Background:A differential gene,triggering receptor expressed on myeloid cells 1(TREM1),was identiﬁed in blood sequencing datasets from myocardial infarction patients and healthy ***ﬁbrosis following myocardial infarction signiﬁcantly contributes to cardiac ***:This study aimed to unveil the intrinsic regulatory mechanism of TREM1 in myocardialﬁ***:Mimicking pathology by angiotensin II(Ang II)treatment of human cardiacﬁbroblasts(HCFs),the impacts of TREM1 knockdown on its proliferation,migration,and secretion of the pro-ﬁbrotic matrix were identiﬁ*** the Human Transcription Factor Database(HumanTFDB)website,lysine-speciﬁc demethylase 5B(KDM5B)was found to bind to the TREM1 promoter,which was further validated through luciferase reporter and chromatin immunoprecipitation(ChIP).By promoting KDM5B overexpression,its effect on the regulation of TREM1 was ***:TREM1 knockdown suppressed the proliferation,migration,and secretion of the pro-ﬁbrotic matrix in HCFs upon Ang II ***5B bound to the TREM1 promoter and upregulated its transcriptional ***,KDM5B overexpression reversed the regulation of the above cellular phenotypes by TREM1 ***:This study sheds light on the positive regulation of TREM1 by KDM5B,demonstrating their role in promoting myocardialﬁ***ﬁnding provides a theoretical foundation for understanding disease pathology and potentially advancing the development of new targeted therapies.

Gears are pivotal in mechanical drives,and gear contact analysis is a typically difficult problem to *** isogeometric analysis(IGA)methods have developed new ideas to solve this *** this paper,a threedimensional body parametric gear model of IGA is established,and a theoretical formula is derived to realize single-tooth contact *** were benchmarked against those obtained from commercial software utilizing the finite element analysis(FEA)method to validate the accuracy of our *** findings indicate that the IGA-based contact algorithmsuccessfullymet theHertz contact *** juxtaposed with the FEA approach,the IGAmethod demonstrated fewer node degrees of freedomand reduced computational units,all whilemaintaining comparable ***,the IGA method appeared to exhibit consistency in analysis accuracy irrespective of computational unit density,and also significantlymitigated non-physical oscillations in contact stress across the tooth *** underscores the prowess of IGA in contact *** conclusion,IGA emerges as a potent tool for addressing contact analysis challenges and holds significant promise for 3D gear modeling,simulation,and optimization of various mechanical components.

The association between PM_(2.5)(particulate matter≤2.5μm)short-term exposure and its health effect is non-linear from the epidemiological *** this nonlinearity is suggested to be related with the PM_(2.5)heterogeneity,however,the underlying biological mechanism is still ***,a total of 38 PM_(2.5)filters were collected continuously for three weeks in winter Beijing,with the ambient PM_(2.5)varying between 10 and 270μg/m^(3).Human monocytes-derived macrophages(THP-1)were treated with PM_(2.5)water-soluble elutes at10μg/mL to investigate the PM_(2.5)short-term exposure effect from a proinflammatory *** proinflammatory cytokine tumor necrosis factor(TNF)induced by the PM_(2.5)elutes at equal concentrations were unequal,showing the heterogeneity of PM_(2.5)proinflammatory *** the various chemical and biological components,lipopolysaccharide(LPS)showed a strong positive association with the TNF ***,some outliers were observed among the TNF-LPS ***,for PM_(2.5)from relatively clean air episodes,the higher LPS amount corresponded to relatively low TNF *** this phenomenon was also observed in the promotion tests by treating macrophages with PM_(2.5)elutes dosed with additional trace *** expression analysis indicated the involvement of oxidative-stress related genes in the LPS signaling ***,a potential oxidative-stress-mediated suppression on the PM_(2.5)-borne LPS proinflammatory effect was proposed to be accounted for the ***,the results showed the differential role of LPS in the heterogeneity of PM_(2.5)proinflammatory effects from a component-based *** experimental studies are needed to elucidate the signaling pathway of LPS attached on PM_(2.5)from different air quality episodes.

Objective:To investigate the anti-liver cancer effects and aspartic acid(Asp)-related action mechanism of Euphorbia fischeriana Steud.(LD).Methods:The mice model of liver cancer was established by injection of H22 *** 5 days,mice were randomly divided into model group,sorafenib group(20 mg/kg),LD high-dose(LDH,1.36 g/kg) group,LD medium-dose(LDM,0.68 g/kg) group,and LD low-dose(LDL,0.34 g/kg)group,10 mice each *** were intragastrically administered to the mice once daily for 10 days,*** weight,tumor size and tumor weight were *** index was *** changes of liver cancer tissues were evaluated by hematoxylin and eosin staining and TUNEL *** chromatography-mass spectrometer was used to analyze different metabolites between the model and LDH ***:After LD treatment,tumor weight,tumor size and hepatic index were reduced compared with the model *** and karyorrhexis of tumor cells were ***,61 differential metabolites(18 up-regulated,43 down-regulated) were affirmed and 20 pathways of KEGG(P<0.05) were *** addition,Bel-7402,HepG2 and H22 cell viabilities were significantly increased after adding Asp into the *** then,the cell proliferation effect induced by Asp was ameliorated by ***:The anti-liver cancer efficacy of LD extract was validated in H22 mice model,and inhibition of Asp level might be the underlying mechanism.

To accurately obtain the waveform template of gravitational waves, substantial computational resources and extremely high precision are often required. In a previous study, we employed the confluent Heun function to obtain an exact solution to the Teukolsky equation. This approach allowed us to efficiently and accurately calculate the gravitational wave flux for a particle in circular orbits around a Schwarzschild black hole. Building on this method, we now extend its application to calculate the asymptotic gravitational wave fluxes from a particle in generic orbits around a near-extreme Kerr black hole. Our extended method proves particularly effective in handling computational challenges associated with large eccentricities(e = 0.9), higher spins(a = 0.999), higher harmonic modes, and strong-field regions. The results we obtained significantly outperform those derived from the numerical integration method based on the Mano-Suzuki-Takasugi method.
