Introduction:All countries face the potential threat of imported polioviruses,including both wild type and circulating vaccine-derived *** response,we conducted a province-level assessment in China to evaluate the risk of importation and transmission of type 1 wild poliovirus(WPV1)and type 2 vaccine-derived poliovirus(VDPV2).Methods:Distinct risk assessment tools for WPV1 and VDPV2 were employed,incorporating three primary indicators—population immunity,poliovirus surveillance,and importation ***1 was assessed using 13 secondary indicators,whereas VDPV2 utilized 21 secondary *** used comprehensive provincial data from the preceding five ***(S-values)were derived from the secondary indicators’criteria and ratings,and used to classify the provincial-level administrative divisions(PLADs)into three risk categories:high,medium,and *** top 10%of PLADs were designated as highrisk,with the remaining provinces equally distributed into medium-and low-risk ***:In 2023,Xizang,Qinghai,and Xinjiang PLADs faced the highest risk of WPV1 importation and transmission;Xizang,Shaanxi,and Hainan PLADs were at the greatest risk for VDPV2 importation and ***:Risk assessment for VDPV2 importation and transmission has identified a distinct set of high-risk provinces compared to those identified by WPV1 risk *** and proactive response measures tailored to the specific risks should be implemented to maintain China’s polio-free status.

Stem cells remain in a quiescent state for long-term maintenance and preservation of potency;this process requires fine-tuning regulatory *** this study,we identified the epigenetic landscape along the developmental trajectory of skeletal stem cells(SSCs)in skeletogenesis governed by a key regulator,Ptip(also known as Paxip1,Pax interaction with transcription-activation domain protein-1).Our results showed that Ptip is required for maintaining the quiescence and potency of SSCs,and loss of Ptip in type II collagen(Col2)^(+)progenitors causes abnormal activation and differentiation of SSCs,impaired growth plate morphogenesis,and long bone *** also found that Ptip suppressed the glycolysis of SSCs through downregulation of phosphoglycerate kinase 1(Pgk1)by repressing histone H3 lysine 27 acetylation(H3K27ac)at the promoter ***,inhibition of glycolysis improved the function of SSCs despite Ptip *** the best of our knowledge,this is the first study to establish an epigenetic framework based on Ptip,which safeguards skeletal stem cell quiescence and potency through metabolic *** framework is expected to improve SSC-based treatments of bone developmental disorders.

High-rate CH_(4)-to-C_(2)H_(6) photoconversion enabled by Au/ZnO porous nanosheets under oxygen-free system

Photocatalytic CH_(4) coupling into high-valued C_(2)H_(6) is highly attractive,whereas the photosynthetic rate,especially under oxygen-free system,is still ***,we designed the negatively charged metal supported on metal oxide nanosheets to activate the inert C-H bond in CH_(4)and hence accelerate CH_(4) coupling *** an example,the synthetic Au/ZnO porous nanosheets exhibit the C_(2)H_(6) photosynthetic rate of 1,121.6μmol g^(-1)_(cat)h^(-1)and the CH_(4) conversion rate of 2,374.6μmol g^(-1)_(cat)h^(-1) under oxygen-free system,2 orders of magnitude higher than those of previously reported *** virtue of several in situ spectroscopic techniques,it is established that the generated Au^(δ-)and O^-species together polarized the C-H bond,while the Au^(δ-)and O^-species jointly stabilized the CH_(3) intermediates,which favored the coupling of CH_(3) intermediate to photosynthesize C_(2)H_(6) instead of overoxidation into CO_(x).Thus,the design of dual active species is beneficial for achieving high-efficient CH_(4)-to-C_(2)H_(6) photoconversion.

Guaico Culex virus(GCXV)is a newly identified segmented jingmenvirus from Culex *** in Central and South *** genome of GCXV is composed of four or five single-stranded positive RNA ***,the infection kinetics and transmission capability of GCXV in mosquitoes remain *** this study,we used reverse genetics to rescue two GCXVs(4S and 5S)that contained four and five RNA segments,respectively,in C6/36 *** in vitro characterization revealed that the two GCXVs exhibited comparable replication kinetics,protein expression and viral ***,GCXV RNAs were detected in the bodies,salivary glands,midguts and ovaries of Culex quinquefasciatus at 4–10 days after oral *** addition,two GCXVs can colonize *** eggs,resulting in positive rates of 15%–35%for the second gonotrophic *** conclusion,our results demonstrated that GCXVs with four or five RNA segments can be detected in *** eggs during the first and second gonotrophic cycles after oral infection.

Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT03950154)assessed the efficacy and adverse events(AEs)of the combination of PD-1 blockade-activated DC-CIK(PD1-T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 202 participants were enrolled and randomly assigned in a 1:1 ratio to receive either first-line XELOX plus bevacizumab(the control group,n=102)or the same regimen plus autologous PD1-T cell immunotherapy(the immunotherapy group,n=100)every 21 days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 19.5 *** PFS was 14.8 months(95%CI,11.6-18.0)for the immunotherapy group compared with 9.9 months(8.0-11.8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 25.6 months(95%CI,18.3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 20.0%of patients in the immunotherapy group and 23.5%in the control groups,with no toxicity-associated deaths *** addition of PD1-T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

BACKGROUND Whether operation is superior to non-operation for humeral shaft fracture remains *** hypothesized that operation could decrease the nonunion and reintervention rates and increase the functional *** To compare the clinical efficacy between operative and nonoperative approaches for humeral shaft *** We searched the PubMed,Web of Science,ScienceDirect,and Cochrane databases from 1990 to December 2023 for clinical trials and cohort studies comparing the effects of operative and conservative methods on humeral shaft *** investigators independently extracted data from the eligible studies,and the other two assessed the methodological quality of each *** quality of the included studies was assessed using the Cochrane risk bias or Newcastle-Ottawa *** nonunion,reintervention and the overall complications and functional scores were pooled and analyzed using Review Manager software(version 5.3).RESULTS A total of four randomized control trials and 13 cohort studies were included,with 1285 and 1346 patients in the operative and nonoperative groups,*** in the operative group were treated with a plate or nail,whereas those in the conservative group were managed with splint or functional *** studies were assessed as having a high risk of bias,and the other 13 were of a low risk of bias according to the Newcastle-Ottawa Scale or Cochrane risk bias *** operative group had a significantly decreased rate of nonunion[odds ratio(OR)0.30;95%CI:0.23 to 0.40,reintervention(OR:0.33;95%CI:0.24 to 0.47),and overall complications(OR:0.62;95%CI:0.49 to 0.78)].The pooled effect of the Disabilities of Arm,Shoulder,and Hand score showed a significant difference at 3[mean difference(MD)-8.26;95%CI:-13.60 to-2.92],6(MD:-6.72;95%CI:-11.34 to-2.10),and 12 months(MD:-2.55;95%CI:-4.36 to-0.74).The pooled effect of Visual Analog Scale scores and the Constant-Murley score did not significantly differ between
