Prolapse gastropathy syndrome may be a predictor of pathologic acid reflux

AIM： To assess the occurrence of gastric acid reflux into the esophagus in endoscopically confirmed prolapse gastropathy syndrome （PGS）. METHODS： Using ambulatory esophageal pH measurement （BRAVOTM wireless esophageal pH monitoring system）, twenty-six patients with PGS were compared with twenty-one patients with erosive esophagitis （EE） as controls. We assessed several reflux parameters, including the percentage of total time at pH 〈 4, and the DeMeester score. RESULTS： There were no statistical differences between the PGS group and the EE group as to mean age, sex ratio and pH recording time. The EE group showed more severe reflux than the PGS group, as evaluated in terms of the longest duration of reflux, the number of reflux episodes, the number of reflux episodes lasting 〉 5 min, the total time with pH 〈 4 during acid reflux episodes, and the DeMeester score, but none of these parameters showed statistically significant difference. Although 53.8% （14/26） of the PGS group and 76.2% （16/22） of the EE group demonstrated pathologic acid reflux （DeMeester score 〉 14.72）, there was no statistically significant difference between the two groups in the incidence of pathologic acid reflux （P = 0.11）. CONCLUSION： There was no statistically significant difference in pathologic acid reflux between the PGS and EE group. These data suggest that endoscopically diagnosed PGS might be a predictor of pathologic acid reflux.

Phosphorylated p70S6K in urothelial carcinoma of the tumor recurrence noninvasive low-grade bladder： correlation with

We investigated whether inhibiting phosphorylated p70S6K （p-p70S6K） suppresses the proliferation and growth of noninvasive low-grade urothelial carcinoma （LG-URCa） in vitro and whether p-p70S6K can serve as a predictive biomarker for the recurrence of noninvasive LG-URCa of the bladder in patients. We constructed a tissue microarray （TMA） for 95 LG-URCa and 35 benign urothelium samples and performed immunohistochemical staining for p-p70S6K and p-4E-BP1. A Cox regression model was used to investigate the predictive factors for recurrence of LG-URCa. We investigated the dose-dependent antiproliferative effect of rapamycin, its antiproliferative effect and the growth-inhibition effect of p70S6K siRNA transfection in RT4 and 253J cell lines. The pT1 staged group （P 〈 0.05; hazard ratio （HR）, 2.415） and the high p-p70S6K staining group （P 〈 0.05; HR, 2.249） were independent factors for predicting recurrence. Rapamycin inhibited RT4 and 253J cell proliferation in a dose-dependent manner （r = -0.850, P 〈 0.001 in RT4 cells; r = -0.835, P 〈 0.001 in 253J cells）. RT4 and 253J cell proliferation and growth were inhibited by the transfection of p70S6K siRNA and rapamycin, respectively （P 〈 0.05）. Transfection of p70S6K siRNA resulted in inhibitory effects on cell proliferation and growth that were similar to those of rapamycin. Our results suggest that inhibiting p70S6K phosphorylation is important to prevent recurrence and that p70S6K phosphorylation can be used as a molecular biomarker to predict recurrence of certain LG-URCa of the bladder.

Efficacy of horse oil on lipopolysaccharide-induced inflammation in human keratinocytes

OBJECTIVE: To investigate the efficacy of horse oil on lipopolysaccharide(LPS)-induced inflammation in human ***: Western blot analysis was performed to measure the expression of cyclooxygenase-2(COX-2) and IκBα. ELISA was used to analyze prostaglandin E2(PGE2) ***: Horse oil decreased LPS-induced COX-2 and PGE2 levels in a dose-dependent manner. Nuclear factor-kappa B(NF-κB) plays a key role in the expression of inflammatory cytokines and mediators. Therefore, we investigated the influence of horse oil on the NF-κB signaling pathways. Horse oil inhibited translocation of NF-κB from the cytosol to the nucleus. Furthermore, LPS-induced degradation of IκBα was recovered by horse oil. The activation of p38 mitogen-activated protein kinase(MAPK) reportedly induces degradation of IκBα In agreement with this, LPS activated p38 MAPK and caused IκBα degradation. Conversely, horse oil inhibited LPS-induced p38 MAPK activation and IκBαdegradation. In addition, a specific p38 MAPK inhibitor, SB203580, blocked IκBα ***: Horse oil decreased COX-2 and PGE2 by inhibiting p38 MAPK activation, IκBα degradation, and the translocation of NF-κB.

High-efficiency technique based on dielectrophoresis for assembling metal,semiconductor,and polymer nanorods

This paper presents a high-efficiency technique based on dielectrophoresis (DEP) for assembling metal, semiconductor, and polymer nanorods, which are synthesized by electrochemical deposition (ECD). The assembly patterns of these nanorods (width: 20 nm; length: 7 μm) were designed using a finite element method (FEM) simulation tool. Further, these nanorods were used in our experiment after their assembly patterns were fabricated. The assembly yield was found to be approximately 70% at an AC voltage of 30 Vp-p and at frequencies of 20 and 30 kHz, and the DC voltage prevented the random alignment of the nanorods at the edge of the assembly pattern. Moreover, the above-mentioned nanorods, which had different permittivities, were found to have similar assembly yields. The proposed method can be improved and applied to nanostructure device fabrication.

