Maize plant architecture influences planting density and,in turn,grain yield.Most of the plant architecture-related traits can be described as organ size.We describe a miniature maize mutant,Tiny plant 4(Tip4),which exhibits reduced size of multiple organs and exhibits a semi-dominant monofactorial inheritance characteristic.Positional cloning confirmed that a 4-bp deletion in the NAC TF with transmembrane motif 1-Like(NTL)gene ZmNTL2,denoted as ZmNTL2^(Δ),confers the Tip4 mutation.qRT-PCR showed that ZmNTL2 was expressed in all tested tissues.ZmNTL2 functions as a transcriptional activator and is located in both the nucleus and biomembranes.The mutation does not affect the mRNA abundance of ZmNTL2 locus,but it does result in the loss of transmembrane domain and confines the ZmNTL2^(Δ)protein to the nucleus.Knocking out ZmNTL2 has no effect on maize organ size development,indicating that the 4-bp deletion might be a gain-of-function mutation in organ size regulation.Combining transcriptome sequencing with cytokinin and auxin content determination suggests that the decreased organ size may be possibly mediated by changes in hormone homeostasis.

The extreme instability of pureα-phase FAPbI_(3) under high humidity conditions restricts the highthroughput fabrication in unmodified air environments,resulting in poor performance ofα-phase FAPbI_(3) perovskite devices obtained by scalable fabrication methods.Here we synthesized hyperbranched copper phthalocyanine(HCuPc)as a supramolecular additive with twisted phthalocyanine units to realize the molecular-level encapsulation at the grain boundaries through supramolecular interaction,which greatly broadened the processing window of FAPbI_(3) under high humidity.At the same time,unlike traditional encapsulation layer that carrier can only be collected by tunneling effect,the twisted phthalocyanine ring of HCu Pc in perovskite films is more conducive to hole extraction.Finally,a record efficiency was achieved in pure FAPbI_(3) based inverted structured solar cell by blade-coating to the best of our knowledge,even under unmodified humid air conditions(relative humidity of 65%–85%).The best operational stability of 3D pure FAPbI_(3) devices can also be achieved at the same time and unencapsulated HCuPc-FAPbI_(3) device can even operate with negligible degradation for 100 h in the open air(RH 30%–40%).

ObjectiveTo explore the regulatory effect of Pien Tze Huang (PZH) on targeting partner of NOB1 (PNO1) and it’s down-stream mediators in colorectal cancer (CRC) cells.MethodsQuantitative polymerase chain reaction was performed to determine mRNA levels of PNO1, TP53, and CDKN1A. Western blotting was performed to determine protein levels of PNO1, p53, and p21. HCT-8 cells were transduced with a lentivirus over-expressing PNO1. Colony formation assay was used to detect cell survival in PNO1 overexpression of HCT-8 cells after PZH treatment. Cell-cycle distribution, cell viability and cell apoptosis were performed to identify the effect of PNO1 overexpression on cell proliferation and apoptosis of HCT-8 cells after PZH treatment. Xenograft BALB/c nude mice bearing HCT116 cells transduced with sh-PNO1 or sh-Ctrl lentivirus were evaluated. Western blot assay was performed to detect PNO1, p53, p21 and PCNA expression in tumor sections. Terminal deoxynucleotidyl transferase dUTP nick end labling (TUNEL) assay was used to determine the apoptotic cells in tissues.ResultsPZH treatment decreased cell viability, down-regulated PNO1 expression, and up-regulated p53 and p21 expressions in HCT-8 cells (P<0.05). PNO1 overexpression attenuated the effects of PZH treatment, including the expression of p53 and p21, cell growth, cell viability, cell cycle arrest and cell apoptosis in vitro (P<0.05). PNO1 knockdown eliminated the effects of PZH treatment on tumor growth, inhibiting cell proliferation inhibition and apoptosis induction in vivo (P<0.05). Similarly, PNO1 knockdown attenuated the effects of PZH treatment on the down-regulation of PNO1 and up-regulation of p53 and p21 in vivo (P<0.05).ConclusionThe mechanism by which PZH induces its CRC anti-proliferative effect is at least in part by regulating the expression of PNO1 and its downstream targets p53 and p21.

