Deciphering the molecular mechanisms of Simiaowan in the treatment of hyperuricemia: in vivo and in silico approaches

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

Photoactive complexes of nonprecious transition metals, mainly including those in the first-row and partially the second-row of the Periodic table of elements, have received increasing attention in view of their low cost and long-term sustainability. They are recognized as promising alternatives to noble transition metal complex congeners that have been extensively studied in optoelectronic devices, artificial photosynthesis, photocatalysis, biodiagnostics, and therapeutics, etc. This review is devoted to a comprehensive summary on the classical and recent advances on photoactive nonprecious transition metal complexes, including photoactive Zr, V, Cr, Mo, and W complexes, Mn complexes and hybrids, Fe, Co, Ni, and Cu complexes, and Zn and Cd complexes and hybrids. A particular focus is given on the molecular design, modulation of photophysical and photochemical properties, and applications of the representative and lately-developed nonprecious metal complexes. In addition, a perspective on the future development in this field is provided at the end of this review.

Atherosclerosis(AS) is a prevalent condition associated with cardiovascular and cerebrovascular diseases, featuring lipid-laden plaque; however, the insufficient resolution and low sensitivity of traditional medical imaging techniques limit their application in lipid imaging. Currently, the second near-infrared(NIR-II) fluorescence imaging offers promising sensitivity and convenience. However, existing probes have limitations in terms of signal-to-background ratio, penetration depth, and tissue scattering. Herein, we use a molecular regulatory strategy to design a lipid-activatable probe Rh-965 with high specificity and NIR-II emission for improved imaging potential in deep tissues. The potential of Rh-965 for imaging plaque lipids in mice with AS is demonstrated by localizing carotid artery plaques using NIR-II imaging. The development of more stable lipid-activated NIR-II probes can significantly enhance atherosclerosis diagnosis and facilitate in vivo imaging in deeper tissues.

High-precision tip positioning composite feedforward control of linear colocated motion systems with Bi-Loop iterative feedforward tuning

To enhance the tip positioning accuracy and robustness against disturbances of linear colocated motion systems, a novel composite feedforward (COMFF) control with Bi-Loop iterative feedforward tuning (IFFT) is proposed. The tip positioning error consists of the coupled tip residual vibration and linear motor tracking error. Given the influence of nonlinear friction and load flexibility, the topology of COMFF is designed as decoupled structures so that COMFF can suppress the tip residual vibration and compensate for the linear motor tracking error compatibly. Furthermore, Bi-Loop IFFT is built to improve the robustness of COMFF in high-precision motor tracking performance. In the state space of the linear motor tracking error based on the projection theorem, two loops are performed in parallel along the iteration direction. One is the P-type iteration loop that drives linear motor tracking errors to converge to the zero state. The other is the observation loop constructed by the recursive least square with forgetting factors to accelerate the P-type iteration loop. Driven by the P-type iteration loop with the aid of the observation loop, the proposed COMFF is integrated with fast convergence and high robustness when confronted with model disturbances. The effectiveness of the proposed COMFF combined with Bi-Loop IFFT is validated by experiments.

Effects of mesoscale gravity waves on sporadic E simulated by a one-dimensional dynamic model

In addition to being driven by tidal winds,the sporadic E(Es)layers are modulated by gravity waves(GWs),although the effects are not yet comprehensively *** this article,we discuss the effects of mesoscale GWs on the Es layers determined by using a newly developed model,MISE-1D(one-dimensional Model of Ionospheric Sporadic E),with low numerical dissipation and high *** by the wind fields resolved by the high-resolution version of the Whole Atmosphere Community Climate Model with thermosphere and ionosphere extension(WACCM-X),the MISE-1D simulation revealed that GWs significantly influence the evolution of the Es layer above 100 km but have a very limited effect at lower *** effects of GWs are diverse and complex,generally including the generation of fluctuating wavelike structures on the Es layer with frequencies similar to those of the *** mesoscale GWs can also cause increases in the density of Es layers,or they can disperse or diffuse the Es layers and increase their *** addition,the presence of GWs is a key factor in sustaining the Es layers in some cases.

Isolation and Structural Indentification of Herbicidal Toxin Fractions Produced by Pythium aphanidermatum

In order to understand the compsition and structure of herbicidal component of Pythium aphanidermatum,the isolation and structural indentification were *** culture filtrate was extracted by ethyl acetate,petroleum,and chloroform with the same volume respectively and the activity of the crude toxin was *** toxin was separated by using the method of thin layer chromatography（TLC）,then the main fraction was separated by HPLC,and the structure was analyzed by the sepctrum of IR,13C-NMR and *** results showed that the ethyl acetate extracts had the strongest herbicidal *** the method of TLC,the bioassay results showed that the extracts with Rf 0.19 had the strongest effect on weeds and the inhibition to Digitaria sanguinalis and Amaranthus retroflexus reached five levels,and the component was proved to be dimethyl o-phthalate from the spectrum of IR,13C-NMR and 1HNMR,which was one of the components from the toxin,and it had herbicidal activity.

[Solid-phase extraction coupled with high performance liquid chromatography-triple quadrupole mass spectrometry for simultaneous determination of seven coumarins in water samples from drinking water treatment plants]

Chlorinated coumarins, which are as cytotoxic as highly toxic halobenzoquinones toward CHO-K1 cells, have recently been identified as disinfection byproducts in drinking water disinfection processes. Therefore, detecting coumarins in water samples collected at various stages from drinking water treatment plants helps assess the formation of chlorinated coumarins in drinking water. Hence, a simple, rapid, accurate, and sensitive method for quantifying coumarins in water samples is required. In this study, a method was developed based on solid-phase extraction coupled with high performance liquid chromatography-triple-quadrupole mass spectrometry for analyzing seven coumarins in water samples from drinking water treatment plants, including 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, 8-chloro-7-hydroxycoumarin, coumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin. Sample pretreatment involved solid-phase extraction using HLB columns, followed by elution with water and methanol, each containing 0.25% formic acid. The extracted solution was separated via gradient elution using a Phenomenex Luna C18 column (100 mm×2.0 mm, 3 mm) with 0.1% formic acid aqueous solution and methanol as the mobile phases, with analytes detected by triple-quadrupole mass spectrometry equipped with electrospray ionization source in multiple reaction monitoring mode. The matrix effect, precision, and accuracy of the developed method were investigated using raw and treated water as matrices. Matrix effects of 0.84-1.12 were recorded for the detection of 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, and coumarin in raw water, while values of 0.67-0.70 were recorded for 8-chloro-7-hydroxycoumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin in finished water. Micropillar compression using discrete dislocation dynamics and machine learning
