SQSTM1/p62 regulate breast cancer progression and metastasis by inducing cell cycle arrest and regulating immune cell infiltration

The autophagy adaptor protein SQSTM1/p62 is overexpressed in breast cancer and has been identified as a metastasis-related ***,the mechanism by which SQSTM1/p62 contributes to breast cancer progression and tumor microenvironment remains *** study revealed that silencing SQSTM1/p62 expression suppressed breast cancer progression via regulating cell proliferation and reshaping the tumor microenvironment(TME).here,we found that SQSTM1/p62 was overexpressed in multiple human cancer tissue types and that was correlated with poor patient overall survival(OS)and disease-free survival(DFS).Moreover,we found that short-hairpin RNA(shRNA)-mediated knockdown of p62 expression significantly inhibited cell proliferation,migration,and invasion,and promoted cell death in vitro,as well as suppressed breast cancer growth and lung metastasis in *** addition,flow cytometry analysis of splenocytes and tumor infiltrating lymphocytes(TILs)indicated that the numbers of CD8α+interferon(IFN)-γ+cells(CTLs)and CD4+IFN-γ+(Th1)cells were increased while those of CD4+IL-4+(Th2)cells,tumor-associated macrophages(TAMs)and myeloid-derived suppressor cells(MDSCs)were ***-PCR analyses showed that the gene expression of Th1/Th2 cytokines changed in the tumor *** SQSTM1/p62 suppressed tumor cell lung ***,our results provide strong evidence that silencing tumor cell SQSTM1/p62 inhibited tumor growth and metastasis through cell cycle arrest and TME *** finding provides a novel molecular therapeutic strategy for breast cancer progression and metastasis treatment.

Within the framework of nonrelativistic-QCD factorization,we calculate both the next-to-leading-order relativistic and QCD corrections to prompt J/ψpair production,with feed-down fromψ(2S)mesons,via photon-photon collisions at future e^(+)e^(−)colliders,including the Future Circular Lepton Collider(FCC-ee),the Circular Electron Positron Collider(CEPC),and the Compact Linear Collider(CLIC).We present total cross sections and distributions in single J/ψtransverse momentum and rapidity,as well as in J/ψpair invariant *** relativistic and QCD corrections both turn out to be large and ***,the production rates are large enough for useful experimental studies.

Answering reachability queries with ordered label constraints over labeled graphs

Reachability query plays a vital role in many graph analysis *** researches proposed many methods to efficiently answer reachability queries between vertex *** many real graphs are labeled graph,it highly demands Label-Constrained Reachability(LCR)query inwhich constraint includes a set of labels besides vertex *** researches proposed several methods for answering some LCR queries which require appearance of some labels specified in constraints in the *** that constraint may be a label set,query constraint may be ordered labels,namely OLCR(Ordered-Label-Constrained Reachability)queries which retrieve paths matching a sequence of ***,no solutions are available for ***,we propose DHL,a novel bloom filter based indexing technique for answering OLCR *** can be used to check reachability between vertex *** the answers are not no,then constrained DFS is ***,we employ DHL followed by performing constrained DFS to answer OLCR *** show that DHL has a bounded false positive rate,and it's powerful in saving indexing time and *** experiments on 10 real-life graphs and 12 synthetic graphs demonstrate that DHL achieves about 4.8-22.5 times smaller index space and 4.6-114 times less index construction time than two state-of-art techniques for LCR queries,while achieving comparable query response *** results also show that our algorithm can answer OLCR queries effectively.

Bioisosteres play an important role in pharmaceutical and agricultural compound design that has been developed to enhance the properties of *** a sp3-rich small ring cage hydrocarbons,bicyclo[1.1.1]pentanes(BCPs)were commonly considered as a bioisostere of the para-substituted arenes,internal alkynes,and tert-butyl groups,and its introduction can significantly improve pharmacokinetic properties,including passive permeability,aqueous solubility,metabolic stability[1].Over the past decades,numerous pharmaceutically relevant molecules with biaryl frameworks were improved by the approach of bioisosteric *** strategies for the synthesis of such BCP-aryls derivatives mostly depend on the stepwise C-C formation via the direct addition to bridgehead C-C bond of[1.1.1]propellane to provide active BCP-intermediates and followed by a second transitionmetal cross-coupling ***,these pioneer works were limited by the low step economy,the use of unstable reagents,and harsh conditions in industrial productions.

Creep-Resistant Covalent Adaptable Networks with Excellent Selfhealing and Reprocessing Performance via Phase-Locked Dynamic Covalent Benzopyrazole-Urea Bonds

Covalent adaptive networks(CANs)are capable of undergoing segment rearrangement after being heated,which endows the materials with excellent self-healing and reprocessing performance,providing an efficient solution to the environment pollution caused by the plastic *** main challenge remains in developing CANs with both excellent reprocessing performance and creep-resistance *** this study,a series of CANs containing dynamic covalent benzopyrazole-urea bonds were developed based on the addition reaction between benzopyrazole and isocyanate *** calculation confirmed that relatively low dissociation energy is obtained through undergoing a five-member ring transition state,confirming excellent dynamic property of the benzopyrazole-urea *** verified by the FTIR results,this nice dynamic property can be well maintained after incorporating the benzopyrazole-urea bonds into polymer *** self-healing and reprocessing performance is observed by the 3-ABP/PDMS elastomers owing to the dynamic benzopyrazole-urea *** separation induced by the aggregation of the hard segments locked the benzopyrazole-urea bonds,which also makes the elastomers display excellent creep-resistance *** hard phase locking strategy provides an efficient approach to design CANs materials with both excellent reprocessing and creep-resistance performance.

A generalized deep neural network approach for improving resolution of fluorescence microscopy images

Deep learning is capable of greatly promoting the progress of super-resolution imaging technology in terms of imaging and reconstruction speed,imaging resolution,and *** paper proposes a deep neural network based on a generative adversarial network(GAN).The generator employs a U-Net-based network,which integrates Dense Net for the downsampling *** proposed method has excellent properties,for example,the network model is trained with several different datasets of biological structures;the trained model can improve the imaging resolution of different microscopy imaging modalities such as confocal imaging and wide-field imaging;and the model demonstrates a generalized ability to improve the resolution of different biological structures even out of the *** addition,experimental results showed that the method improved the resolution of caveolin-coated pits(CCPs)structures from 264 nm to 138 nm,a 1.91-fold increase,and nearly doubled the resolution of DNA molecules imaged while being transported through microfluidic channels.
