Growth inhibitory effects of Phyllanthus niruri extracts in combination with cisplatin on cancer cell lines

AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in combination with cis- platin on two cancer cell lines. METHoDS： Colorectal carcinoma （HT29） and human hepatocellular carcinoma （HepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDEPN） either alone or in combination with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viability, we stained the cells with prop- idium iodide and assessed them by flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences between groups were determined using analysis of variance and Bonferroni＇s test, as indicated. A value of P 〈 0.05 was considered to be statistically significant. RESULTS： SDEPN had significantly different cyto- toxic effects on HT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and HepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDEPN also had significantly different cytotoxic effects on HT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and HepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. Both cell lines were killed by cisplatin in a dose-dependent manner compared to control cells （HepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and HT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In HT29 cells, pretreat- ment with SDEPN and subsequent treatment with cis-platin resulted in a greater number of cells being killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）. objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIC)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MICs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and LEU255 were present commonly in stabilizing all *** PCA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

Care Technologies in Nursing for People with Colorectal Neoplasia: Integrative Review

objective: Identifying in the literature the care technologies used by nurses for people with colorectal neoplasia on Nietzsche’s conceptual perspective. Method: It consists in an integrative review of the literature held in the database: Latin-American Literature and Caribbean in medical health sciences Literature Analysis and Retrieval System online, Database in Nursing, US National Library of Medicine, Cumulative Index to Nursing & Allied Health Literature, Scopus Info Site and Web of Science in December 2015. Results: 28 articles were selected to compose the final sample of this review, which were analyzed and categorized in management technology (86.20%), assistance technology (20.68%) and educational technology (10.34%). Studies published in the last five years have predominated (62.06%), in international territory (82.75%) and with evidence level IV (44.82%). Conclusion: Today the growth on the production about this theme has been highlighted, but there is an important discrepancy between the researches with high and low level of scientific evidence, showing the need of more studies about technologies that strengthen the experience of nursing.

Antithrombotic treatment in chronic heart failure and sinus rhythm: Systematic review

AIM: To assess the efficacy and safety of antithrombotic drugs(antiplatelet or anticoagulant drugs) compared to no antithrombotic treatment or placebo in patients with heart failure(HF) and sinus rhythm. METHoDS: We searched Medline and Cochrane Library for randomized controlled trials evaluating antithrombotic treatment and no antithrombotic treatment in patients with HF and sinus rhythm. Risk ratio(RR) and 95%CIs were estimated performing meta-analysis with random effects method. RESULTS: Two studies met the inclusion criteria: Heart failure Long-term Antithrombotic Study and Warfarin/Aspirin Study in Heart failure, with 336 patients and mean follow-up 1.8-2.25 years. Stroke risk was not reduced by acetylsalicylic acid(RR = 1.18, 95%CI: 0.17-8.15), oral anticoagulation(RR = 0.30, 95%CI: 0.03-2.65) or overall antithrombotic drugs(RR = 0.52, 95%CI: 0.10-2.74). Acetylsalicylic acid showed a significant increased risk of worsening HF(RR = 1.78, 95%CI: 1.08-2.92), while oral anticoagulation had no impact in this outcome(RR = 1.03, 95%CI: 0.61-1.75). overall antithrombotic drugs showed a significant risk increase of major bleeding(RR = 6.99, 95%CI: 0.89-54.64). CoNCLUSIoN: Best available evidence does not support the routine use of antithrombotic drugs in patients with HF and sinus rhythm. These drugs, particularly oral anti-coagulation has the hazard of increase significantly major bleeding risk.

objective: To evaluate the anti Candida activity of Hyptis martiusii decoction and its major compound, caffeic acid alone or in the presence of fluconazole, as well as their cytotoxic effect. Methods: The decoction was characterized using high performance liquid chromatography coupled with diode array detector. For the antifungal activity, the minimum inhibitory concentration(MIC) and the potential effect of the decoction with the fluconazole were evaluated by microdilution method using 96-well microtiter trays. The osmotic fragility test was performed using erythrocytes under saline stress. All tests were performed in triplicate. Results: The chemical characterization of the decoction was performed by high performance liquid chromatography and revealed the presence of seven compounds, including caffeic acid as major constituent. The antifungal tests demonstrated that both decoction(DHm) and caffeic acid obtained from Hyptis martiusii presented MIC and MFC ≥4096 μg/mL against Candida albicans and Candida tropicalis strains. However, in the presence of fluconazole, DHm and caffeic acid presented IC_(50) of 2.60 and 2.53 μg/mL respectively, demonstrating significant synergistic effects against Candida strains. The modulator activity of DHm might be due to the presence of caffeic acid. Moreover, DHm and caffeic acid did not cause significant hemolytic effects, indicating that they present low cytotoxicity. Conclusions: These data indicate that DHm potentiates the activity of the fluconazole, without enhancement of the toxicity, encouraging further toxicological, pharmacological and phytochemical studies to provide consistent evidence of the potential of this plant to be used in drug development.

