BACKGROUND Acquired hemophilia A (AHA) is a rare and potentially severe bleeding disordercaused by circulating autoantibodies against factor Ⅷ (FⅧ). In approximately50% of the patients, the condition is associated with autoimmune diseases,cancers, medication use, pregnancy, and the post-partum period. Bullous pemphigoid(BP) is a chronic autoimmune subepidermal blistering disease associatedwith tissue-bound and circulating autoantibodies against BP antigens 180 (BP180)and 230 (BP230). AHA-associated BP has a high mortality rate;hence, the understandingof this disease must *** SUMMARY A 69-year-old man presented with erythema, blisters, blood blisters, and crustsaccompanied by severe pruritus for more than 20 days, and ecchymosis andswelling on his left upper arm for 3 days. Pathological examination revealed asubepidermal blister that contained eosinophils. Laboratory tests showed that theBP180 autoantibody levels had increased, isolated activated partial thromboplastintime was notably prolonged (115.6 s), and coagulation FⅧ activity wasextremely low (lity rate. Therefore, its timely diagnosis and effectivetreatment are necessary.

Halogenated organic pollutants (HOPs) have attracted considerable attention owing to their persistence, bioaccumulation, and toxicity. The development of methods to detect HOPs in fish is challenging owing to the compositional complexity of fish matrices, which contain high levels of lipids and relatively low concentrations of HOPs. In addition, the lipophilicity of most HOPs renders their extraction difficult. Moreover, the simultaneous determination of multiple HOPs to achieve the high-throughput screening of these analytes is complex. In this study, a reliable and efficient pretreatment method based on ultrasound-assisted extraction, gel permeation chromatography purification, and ultra performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS) was developed for the determination of 12 HOPs in edible fish. The procedures of sample extraction and purification and LC-HRMS detection parameters were optimized to improve the performance of the method. Fresh fish samples were thoroughly rinsed with water, and non-edible parts, including the skin, bones, and phosphorus, were removed. The fish were weighed, cut into small pieces, and vacuum freeze-dried for 48 h. Subsequently, a freeze grinder was used to grind the dried fish into a fine powder. Exactly 2 g of the fish powder was weighed, fortified with isotope-labeled internal standards of the HOPs, and allowed to stand for 5 min. Methanol-acetonitrile (1∶1, v/v) was then added, followed by vortex mixing and ultrasonication. After centrifugation, the supernatant was transferred to a fresh tube. The extraction process was repeated twice and all extracts were combined. The extract was evaporated under a gentle nitrogen flow and redissolved in a mixture of ethyl acetate-cyclohexane (1∶1, v/v). The sample mixture was cleaned using gel permeation chromatography, and the eluate was collected and concentrated under a nitrogen flow. Skeletal muscle plays a significant role in both local and systemic energy metabolism. The current investigation aims to explore the role of the Bambi gene in skeletal muscle, focusing on its implications for muscle hypertrophy and systemic metabolism. We hypothesize that skeletal muscle-specific deletion of Bambi induces muscle hypertrophy, improves metabolic performance, and activates thermogenic adipocytes via the reprogramming of progenitor of i WAT, offering potential therapeutic strategies for metabolic syndromes. Leveraging the Chromatin immunoprecipitation(ChIP)-seq and bioinformatics analysis, Bambi gene is shown to be a direct target of HIF2α, which is further confirmed by ChIP-qPCR and promoter luciferase assay. Skeletal muscle-specific Bambi deletion led to significant muscle hypertrophy and improved metabolic parameters, even under high-fat diet conditions. This deletion induced metabolic reprogramming of stromal vascular fractions(SVFs) into thermogenic adipocytes, contributing to systemic metabolic improvements, potentially through the secretory ***, mice with skeletal muscle-specific Bambi deletion demonstrate resistance to high-fat diet-induced metabolic disorders, highlighting a potential therapeutic pathway for metabolic syndrome management. Thus, skeletal muscle-specific deletion of Bambi triggers muscle growth, enhances metabolic performance, and activates thermogenic adipocytes. These findings suggest Bambi as a novel therapeutic target for metabolic syndromes, providing new insights into the interaction between muscle hypertrophy and systemic metabolic improvement. The study underscores the potential of manipulating muscle physiology to regulate whole-body metabolism, offering a novel perspective on treating metabolic disorders.

On July 27, 2022, a high-precision system for measuring the magnetic field based on a coherent population trapping magnetometer（referred to as the CPT system） was launched onboard the Space Advanced Technology demonstration satellite（SATech-01） by a ZK-1A rocket. The payload comprises a scalar coherent population trapping magnetometer（CPTM）, triaxial anisotropic magnetoresistance magnetometer（AMRM）, and nano star tracker mounted on a non-magnetic telescopic tubular mast. This configuration enables synchronized measurement of scalar and vector geomagnetic fields, as well as the attitude,making the payload with its compact sensors suitable for applications on miniature satellite platforms. On November 7, 2022,the novel telescopic tubular mast was deployed in orbit, extending to a length of 5.28 m. The CPTM, an absolute scalar magnetometer with an omnidirectional sensor, is the first China-developed quantum/atomic magnetometer successfully operating in space. In orbit, the CPTM has sensitivity of ～10 pT/Hz1/2and can automatically operate under complex magnetic field variations. The difference between the scalar fields obtained using the CPTM and CHAOS model has a mean deviation of–3.73 n T and a standard deviation of 26.11 nT（1σ）, without on-orbit calibration or correction. The miniaturized AMRM has a sensitivity of ～0.3 n T/Hz1/2and maintains stable and reliable operation in orbit. The low-power and miniaturized nano star tracker has a measurement uncertainty of 14.23″（1σ） in orbit. The mission has realized the on-orbit technical verification of the integrated and synchronized detection of the vector magnetic field and attitude and fully validated the system's performance,automation level, and reliability.

If your computer crashes, you can revive it by a reboot, an empirical solution that usually turns out to be effective. The rationale behind this solution is that transient faults, either in hardware or software,can be fixed by refreshing the machine state. Such a "silver bullet", however, could be futile in the future because the faults, especially those existing in the hardware such as Integrated Circuit(IC) chips, cannot be eliminated by refreshing. What we need is a more sophisticated mechanism to steer the system back to the right track. The "magic cure" is the Fault Tolerance On-Chip(FTOC) mechanism, which relies on a suite of built-in design-for-reliability logic, including fault detection, fault diagnosis, and error recovery, working in a self-supportive manner. We have exploited the FTOC to build a holistic solution ranging from on-chip fault detection to error recovery mechanisms to address faults caused by chips progressively aging. Besides fault detection, the FTOC paradigm provides attractive benefits, such as facilitating graceful performance degradation, mitigating the impact of verification blind spots, and improving the chip yield.
