODS： In vitro RAGE expression and extracellular matrix-related gene expression in both rat and human hepatic stellate cells （HSC） were measured after stimulation with the two RAGE ligands, advanced glycation end product-bovine serum albumin （AGE- BSA） and N＇-（carboxymethyl） lysine （CML）-BSA, or with tumor necrosis factor-α （TNF-α）. In vivo RAGE expression was examined in models of hepatic fibrosis induced by bile duct ligation or thioacetamide. The effects of AGE-BSA and CML-BSA on HSC proliferation, signal transduction and profibrogenic gene expression were studied in vitro. RESULTS： In hepatic fibrosis, RAGE expression was enhanced in activated HSC, and also in endothelial cells, inflammatory cells and activated bile duct epithelia. HSC expressed RAGE which was upregulated after stimulation with AGE-BSA, CML-BSA, and TNF-α.RAGE stimulation with AGE-BSA and CML-BSA did not alter HSC proliferation, apoptosis, fibrogenic signal transduction and fibrosis- or fibrolysis-related gene expression, except for marginal upregulation of procollagen α1（ I ） mRNA by AGE-BSA. CONCLUSION： Despite upregulation of RAGE in activated HSC, RAGE stimulation by AGE does not alter their fibrogenic activation. Therefore, RAGE does not contribute directly to hepatic fibrogenesis.

We present a device for passive unidirectional liquid transport. The capillary channels used are bioinspired by the shape of the spermathecae （receptaculum seminis） of rabbit fleas （Spilopsyllus cuniculi） and rat fleas （Xenopsytla cheopis）. The spermatheca is an organ of female fleas that stores sperm until suitable conditions to lay eggs are found. We translated and multiplied the natural form and function of a spermatheca to create a continuous capillary system from which we designed our microfluidic device based directly on the model from nature. Applying the Young-Laplace equation, we derived a theoretical description of local liquid transport, which enables model-guided design. We arranged the bioinspired capillaries in parallel and engraved them in poly（methyl methacrylate）（PMMA） plates by CO2 laser ablation. The fabricated structures transport soapy water passively （i.e., without external energy input） in the forward direction at velocities of about 1 mm＇s i while halting the liquid fronts completely in the backward direction. The bio- inspired capillary channels are capable of unidirectional liquid transport against gravity. Distance and velocity measurements prove the feasibility of the concept. Unidirectional passive liquid transport might be advantageous in technical surfaces for liquid man- agement, for instance, in biomedical microfluidics, lab-on-chip, lubrication, electronics cooling and in micro-analysis devices.

Ectomycorrhizal diversity at five different tree species in forests of the Taunus Mountains in Central Germany

Ectomycorrhizal fungi were investigated on five different forest tree species growing in pure stands on the south slope of the Taunus Mountains, which are situated at the northern end of the Rhine rift valley in Central Germany. Mycorrhizal fungi accompanying the genus Xerocomus were identified and their frequencies counted. Using ITS markers, 22 different fungal species were identified down to species level and 6 down to genus level. On European beech (Fagus sylvatica) 16 fungal species and 4 genera were identified and on Sessile oak (Quercus petraea) 16 ectomycorrhizal species and 2 genera were determined. On both deciduous trees we observed exclusively: Cortinarius subsertipes, Genea hispidula, Lactarius quietus, Tylopilus felleus and a Melanogaster genus. On Norway spruce (Picea abies) we identified 13 different mycorrhizal species and 3 different genera, on Silver fir (Abies alba) 12 species and 3 genera, and in association with European larch (Larix decidua) 11 species and 3 genera. On these conifers Cortinarius anomalus, Lactarius necator and a Piloderma genus occurred exclusively. Comparisons with published data of ectomycorrhizal diversity on the same five tree species, growing in different areas of Germany and Europe, led to the conclusion that there is relative site specificity for ectomycorrhizal communities. Upper soil compartments of the stands investigated in the Taunus Mountainssuffer from soil acidification (pH-H20 ~3.7 to ~4.8). However, a clear correlation between upper soil pH-values and fungal diversity was not observed. On the other hand, nitrate concentrations in upper soil compartments (~26 to ~91 kgNO3-/ha) were higher in older stands as compared to younger ones. Higher nitrate concentrations in upper soils correlated with lower numbers of mycorrhizal individuals.

The interest of Western medicine in Traditional Chinese Medicine(TCM) as a source of drug leads/new drugs to treat diseases without available efficient therapies has been dramatically augmented in the last decades by the extensive work and the outstanding findings achieved within this kind of medicine. The practice of TCM over thousands of years has equipped scientists with substantial experience with hundreds of plants that led to the discovery of artemisinin(qinghaosu), which is extracted from the medicinal plant Artemisia annua L.(qinghao). The unexpected success of artemisinin in combating malaria has drawn strong attention from the scientific community towards TCM. Artemisinin was discovered by Youyou Tu in 1972. Since then, several novel pharmacological activities based on the well-known properties of the sesquiterpene lactone structure with the oxepane ring and an endoperoxide bridge have been unravelled. Beyond malaria, artemisinin and its derivatives(artemisinins) exert profound activities towards other protozoans(Leishmania, Trypanosoma, amoebas, Neospora caninum, and Eimeria tenella), trematodes(Schistosoma, liver flukes), and viruses(human cytomegalovirus, hepatitis B and C viruses). Less clear is the effect against bacteria and fungi. Based on the promising results of artemisinin and the first generation derivatives(artesunate, artemether, arteether), novel drug development strategies have been *** included the synthesis of acetal-and non-acetal-type artemisinin dimeric molecules as well as developing nanotechnological approaches, ***-based liposomes, niosomes, micelles, solid lipid nanocarriers, nanostructured lipid carriers, nanoparticles, fullerenes and nanotubes.
