Toll-Like Receptors as Biomarkers of Gastric Carcinogenesis:Implications for Diagnosis,Prognosis and Treatment

Toll-like receptors (TLR) are essential for Helicobacter pylori (Hp) recognition and subsequent innate and adaptive immunity responses. TLR2 appears to be the receptor responsible for most of the immunologic reaction against Hp infection. However, TLR4, TLR9 and eventually TLR5 may also have a synergic effect with TLR2 against Hp. It has been shown that gastric Hp infection increases TLR expression in the gastric mucosa. Moreover, recent studies have shown that human gastric carcinogenesis is associated not only with increased expression of TLR but also with decreased expression of their inhibitors such as Toll-Interacting Protein (TOLLIP) and peroxisome proliferator-activated receptor (PPAR)-g. Indeed, gastric dysplasia and adenocarcinoma are associated with high expression levels of TLR and low levels of TOLLIP and PPAR-g, suggesting increased activation of these receptors throughout human gastric carcinogenesis. In this article we discuss how these novels findings could be used not only for the diagnosis and prognosis of gastric lesions associated with Hp infection but also for their treatment. Specifically, we discuss the potential use of TLR agonists in addition to antibiotics to improve eradication rates of Hp and of TLR antagonists to slow the progression of gastric preneoplastic lesions. We also discuss the potential value of TLR signalling blockers and quantification of tumoral TLR expression, respectively, in the treatment and prognosis of gastric cancer. In conclusion, TLRs can be an important link between Hp and the sequence of gastric carcinogenesis and they can be used as biomarkers of gastric carcinogenesis. In this article, future lines of investigation related with these novel scientific findings are proposed and discussed.

A detailed Pendulum generators analysis was conducted on various architectures including rotational,pendulum,linear,sliding,cantilever,flexible blade,multidimensional and 1 cm magnetoelectric,and the following hybrid technologies. They enable highly e cient ways to harvest electric energy from many forms of vibrational,rotational,biomechanical,wave,wind and thermal sources,among others. Open-circuit voltages up to 75 V,short-circuit currents up to 60 mA and instantaneous power up to 144 mW were already achieved by these nanogenerators. Their transduction mechanisms,including proposed models to make intelligible the involved physical phenomena,are also overviewed here. A comprehensive analysis was performed to compare their respective construction designs,external excitations and electric outputs. The results highlight the potential of hybrid E-TENGs to convert unused mechanical motion into electric energy for both large-and small-scale ***,this paper proposes future research directions toward optimization of energy conversion e ciency,power management,durability and stability,packaging,energy storage,operation input,research of transduction mechanisms,quantitative standardization,system integration,miniaturization and multi-energy hybrid cells.

In the blossoming field of Cd-free semiconductor quantum dots(QDs),ternary Ⅰ-Ⅲ-VI QDs have received increasing attention due to the ease of the environmentally friendly synthesis of high-quality materials in water,their high photoluminescence(PL)quantum yields(QYs)in the red and near infrared(NIR)region,and their inherently low ***,their oxygen-insensitive long PL lifetimes of up to several hundreds of nanoseconds close a gap for applications exploiting the compound-specific parameter PL *** overcome the lack of reproducible synthetic methodologies and to enable a design-based control of their PL properties,we assessed and modelled the synthesis of high-quality MPA-capped AglnS2/ZnS(AlS/ZnS)*** refined parameters included reaction time,temperature,Ag:In ratio,S:In ratio,Zn:In ratio,MPA:ln ratio,and pH using a design-of-experiment *** for the optimization was provided by mathematical models developed for the application-relevant PL parameters,maximum PL wavelength,QY,and PL lifetime as well as the elemental composition in terms of Ag:ln:Zn *** these experimental data-based models,MPA:ln and Ag:ln ratios and pH values were identified as the most important synthesis parameters for PL control and an insight into the connection of these parameters could be ***,the experimental conditions to synthetize QDs with tunable emission and high QY were *** excellent agreement between the predicted and experimentally found PL features confirmed the reliability of our methodology for the rational design of high quality AlS/ZnS QDs with defined PL *** approach can be straightforwardly extended to other ternary and quaternary QDs and to doped QDs.

Familial hypercholesterolemia (FH) is one of the most prevalent autosomal dominant inherited disorders. Mutations have been found in at least 3 genes: the low-density lipoprotein receptor (LDLR), apolipoprotein B (APOB), and proprotein convertase subtilisin/kexin type 9 (PCSK9). We report on an Azorean family with FH due to a novel mutation in the LDLR gene across three generations. The index-case was first seen at our endocrinology consultation at 12 years old, because of delayed growth and development. Laboratorial investigations revealed a complete failure of the anterior hypophysis due to a congenital malformation of the sella turcica. A total cholesterol of 313 mg/dL (90 - 190 mg/dL) and low-density lipoprotein cholesterol (LDL-C) of 262 mg/dL ( G) causing a premature termination of transcription (stop codon) was identified.