Deoxygenation has frequently appeared in coastal ecosystems over the past century due to the joint infl uence of increasing anthropogenically induced nutrient inputs and global warming.The semi-enclosed Bohai Sea is a typical system that is prone to deoxygenation,with regular hypoxia events consistently recorded in recent decades.Based on in-situ observation data collected in large-scale voyage surveys in the Bohai Sea during 2008-2017,the seasonal variability in dissolved oxygen(DO)and its controlling mechanisms were studied.The results indicated that in spring and autumn,the DO distributions exhibited similar spatial patterns in the surface and bottom layers,while in summer,its spatial distribution was characterized by large-scale oxygen-poor zones distributed off the Qinhuangdao Coast and the central southern Bohai Sea in the bottom layer.The controlling mechanisms of the DO distribution varied from season to season.Spring and autumn DO distributions were dominated by the seawater temperature.Under the combined eff ects of stratifi cation and decomposition,the summer bottom DO exhibited dual-core distribution.On the one hand,stratifi cation could greatly impede vertical mixing,resulting in reduced bottom DO replenishment.On the other hand,the increased bottom organic matter intensifi ed the decomposition processes,inducing massive DO consumption and elevated dissolved inorganic nitrogen concentrations.In addition,the stronger stratifi cation might be the reason for the more severe deoxygenation in the southern oxygen-poor zones in summer.Our study provides guidance for an in-depth understanding of the DO seasonality in the Bohai Sea and the mechanisms that modulate it and for the improvement of hypoxia forecasts in ocean models.

Background and Aims:Long non-coding RNA small nucleolar RNA host genes(SNHGs)play a critical role in the occurrence and development of tumors.In this study,we aimed to investigate the role of SNHG4 in hepatocellular carcinoma(HCC)and its underlining mechanism.Methods:Datasets were acquired from The Cancer Genome Atlas(TCGA)database.lncLocator 2.0 was used to identify the distribution of SNHG4 in HCC cells.Gene expression,Kaplan-Meier survival,microRNA and transcription factor target analyses were performed with the University of Alabama Cancer(UALCAN)Database,Kaplan-Meier Plotter,LinkedOmics,WebGestalt and gene set enrichment analysis,respectively.Gene Ontology and pathway enrichment analyses and assessment of RNA binding proteins were performed by R software,circlncRNAnet and Encyclopedia of RNA Interactomes(EN-CORI).In addition,CirclncRNAnet and ENCORI were used to find the correlation between SNHG4 and important proteins,while the prognostic value was assessed with the Human Protein Atlas database and Kaplan-Meier Plotter.Results:Expression of SNHG4 in HCC is higher in HCC tissue than in normal healthy liver tissues and is mainly distributed in the nucleus.SNHG4 positively correlated with poor prognosis(p<0.01 for overall survival and recurrence-free survival).Functional enrichment analysis revealed SNHG4 involve-ment with regulation of ribosomal RNA synthesis and the RNA processing and surveillance pathway.SNHG4 is closely associated with miR-154 and miR-206,transcription factor target E2F family and the signaling pathway for MAPK/ERK and mTOR.U2 auxiliary factor 2(U2AF2)showed strong correlation with SNHG4,while low-expression of U2AF2 showed good prognosis.Conclusions:Based on our find-ings,we infer SNHG4 may play a role in the formation of HCC via regulation of tumor-related pathways.

图卷积由于其对图数据的强大表示能力被广泛应用于基于骨架的动作识别任务中.但是现有的图卷积方法在所有帧或通道上都使用共享的图拓扑进行特征聚合,这极大限制了图卷积网络的表示能力.为了解决这些问题,本文提出多维动态拓扑学习图卷积用于动态建模具有时序与通道特异性的拓扑结构.多维动态拓扑学习图卷积主要包含三个组成部分:纯粹节点拓扑学习图卷积(pure Joint topology learning Graph Convolution,J-GC)、动态时序特异性拓扑学习图卷积(Dynamic Temporal-Wise topology learning Graph Convolution,DTW-GC)和通道特异性拓扑学习图卷积(Channel-Wise topology learning Graph Convolution,CW-GC).特别地,在DTW-GC中使用了动态骨架拓扑建模方法(Dynamic Skeleton Topology Learning,DSTL),以高效地建模富含全局时空拓扑特征的动态骨架拓扑.将多维动态拓扑学习图卷积与多尺度时间卷积(Multi-Scale Temporal Convolution,MS-TC)相结合,本文构建了具有强大建模能力的图卷积网络.此外,为了对骨架数据的空间信息进行补充,本文额外引入了相对节点数据和相对骨骼数据进行多流网络的融合.本文所提出的方法在NTU-RGB+D与NTU-RGB+D 120数据集上分别取得了92.64%和89.29%的准确率,超过了当前最先进方法.

