Detection of HBV DNA integration in plasma cell-free DNA of different HBV diseases utilizing DNA capture strategy

The landscape of hepatitis B virus(HBV)integration in the plasma cell-free DNA(cfDNA)of HBV-infected patients with different stages of liver diseases[chronic hepatitis B(CHB),liver cirrhosis(LC),and hepatocellular carcinoma(HCC)]remains *** this study,we developed an improved strategy for detecting HBV DNA integration in plasma cfDNA,based on DNA probe capture and next-generation *** this optimized strategy,we successfully detected HBV integration events in chimeric artificial DNA samples and HBV-infected HepG2-NTCP cells at day one post infection,with high sensitivity and *** characteristics of HBV integration events in the HBV-infected HepG2-NTCP cells and plasma cfDNA from HBV-infected individuals(CHB,LC,and HCC)were further investigated.A total of 112 and 333 integration breakpoints were detected in the HepG2-NTCP cells and 22 out of 25(88%)clinical HBV-infected samples,*** vivo analysis showed that the normalized number of support unique sequences(nnsus)in HCC was significantly higher than in CHB or LC patients(P values<0.05).All integration breakpoints are randomly distributed on human chromosomes and are enriched in the HBV genome around nt *** majority of integration breakpoints(61.86%)are located in the gene-coding *** non-homologous end-joining(NHEJ)and microhomology-mediated end-joining(MMEJ)interactions occurred during HBV integration across the three different stages of liver *** study provides evidence that HBV DNA integration can be detected in the plasma cfDNA of HBV-infected patients,including those with CHB,LC,or HCC,using this optimized strategy.

Vulnerability and Conservation of Nearshore Cetaceans to Increasing Vessel Traffic:The Indo-Pacific Humpback Dolphin in the South China Sea

A comprehensive risk assessment to evaluate the vulnerability of the nearshore cetaceans to increasing marine traffic is *** this study,the risk to Indo-Pacific humpback dolphins(Sousa chinensis)in the main habitats in China was assessed based on a semiquantitative spatial vulnerability analysis.85%of the(sub)population which overlapped with the lanes of ferries and/or cargo vessels,with exposure scores of 1.23±0.31(calculated based on the proportion of sea routes that overlap the dolphin’s range).A combination of high exposure to the sea routes and sensitivity to marine traffic would render the(sub)populations to be highly vulnerable(EPRE:5.21 and XM:4.47).A low frequency of vessels and an awareness of the ecology assures a low vulnerability(SH:2.43,EZ:2.73).Uncertainty scores were commonly lower for the higher vulnerable populations,and higher for the lower vulnerable populations,suggesting additional field data and more monitoring are *** years of field data have been used to analyze the influences of various types of marine traffic on humpback dolphin *** and non-fishing vessels were significantly different(χ^(2)=65.19,p<0.01).The humpback dolphins were attracted by fishing vessels(83.1%)but avoided non-fishing vessels(95.2%).We recommend a management with seasonal and geographical constraints,to balance economic development and environmental conservation.

Prediction of soil organic carbon stock combining Sentinel-1 and Sentinel-2 images in the Zoige Plateau, the northeastern Qinghai-Tibet Plateau

Background Soil organic carbon（SOC） is a critical component of the global carbon cycle, and an accurate estimate of regional SOC stock（SOCS） would significantly improve our understanding of SOC sequestration and cycles. Zoige Plateau, locating in the northeastern Qinghai-Tibet Plateau, has the largest alpine marsh wetland worldwide and exhibits a high sensitivity to climate fluctuations. Despite an increasing use of optical remote sensing in predicting regional SOCS, optical remote sensing has obvious limitations in the Zoige Plateau due to highly cloudy weather, and knowledge of on the spatial patterns of SOCS is limited. Therefore, in the current study, the spatial distributions of SOCS within 100 cm were predicted using an XGBoost model—a machine learning approach, by integrating Sentinel-1, Sentinel-2 and field observations in the Zoige *** The results showed that SOC content exhibited vertical distribution patterns within 100 cm, with the highest SOC content in topsoil. The tenfold cross-validation approach showed that XGBoost model satisfactorily predicted the spatial patterns of SOCS with a model efficiency of 0.59 and a root mean standard error of 95.2 Mg ha-1. Predicted SOCS showed a distinct spatial heterogeneity in the Zoige Plateau, with an average of 355.7 ± 123.1 Mg ha-1within 100 cm and totaled 0.27 × 10～9 Mg *** High SOC content in topsoil highlights the high risks of significant carbon loss from topsoil due to human activities in the Zoige Plateau. Combining Sentinel-1 and Sentinel-2 satisfactorily predicted SOCS using the XGBoost model, which demonstrates the importance of selecting modeling approaches and satellite images to improve efficiency in predicting SOCS distribution at a fine spatial resolution of 10 m. Cathodic corrosion as a facile and universal method for scalable preparation of powdery single atom electrocatalysts

