Phenotypic Characterization of Porcine IFNc-Producing Lymphocytes in Porcine Reproductive and Respiratory Syndrome Virus Vaccinated and Challenged Pigs

Porcine reproductive and respiratory syndrome(PRRS) continues to be one of the most important swine diseases worldwide. Interferon-γ(IFNγ)-mediated type Ⅰ cell-mediated immune response plays an important role in protection from,and clearance of, PRRS virus(PRRSV). Several lymphocyte subsets including T-helper, CTLs, Th/memory cells, and cd T lymphocytes were previously reported to produce IFNc during PRRSV infection. However, the proportion and phenotypic characterization of these IFNγ-secreting lymphocytes have not been explored. In this study, IFNc producted by different lymphocyte subsets was assessed by multi-color flow cytometry after vaccination with PRRSV modified live vaccine(PRRSV-MLV) and challenge with homogeneous or heterogeneous PRRSV. The results showed that T-helper cells were the major IFNγ-secreting cell population after PRRSV-MLV vaccination and PRRSV challenge. Additionally, the proportion of IFNγ producing Th/memory cells and γδ T cells increased after PRRSV challenge. This difference was accounted for an enhanced ability to produce IFNγ in Th/memory cells and an enlarged quantity of γδ T cells. The results presented here could contribute to our understanding of the roles of IFNγ in protective immunity against PRRSV infection and may be useful for assessment of cell-mediated immunity in vaccine tests.

Caspase-1 activation and mature interleukin-1β release are uncoupled events in monocytes

Montanide<sup>TM</sup>Gel01 ST Adjuvant Enhances PRRS Modified Live Vaccine Efficacy by Regulating Porcine Humoral and Cellular Immune Responses

Porcine reproductive and respiratory syndrome (PRRS) is a devastating disease caused by the PRRS virus. The MontanideTM class of flexible polymeric adjuvants has recently been shown to enhance protective immunity against PRRSV infection in piglets when used in combination with PRRS modified live vaccines (MLV). In this study, we explored the efficacy and immunological mechanisms of protection of MontanideTM Gel01 ST (Gel01) adjuvanted modified live PRRSV vaccine in pigs challenged with two genetically distinct strains of PRRSV. Gel01-MLV reduced lymph node pathology scores in pigs challenged with VR-2332 (parental strain of MLV vaccine) but not that in pigs challenged with MN184A (heterologous strain), when compared to that in pigs vaccinated with un-adjuvanted MLV. Pigs vaccinated with Gel01-MLV had higher levels of PRRS-specific antibodies, as measured by IDEXX ELISA and virus neutralizing antibodies, after vaccination and VR-2332 challenge. In addition, pigs vaccinated with Gel01-MLV had decreased levels of IFN-γ, IL-10, and T-regulatory lymphocytes in the blood as compared to that in pigs vaccinated with MLV alone. Interestingly, we found that addition of Gel01 did not change the profile of other T lymphocyte populations after PRRSV challenge. These results demonstrate that the MLV adjuvanted with Gel01 provides enhanced protection against homologous PRRSV infection, possibly by regulating the production of PRRSV-specific antibodies and cytokines involved in the development of T-regulatory cells. Thus, Gel01 ST is a promising adjuvant that can be formulated with PRRSV MLV vaccines to reduce disease severity and tissue damage caused by PRRSV infection in pigs.

As the causative agent of classical swine fever,the economically devastating swine disease worldwide,classical swine fever virus（CSFV） is currently classified into the 11 subgenotypes,of which subgenotype 2.1 is distributed worldwide and showing more genetic diversity than other *** to this report,subgenotype 2.1 was divided into three sub-subgenotypes（2.1a-2.1c）.To further analyze the genetic diversity of CSFV isolates in China,39 CSFV isolates collected between 2004-2012 in two Chinese provinces Guangxi and Guangdong were sequenced and subjected to phylogenetic analysis together with reference sequences retrieved from Gen *** analyses based on the 190-nt and/or 1119-nt full length E2 gene fragments showed that current CSFV subgenotype 2.1 virus isolates in the world could be divided into 10 sub-subgenotypes（2.1a-2.1j） and the 39 isolates collected in this study were grouped into 7 of them（2.1a-2.1c and 2.1g-2.1j）.Among the 10 sub-subgenotypes,2.1d-2.1j were newly ***-subgenotypes 2.1d isolates were circulated only in India,however the rest 9 sub-subgenotypes were from China with some of them closely related to isolates from European and neighboring Asian *** to the temporal and spatial distribution of CSFV subgenotype 2.1 isolates,the newly classified 10 sub-subgenotypes were further categorized into three groups:dominant sub-subgenotype,minor sub-subgenotype and silent sub-subgenotype,and each sub-subgenotype can be found only in certain geographical *** together,this study reveals the complex genetic diversity of CSFV subgenotype 2.1 and improves our understanding about the epidemiological trends of CSFV subgenotype 2.1 in the world,particularly in China.

Background Pancreaticoduodenectomy(PD)has been widely applied in general hospitals in China;however,there is still a lack of unified standards for each surgical technique and *** survey is intended to investigate the current status of digestive tract reconstruction after PD in university hospitals in *** A cross-sectional survey was conducted among the members of the Young Elite Pancreatic Surgery Club of China by using the Questionnaire for Digestive Tract Reconstruction after *** questionnaire was disseminated and collected by point-to-point communication via WeChat public *** A total of 73 valid questionnaires were returned from 65 university hospitals in 28 provincial divisions of China's *** respondents who performed PD surgery with an annual volume of over 100 cases accounted for 63%.Generally,laparoscopic PD was performed less often than open *** and Whipple reconstructions accounted for 70%and 26%,*** sequence of pancreatoenteric,biliary-enteric,and gastrointestinal reconstruction accounted for 84%of *** pancreatoenteric anastomosis,double-layer anastomosis is the most commonly employed type,accounting for approximately 67%,while single-layer anastomosis accounts for 30%.Of the double-layer anastomoses,duct-to-mucosa/dunking(94%/4%)PJ was performed with duct-mucosa using the Blumgart method(39%)and Cattel-Warren(29%),with continuous/interrupted sutures in the inner layer(69%/31%)and continuous/interrupted sutures in the outer layer(53%/23%).In single-layer anastomosis,continuous/interrupted sutures accounted for 41%/45%.In hepatojejunostomy,single-layer/double-layer suture accounted for 79%/4%,and continuous/interrupted suture accounted for 75%/9%.Forty-six percent of the responding units had not applied double-layer biliary-intestinal anastomosis in the last 3 years,75%of the responding surgeons chose the anastomosis method according to bile duct diameter,with absorbab