RNA-binding proteins(RBPs)-RNA networks have contributed to cancer development.Circular RNAs(circRNAs)are considered as protein recruiters;nevertheless,the patterns of circRNA-protein interactions in colorectal cancer(CRC)are still lacking.Processing bodies(PBs)formed through liquid-liquid phase separation(LLPS)are membrane-less organelles(MLOs)consisting of RBPs and RNA.Previous evidence suggests a connection between PBs dynamics and cancer progression.Despite the increasingly acknowledged crucial role of RBPs and RNA in the accumulation and maintenance of MLOs,there remains a lack of specific research on the interactions between PBs-related RBPs and circRNAs in CRC.Herein,we identify that MEX-3 RNA binding family member A(MEX3A),frequently upregulated in CRC tissues,predicts poorer patient survival.Elevated MEX3A accelerates malignance and inhibits autophagy of CRC cells.Importantly,MEX3A undergoes intrinsically disordered regions(IDRs)-dependent LLPS in the cytoplasm.Specifically,circMPP6 acts as a scaffold to facilitate the interaction between MEX3A and PBs proteins.The MEX3A/circMPP6 complex modulates PBs dynamic and promotes UPF-mediated phosphodiesterase 5A(PDE5A)mRNA degradation,consequently leading to the aggressive properties of CRC cells.Clinically,CRC patients exhibiting high MEX3A expression and low PDE5A expression have the poorest overall survival.Our findings reveal a collaboration between MEX3A and circMPP6 in the regulation of mRNA decay through triggering the PBs aggregation,which provides prognostic markers and/or therapeutic targets for CRC.

Determining reasonable fracturing stage spacing is the key to horizontal well fracturing.Different from traditional stage spacing optimization methods based on the principle of maximum stimulated reservoir volume,in this paper,by considering the integrity of the wellbore interface,a fracture propagation model was established based on displacement discontinuity method and the competition mechanism of multifracture joint expansion,leading to the proposal of an unequal stage spacing optimization model.The results show that in the first stage,the interfacial fractures spread symmetrically along the axis of the central point during that stage,while in the second and subsequent stages,the interfacial fractures of each cluster extend asymmetrically along the left and right sides.There are two kinds of interface connectivity behaviour:in one,the existing fractures first extend and connect within the stage,and in the other,the fractures first extend in the direction close to the previous stage,with the specific behaviour depending on the combined effect of stress shadow and flow competition during hydraulic fracture expansion.The stage spacing is positively correlated with the number of fractures and Young’s modulus of the cement and formation and is negatively correlated with the cluster spacing and horizontal principal stress difference.The sensitivity is the strongest when the Young’s modulus of the cement sheath is 10-20 GPa,and the sensitivity of the horizontal principal stress difference is the weakest.

OBJECTIVE: To investigate the effect of Ganoderma Lucidum Spore Oil(GLSO) on the tumor growth and survival of H22 tumor-bearing mice treated with cyclophosphamide(CTX), and explore the underlying mechanism.METHODS: Allograft H22 hepatocellular carcinoma mouse model was applied to investigate the effect of GLSO on the tumor growth and survival of animals, and Kaplan-Meier survival analysis was used to analyze the life span. Plasma biochemical examination was used to determine the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST), urea(UREA) and creatinine(CRE). Western blot analysis was performed to detect Programmed Death-1(PD-1), Programmed Death Ligand 1(PD-L1), Janus Kinase 2(JAK2),phosphorylated Signal Transducer and Activator of Transcription 3(p-STAT3), and Signal Transducer and Activator of Transcription 3(STAT3) expression. RESULTS: GLSO increased the anti-tumor effect of CTX and prolonged the survival of H22 tumor-bearing mice treated with CTX. Meanwhile, GLSO increased the thymus index and showed no obvious toxicity to liver functions of animals. GLSO also decreased the level of UREA in H22 tumor-bearing mice treated with CTX. Furthermore, GLSO could inhibit the expression of PD-1 in spleen, which was independent of JAK2 expression and STAT3 phosphorylation. However, GLSO did not affect the expression of PD-L1, JAK2, and p-STAT3 in tumor tissue. CONCLUSION: GLSO could strengthen the anti-tumor effect of CTX and prolong the life span of H22 tumorbearing mice, while the underlying mechanism might be relevant to the amelioration effect of thymus function and inhibition of PD-1 expression in spleen. Furthermore, these findings implied the promising role of GLSO in combination with CTX to extend the survival of patients in clinical chemotherapy of hepatocellular carcinoma.