MM： To investigate whether, under the influence of pol ypectomy, the incidence of adenoma decreases with age.METHODS： Consecutive patients with colonic adenomas identified at index colonoscopy were retrospectively selected if they had undergone three or more complete colonoscopies, at least 24 mo apart. Patients who had any first-degree relative with colorectal cancer were excluded. Data regarding number of adenomas at each colonoscopy, their location, size and histological classification were recorded. The monthly incidence density of adenomas after the index examination was estimated for the study population, by using the person-years method. Baseline adenomas were excluded from incidence calculations but their characteristics were correlated with recurrence at follow-up, using the X^42 test.RESULTS： One hundred and fifty-six patients were included （109 male, mean age at index colonoscopy 56.8 ± 10.3 years）, with follow-up that ranged from 48 to 232 mo. No significant correlations were observed between the number, the presence of villous component, or the size of adenomas at index colonoscopy and the presence of adenomas at subsequent colonoscopies （P = 0.49, 0.12 and 0.78, respectively）. The incidence of colonic adenomas was observed to decay from 1.4% person-months at the beginning of the study to values close to 0%, at 12 years after index colonoscopy.CONCLUSION： Our results suggest the sporadic formation of adenomas occurs within a discrete period and that, when these adenomas are removed, all neoplasia-prone clones may be extinguished.

Development and implementation of an automated system to aid laboratory diagnosis using image processing

The objective of this work is to provide an automatic system to count white blood cells in a blood smear. To do so an experiment was assembled, composed by a standard microscope with two step motors coupled to its knobs in order to move the microscope in x and y directions and a web cam which was mounted in the top of the microscope responsible for to acquire images from the smear. The step motors and the web cam are controlled by a microcomputer PC standard via software developed inDelphi. The motors use the parallel port to communicate with the PC and the camera use the USB port. The main idea is to set an initial point into the smear and the automated system will carry over the smear acquiring images (frames with 640 × 480 pixels) and counting the white blood cells encountered. The double histogram threshold technique is implemented to initially exclude the red cells from the image leaving only the white ones. Preliminaries results are obtained and show that the system is quite fast and has a good capacity of selection, even when different kinds of smear are used.

Measuring the NBA Teams’ Cross-Efficiency by DEA Game

In this paper, we use DEA to measure the NBA basketball teams’ efficiency in seasons 2006-2007, 2007-2008, 2008-2009 and 2009-2010. In this context, each team is a DMU;we select the payroll and the average attendance to be the inputs while the wins and the average points per game to be the outputs. First, in order to obtain benchmarks, we measure the DMUs efficiency through classic DEA BCC model with an assurance region for each one of the four seasons individually and together. When we consider the four seasons together, we may analyse whether the performance of each team increases or decreases over time. Next, we evaluate the teams cross efficiency by DEA game to consider that there is no cooperation among DMUs. This approach also improves the efficiencies discrimination.

Mapping of the Water Table Levels of Unconfined Aquifers Using Two Interpolation Methods

The spatial prediction of the water table can be used for many applications related to civil works (foundations, excavations) and other urban and environmental management activities. Deterministic and geostatistical interpolation methods were used to predict the spatial distribution of water table levels (unconfined aquifers) of important geological formations of the joao Pessoa City (capital of Paraiba State, Brazil) with dense urban occupation and high demand for new civil works. The deterministic (topo to raster) and geostatistical (ordinary kriging) interpolation methods were evaluated using a Geographic Information System (GIS)-based investigation. The water table levels were obtained from 276 boring logs of Standard Penetration Test (SPT) in situ investigation distributed over the geological formations studied (an area of 59.8 km2, covering 40 districts of the joao Pessoa City). The Nspt values and textural characterization data are stored for levels of 1 m depth. Some boreholes located in the area investigated were not included in the interpolation processes in order to be compared with estimated values (validation of the results). Maps of the water table depths were also produced to further analyze the quality of the water table surfaces interpolated by both methods. The phreatic surface interpolations provided satisfactory results for both methods (RMSE = 1.8 m). The topo to raster method showed a slight general tendency to be less affected by local values in relation to the kriging method and also has the advantage of integrating the drainage flow system, which is a relevant aspect for spatial models of the water table levels of unconfined aquifers. The ordinary kriging (geostatistical method) provides a prediction surface and some measure of the certainty or accuracy of the predictions.

Association between Helicobacterpylori, Epstein-Barr virus, human papillomavirus and gastric adenocarcinomas

AIM TO correlate Helicobacter pylori （H. pylorli）, EpsteinBarr virus （EBV） and human papillomavirus （HPV） with gastric cancer （GC） cases in Para State, *** Tissue samples were obtained from 302 gastric adenocarcinomas. A rapid urease test was used to detect the presence of H. pylori, and the presence of the cagA gene in the HP-positive samples was confirmed by PCR. An RNA in situ hybridization test designed to complement Eberl RNA was used to detect the presence of EBV in the samples, and the L1 region of HPV was detected using nested PCR. Positive HPV samples were genotyped and analyzed for E6 and E7 viral gene expression. Infections were also correlated with the clinical and pathological characteristics of the *** The majority of the 302 samples analyzed were obtained from men （65%） aged 55 years or older （67%） and were classified as the intestinal subtype （55%）. All three pathogens were found in the samples analyzed in the present study （H. pylori： 87%, EBV： 20%, HPV： 3%）. Overall, 78% of the H. pylori-positive （H. pylorl＋） samples were cagA＋（H. pylori-cagA＋）, and there was an association between the cytotoxic product of this gene and EBV. Coinfections of H. pylori-cagA＋ and EBV were correlated with the most advanced tumor stages. Although only 20% of the tumors were positive for EBV, infection with this virus was associated with distant metastasis. Only the HPV 16 and 18 strains were found in the samples, although no expression of the E6 and E7 oncoproteins was detected. The fundus of the stomach was the region least affected by the *** HPV was not involved in gastric tumorigenesis. Prophy- lactic and therapeutic measures against H. pylori and EBV may prevent the development of GC, especially the more aggressive forms.
