Cystic fibrosis（CF） is a severe genetic disease caused by the gene mutation of the cystic fibrosis transmembrane conductance regulator（CFTR） chloride channel. The most common point mutation AF508, which leads to impaired intracellular processing and channel gating of CFTR, appears in about 90% CF patients. The natural compound curcumin was reported to correct the processing defect of AF508-CFTR and proposed as a potential therapeutic drug to cure CF. In the present study, we analyzed the effect of curcumin on AF508-CFTR and demonstrated that curcumin can restore the impaired chloride conductance of AF508 mutant CFTR. The activity is rapid, reversible and cAMP-dependent. However, we couldn＇t reproduce the previously reported correction of the defective membrane trafficking of AF508-CFTR by curcumin. Therefore, curcumin may not be a superior lead compound for developing anti-CF drugs.

BACKGROUND Massive intragastric clotting(MIC) makes endoscopic therapy difficult in patients with acute upper gastrointestinal bleeding. Literature data on how to address this problem are limited. Here, we report on a case of massive stomach bleeding with MIC that was successfully treated endoscopically using an overtube of singleballoon *** SUMMARY A 62-year-old gentleman with metastatic lung cancer was admitted to the intensive care unit due to tarry stools and hematemesis of 1500 mL of blood during hospitalization. Emergent esophagogastroduodenoscopy revealed massive blood clots and fresh blood in the stomach with evidence of active *** sites could not be observed even by changing the patient’s position and aggressive endoscope suction. The MIC was successfully removed using an overtube connected with a suction pipe, which was inserted into the stomach with an overtube of a single-balloon enteroscope. An ultrathin gastroscope was also introduced through the nose into the stomach to guide the suction. A massive blood clot was successfully removed, and an ulcer with oozing bleeding at the inferior lesser curvature of the upper gastric body was revealed, facilitating endoscopic hemostatic *** This technique appears to be a previously unreported method to suction MIC out of the stomach in patients with acute upper gastrointestinal bleeding. This technique could be considered when other methods are not available or if they fail to remove massive blood clots in the stomach.

The plasma transferred arc （PTA） forming remanufacturing technology was introduced in this paper. This technology includes plasma surfacing, deposition and rapid forming technology. With self-developed plasma forming system, the thrust of engine cylinder body was remanufactured by PTA powder surfacing. In the concrete, the Nil5 alloy was deposited on the thrust face of the body in order to recover its dimension. In addition, the reman- ufacturing forming with Fe-based, Inconel 625 alloy was studied. The microstructure and hardness of the as-depos- ited materials were investigated.

MFCIS: an automatic leaf-based identification pipeline for plant cultivars using deep learning and persistent homology

Recognizing plant cultivars reliably and efficiently can benefit plant breeders in terms of property rights protection and innovation of germplasm *** leaf image-based methods have been widely adopted in plant species identification,they seldom have been applied in cultivar identification due to the high similarity of leaves among ***,we propose an automatic leaf image-based cultivar identification pipeline called MFCIS(Multi-feature Combined Cultivar Identification System),which combines multiple leaf morphological features collected by persistent homology and a convolutional neural network(CNN).Persistent homology,a multiscale and robust method,was employed to extract the topological signatures of leaf shape,texture,and venation details.A CNN-based algorithm,the Xception network,was fine-tuned for extracting high-level leaf image *** fruit species,we benchmarked the MFCIS pipeline on a sweet cherry(Prunus avium L.)leaf dataset with>5000 leaf images from 88 varieties or unreleased selections and achieved a mean accuracy of 83.52%.For annual crop species,we applied the MFCIS pipeline to a soybean(Glycine max ***.)leaf dataset with 5000 leaf images of 100 cultivars or elite breeding lines collected at five growth *** identification models for each growth period were trained independently,and their results were combined using a score-level fusion *** classification accuracy after score-level fusion was 91.4%,which is much higher than the accuracy when utilizing each growth period independently or mixing all growth *** facilitate the adoption of the proposed pipelines,we constructed a user-friendly web service,which is freely available at http://***.