The growth and production modeling of individual trees of Eucalyptus urophylla plantations

Individual tree models(ITMs)are classified as growth and production models for projecting current and future forest *** are more complex than other growth and production models,show a higher level of detail and,consequently,produce a better modeling ***,the accuracy and efficiency of ITMs have not been properly assessed to *** this study,we estimated the growth in height,diameter,and individual tree volume of a Eucalyptus urophylla plantation by applying an *** used a continuing forest inventory dataset in which 1554 individual trees within 29 permanent plots were measured in the field over a 6-year period(24 to 72 months).Each individual tree volume was estimated for future tree *** achieve this,we adjusted the model to predict the height and diameter growth,and the probability of mortality as a function of the competition *** ITM accuracy was assessed based on the analysis of variance results and,subsequently,the multiple mean comparison test at the 5%significance *** tree volumes predicted by the ITM for the forest stand aged 72 months,beginning at ages 24,36,48,and 60 months,were compared to the field measured tree volume acquired from the 72-month forest inventory that was used as the reference *** and observed tree volumes were similar when the estimation was based on the 48-month forest *** results might help to reduce financial costs of forest inventory because the ITM produces accurate future predictions of forest stand *** estimated ITM for Eucalyptus plantations using measurement intervals up to 2 years is recommended because it significantly reduced the projected volume discrepancy compared to the field measurements.

Contraception Knowledge, Attitudes and Practices of Adolescents at Risk of Pregnancy in Northeastern Brazil

Background: Contraceptive methods (CM) are important resources in the prevention of adolescent pregnancy. This study aimed to investigate adolescent knowledge, attitudes and practices related to contraception. Study Design: A cross-sectional and exploratory study was conducted with 7th to 9th grade adolescents in northeastern Brazil. Their knowledge, attitudes, and contraceptive method use were analysed by gender, age and sexual initiation groups. Results: A sexual debut had been experienced by 21% (n = 120) of all respondents (n = 570). The majority of the respondents who had experienced a sexual debut were male (73.3%, n = 88), their mean age was 15 years (SD = 1.46), and 49.3% (n = 59) never used any CM. The participants displayed low knowledge and unfavourable and ambivalent attitudes concerning contraception. Conclusion: Adolescents initiate sexual intercourse with knowledge and attitudes that limit the use of CM, which makes them more at risk of pregnancy. Further research that will be representative of the specific population in need of interventions is necessary.

The aim of the current study was to determine the nature of the seasonal variability of the Suspended Particulate Matter(SPM) fluxes from the drainage basin to the estuary in a macrotidal region(Northeastern Brazil),and the estuarine response to a seawater intrusion regarding sediment deposition,which will support the understanding of the global transport of materials at the continent-ocean *** structure data was acquired using a Conductivity,Temperature,and Depth(CTD) probe with a sampling frequency of 4 *** particulate material was measured by gravimetric measurements applied to exact filtered volume *** outflows were measured through the use of an Acoustic Doppler Current Profiler(ADCP) with frequency of 1.5 *** horizontal thermal and saline gradients varied from warmer and less saline waters(2014) to cooler and saline waters(2015).The gradient behavior when linked to volume transport and SPM flows,suggests a minimization of the fluvial flows in 2015,easing the advance of coastal water(CW) towards the inner estuary,leading to an inversion of the baroclinic pressure *** bottom saline front,generated by the entrance of coastal water masses,caused an increase in SPM concentrations due to increased fluid density,resuspension of previously deposited sediment,and erosion of *** concentrations of SPM indicate higher volume transport suggesting a hydraulic barrier due to the change/inversion of the baroclinic pressure gradient,resulting in water and material *** deposition was observed during neap tide,while during spring tide the material is resuspended,increasing the concentration,generating cycles of deposition and erosion during the neap-spring *** sediment in suspension that reach the estuary,even with low fluvial volume,stay in this environment forming new islands because of *** deposition rates or sediment cycling,if generated by the hydraulic barrier,may indicate that the flo

Background:The impact of leprosy reduces health-related quality of life of affected patients,interfering with different factors such as *** study investigated the lipid profile,nutritional status,and risk for cardiovascular disease(CVD)in patients who underwent leprosy treatment in ***:Eighty-four adult patients of both genders ranging in age from 20 to 60 years and diagnosed with paucibacillary(PB)or multibacillary(MB)leprosy were selected after undergoing multidrug *** following data were collected:sociodemographic and clinical data;food intake;anthropometric measures(weight,height,and waist circumference);and lipid profile components(total cholesterol,high-density lipoprotein cholesterol[HDL-c],low-density lipoprotein cholesterol[LDL-c],and triglycerides).Results:Among the study population,there was a predominance of males(65.48%)aged 50 to 60 years,patients with an income of 248-496 American dollars(63.10%),patients who completed elementary school(65.48%),inactive patients(76.19%),non-smokers(46.43%),and non-drinking patients(69.05%).The levels(mean±standard deviation)of total cholesterol were 193.8±29.4 mg/dL in the PB form and 203.5±41.7 mg/dL in the MB *** mean LDL-c was 116.9±22.7 mg/dL in PB patients and 121±31.3 mg/dL in MB *** triglyceride levels were 123.4±45.2 mg/dL in the PB form and 147.4±88.9 mg/dL in the MB *** evaluation of nutritional status showed that 41.67%of the patients were eutrophic,while 55.96%had excess *** intake was significantly associated with HDL-c in male patients(P=0.0264)and with triglycerides in patients above the ideal weight(P=0.0049).Conclusions:The risk of acquiring CVDs was observed to be high due to patients’excess weight and increased waist *** study will guide clinicians in the adequate treatment of patients with leprosy in order to avoid adverse cardiovascular events.
