Curcumin induces apoptosis through the mitochondria-mediated apoptotic pathway in HT-29 cells

Objective： To investigate the effects of curcumin on release of cytochrome c and expressions of Bcl-2, Bax, Bad, Bcl-xL, caspase-3, poly ADP-ribose polymerase （PARP）, and survivin of HT-29 cells. Methods： HT-29 cells were treated with curcumin （0-80 μmol/L） for 24 h. The release of cytochrome c from the mitochondria and the apoptosis-related proteins Bax, Bcl-2, Bcl-xL, Bad, caspase-3, PARP, and survivin were determined by Western blot analysis and their mRNA expressions by reverse transcriptase-polymerase chain reaction （RT-PCR）. Results： Curcumin significantly induced the growth inhibition and apoptosis of HT-29 cells. A decrease in expressions of Bcl-2, Bcl-xL and survivin was observed after exposure to 10-80 μmol/L curcumin, while the levels of Bax and Bad increased in the curcumin-treated cells. Curcumin also induced the release of cyto- chrome c, the activation ofcaspase-3, and the cleavage of PARP in a dose-dependent manner. Conclusion： These data suggest that curcumin induced the HT-29 cell apoptosis possibly via the mitoehondria-mediated pathway.

This paper focuses on the development of three types of activated carbon （AC） adsorbents, i.e. granular AC, consolidated AC with chemical binder, and consolidated AC with expanded natural graphite （ENG）. Their thermal conductivity was investigated with the steady-state heat source method and the permeability was tested with nitrogen as the gas source. Results show that the thermal conductivity of granular AC with different sizes al-most maintains a constant at 0.36 W-（m.K）-＇, while the value modestly increases to 0.40 W.（m.K）-＇ for the con- solidated AC with chemical binder. The consolidated AC with ENG at the density of 600 kg. m-3 shows the best heat transfer performance and their thermal conductivity vary from 2.08 W-（m.K）- to 2.61 W. （m.K）-1 according toits fraction of AC. However, the granular AC and consolidated AC with chemical binder show the better permeabil- ity performance than consolidated AC with ENG binder whose permeability changes from 6.98x10-13 m2 to 5.16x10TM m2 and the maximum occurs when the content of AC reaches 71.4% （by mass）. According to the differ- ent thermal properties, the refrigeration application of three types of adsorbents is analyzed.

In comparison to bright pulses, better stability that is not susceptible to loss makes dark pulses accessible for applications in such fields as signal processing, optics sensing, and quantum communication. Here we investigate the dual-wavelength domain-wall dark pulse generation in a graded-index multimode fiber(GIMF) based anomalous dispersion single-mode fiber(SMF) laser. By optimizing intra-cavity nonlinearity and pulse polarization, the mode-locked states can evolve each other between bright pulses, dark pulses, and bright-dark pulse pairs. The evolution mechanism among them may be relevant to the coherent mode superposition, spectral filtering, and mode selection in SMF-GIMF-SMF hybrid-fiber modulation devices that affect the pulse formation and evolution in temporal, frequency, and space domains. These results provide a valuable reference for promoting further development of nonlinear optics and ultrafast optics, in which ultrafast photonic devices, with low cost, simple manufacture as well as wide adaptability, as novel pulsed generation technique, play a vital role.

RESULTS:Treatment with 10-80μmol/L curcumin induced typical features of apoptosis and activated the caspase-3 in HT-29 *** expression of PPARδ, 14-3-3εand VEGF was reduced and the activity of β-catenin/Tcf-4 signaling was inhibited by curcumin treatment. CONCLUSION:Curcumin can induce apoptosis of HT-29 cells and down-regulate the expression of PPARδ,14-3-3εand VEGF in HT-29.

For space-based gravitational wave （GW） detection,the main noise source for massive black hole binaries （MBHBs） is attributed to approximately 10～7double white dwarf binaries in the *** a GW source,the amplitude of the detector response,recorded by a space-based gravitational wave detector,exhibits a modulation effect with a year period when observing the source from various orbital *** the adverse conditions mentioned above,where there is a strong foreground noise and annual modulation in the signals,we employed the wavelet transform and the strong-amplitude relevant orbital position search methods,which allows the weak MBHB sources to achieve higher locating *** detail,for two MBHB sources of lower intensity,the precision of luminosity distance,represented by the ratio ?DL/DLat the 95%confidence level,is enhanced by factors of ～*** angular resolutions,denoted by??s,are enhanced by a factor of ～*** improvements increase the number of detectable GW sources,facilitate multi-messenger follow-up observations and provide constraints on the cosmological constant.

FK506(Tacrolimus)is a systemic immunosuppressant approved by the *** and Drug ***506 has been shown to promote peripheral nerve regeneration,however,its precise mechanism of action and its pathways remain *** this study,we established a rat model of sciatic nerve injury and found that FK506 improved the morphology of the injured sciatic nerve,increased the numbers of motor and sensory neurons,reduced inflammatory responses,markedly improved the conduction function of the injured nerve,and promoted motor function *** findings suggest that FK506 promotes peripheral nerve structure recovery and functional regeneration by reducing the intensity of inflammation after neuronal injury and increasing the number of surviving neurons.

Background Acute subdural haematoma （ASDH） is a common traumatic brain injury with a relatively high mortality rate. However, few studies have examined the factors predicting the outcome of isolated traumatic ASDH. This clinical study examined the hospital mortality and analyzed the risk factors for mortality in patients treated surgically for isolated traumatic ASDH.Methods We collected 308 consecutive patients who underwent neurosurgery for isolated traumatic ASDH between January 1999 and December 2007 and used multivariate Logistic regression analysis to evaluate the influence of 11 clinical variables on hospital mortality.Results The overall hospital mortality was 21.75% （67/308）. Age （OR=1.807）, preoperative Glasgow Coma Score （OR=0.316）, brain herniation （OR=2.181） and the time from trauma to decompression （OR=1.815） were independent predictors of death, while no independent association was observed between hospital mortality and haematoma volume, midline shift, acute brain swelling or brain herniation duration, although these variables were correlated with hospital mortality in univariate analyses.Conclusions This study identified the risk factors for hospital mortality in patients who underwent surgical treatment for isolated traumatic ASDH. An increased risk of death occurs in patients who are over 50 years of age and have lower preoperative Glasgow Coma Scores, the presence of brain herniation and a long interval between trauma and decompression. The findings should help clinicians determine management criteria and improve survival.