Top-down strategy has been widely applied for synthesis of metal single atom catalysts(SACs)via converting metal nanoparticles or bulk metals into atomically dispersed ***,we report a simple electrochemical atomic migration strategy for top-down synthesis of SACs via a facile cathodic corrosion process without involving high temperature or harsh *** of metal nanoparticles on cathode are firstly disbanded under high negative voltage,and emitted into the electrolyte in the form of atomic metal anions in Zintl *** escaped atomically dispersed metal species are then oxidized by water molecules and captured by the defects on the pre-added nitrogen doped carbon carriers in the *** cathodic corrosion strategy is confirmed to be suitable for scalable synthesis of kinds of metal SACs(e.g.,Pt,Pd,and Ir)on doped carbon ***,the as-prepared nitrogen doped carbon powder supported Pt SACs exhibit superior catalytic activity toward hydrogen evolution reaction(HER)with a low overpotential of 0.024 V at 10 mA·cm^(−2)and a low Tafel slope of 29.7 mV·dec^(−1)as well as a long-term durability.

BACKGROUND Insulin autoimmune syndrome(IAS)is a severe manifestation of spontaneous *** is characterized by elevated levels of immune-reactive insulin and highly potent insulin autoantibodies(IAAs),which are induced by endogenous insulin circulating in the *** is distinguished by recurring instances of spontaneous hypoglycemia,the presence of IAA within the body,a substantial elevation in serum insulin levels,and an absence of prior exogenous insulin ***,recent studies show that both conventional insulin and its analogs can induce IAS episodes,giving rise to the notion of nonclassical ***,more attention should be paid to these *** SUMMARY In this case report,we present a rare case of non-classical IAS in an 83-year-old male patient who present with symptoms of a psychiatric *** symptom onset,the patient exhibited Whipple's triad(including hypoglycemia,blood glucose level less than 2.8 mmol/L during onset,and rapid relief of hypoglycemic symptoms after glucose administration).Concurrently,his serum insulin level was significantly elevated,which contradicted his C-peptide *** a comprehensive examination,the patient was diagnosed with exogenous insulin autoimmune *** that the patient had type 2 diabetes mellitus and a history of exogenous insulin use before disease onset,it was presumed that non classical IAS was induced by this *** PubMed database was used to search for previous cases of IAS and non-classical IAS to analyze their characteristics and treatment *** The occurrence of non-classical IAS is associated with exogenous insulin or its analogs,as well as with sulfhydryl *** can be effectively alleviated through the discontinuation of relevant medications,administration of hormones or immunosuppressants,plasma exchange,and lifestyle adjustments.

Mechanistic insights into the synergetic remediation and amendment effects of zeolite/biochar composite on heavy metal-polluted red soil

Harnessing antimicrobial peptide-coupled photosensitizer to combat drug-resistant biofilm infections through enhanced photodynamic therapy

Bacterial biofilm-associated infection was one of the most serious threats to human health. However, effective drugs for drug-resistance bacteria or biofilms remain rarely reported. Here, we propose an innovative strategy to develop a multifunctional antimicrobial agent with broad-spectrum antibacterial activity by coupling photosensitizers (PSs) with antimicrobial peptides (AMPs). This strategy capitalizes on the ability of PSs to generate reactive oxygen species (ROS) and the membrane-targeting property of AMPs (KRWWKWIRW, a peptide screened by an artificial neural network), synergistically enhancing the antimicrobial activity. In addition, unlike conventional aggregation-caused quenching (ACQ) photosensitizers, aggregation-induced emission (AIE) PSs show stronger fluorescence emission in the aggregated state to help visualize the antibacterial mechanism. In vitro antibacterial experiments demonstrated the excellent killing effects of the developed agent against both Gram-positive (G^(+)) and Gram-negative (G^(–)) bacteria. The bacterial-aggregations induced ability enhanced the photoactivatable antibacterial activity against G^(–) bacteria. Notably, it exhibited a significant effect on destroying MRSA biofilms. Moreover, it also showed remarkable efficacy in treating wound infections in mice in vivo. This multifunctional antimicrobial agent holds significant potential in addressing the challenges posed by bacterial biofilm-associated infections and drug-resistant bacteria.