Chaga mushroom (Inonotus obliquus) induces G0/G1 arrest and apoptosis in human hepatoma HepG2 cells

AIM: To investigate the anti-proliferative and apoptotic effects of Chaga mushroom (Inonotus obliquus) water extract on human hepatoma cell lines,HepG2 and Hep3B cells. METHODS: The cytotoxicity of Chaga extract was screened by 3-4,5-dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide (MTT) assay. Morphological observation,flow cytometry analysis,Western blot were employed to elucidate the cytotoxic mechanism of Chaga extract. RESULTS: HepG2 cells were more sensitive to Chaga extract than Hep3B cells,as demonstrated by markedly reduced cell viability. Chaga extract inhibited the cell growth in a dose-dependent manner,which was accompanied with G0/G1-phase arrest and apoptotic cell death. In addition,G0/G1 arrest in the cell cycle was closely associated with down-regulation of p53,pRb,p27,cyclins D1,D2,E,cyclin-dependent kinase (Cdk) 2,Cdk4,and Cdk6 expression. CONCLUSION: Chaga mushroom may provide a new therapeutic option,as a potential anticancer agent,in the treatment of hepatoma.

High-strength bainitic steels have created a lot of interest in recent times because of their excellent combination of strength,ductility,toughness,and high ballistic mass *** steels have great potential in the fabrication of steel armor *** various approaches and methods have been conducted to utilize the retained austenite(RA)in the bainitic matrix to control mechanical properties,very few attempts have been conducted to improve ballistic performance utilizing transformationinduced plasticity(TRIP)*** this study,high-strength bainitic steels were designed by controlling the time of austempering process to have various volume fractions and stability of RA while maintaining high *** dynamic compressive and ballistic impact tests were conducted,and the relation between the effects of TRIP on ballistic performance and the adiabatic shear band(ASB)formation was *** results show for the first time that an active TRIP mechanism achieved from a large quantity of metastable RA can significantly enhance the ballistic performance of high-strength bainitic steels because of the improved resistance to ASB ***,the ballistic performance can be effectively improved by a very short austempering time,which suggests that the utilization of active TRIP behavior via tuning RA acts as a primary mechanism for significantly enhancing the ballistic performance of high-strength bainitic steels.

Do microRNA 96, 145 and 221 expressions really aid in Lhe prognosis of prostate carcinoma？

MicroRNAs （miRs） are small noncoding RNAs that have been reported to be premising diagnostic tools. We used quantitative real-time reverse transcription PCR （RT-qPCR） to analyze differentially expressed miRNAs in prostate tumor samples to determine its prognostic value. From 2007 to 2009, tumor tissues were obtained from 73 radical prostatectomy specimens. Differentially expressed miR-96, -145 and -221 were validated by TaqMan RT-qPCR using all 73 tissues. The prognostic value was assessed in terms of biochemical recurrence using Kaplan-Meier and Cox regression analyses. For our patient cohort, the mean age was 64.7 years （50-76 years） and the mean prostate-specific antigen （PSA） was 7.5 ng m1-1. During the follow-up period （mean, 19.4 months）, 14 of 73 （19.2%） patients developed biochemical recurrence. Expression of miR-96, -145 and -221 correlated strongly with each other, but there were no correlations between miRNA expression and clinicopathologic parameters. Kaplan-Meier survival curves using the log-rank test showed a decreased biochemical recurrence-free interval with pathologic stage （P〈O.O01）. In addition, patients with Gleason scores over 8, compared with those with a Gleason score of 6, showed a decreased biochemical recurrence-free interval in Kaplan-Meier analysis （P=O.O01）. However, expression of miR-96, -145 and -221 did not correlate with the biochemical recurrence interval in Kaplan-Meier survival curves or by multivariate analysis using the Cox proportional hazard regression model, either. In conclusion, we did not observe a significant correlation between the expression of miR-96, -145 and -221 and clinicopathologic parameters. To utilize miRNA as a diagnostic tool in clinical practice, more research is needed to understand miRNA mechanisms, identify miRNA targets, and further characterize miRNA function.

Fabrication of 3D Printed PCL/PEG Polyblend Scaffold Using Rapid Prototyping System for Bone Tissue Engineering Application

Three-dimensional （3D） printing is a novel process used to manufacture bone tissue engineered scaffolds. This process allows for easy control of the architecture at the micro structure. However, the scaffold properties are typically limited in terms of cellular activity at the scaffold surface due to the printed materials properties. In this study, we developed a polycaprolactone （PCL） blended with polyethylene glycol （PEG） 3D printed scaffold using a rapid prototyping system. The manufactured scaffolds were then washed out to form small pores on the surface in order to improve the scaffolds hydrophilicity. We analyzed the resultant material by using Scanning Electron Microscopy （SEM）, water absorption, water contact angle, in vitro WST-1, and the Bradford assay. Additionally, cells incubated on the fabricated scaffolds were visualized by Confocal Laser Scanning Microscopy （CLSM）. The developed scaffolds exhibited small pores on the strand surface which served to increase hydrophilicity as well as improve cellular proliferation and increase total protein content. Our findings suggest that the presence of small pores on the scaffolds can be used as an effective tool for improving implant cellular interaction. This research indicates that these modified scaffolds can be considered useful for bone tissue engineering applications to improve human health.

Construction of random sheared fosmid library from Chinese cabbage and its use for Brassica rapa genome sequencing project

As a part of the Multinational Genome Sequencing Project of Brassica rapa, linkage group R9 and R3 were sequenced using a bacterial artificial chromosome （BAC） by BAC strategy. The current physical contigs are expected to cover approximately 90% euchromatins of both chromosomes. As the project progresses, BAC selection for sequence extension becomes more limited because BAC libraries are restriction enzyme-specific. To support the project, a random sheared fosmid library was constructed. The library consists of 97536 clones with average insert size of approximately 40 kb corresponding to seven genome equivalents, assuming a Chinese cabbage genome size of 550 Mb. The library was screened with primers designed at the end of sequences of nine points of scaffold gaps where BAC clones cannot be selected to extend the physical contigs. The selected positive clones were end-sequenced to check the overlap between the fosmid clones and the adjacent BAC clones. Nine fosmid clones were selected and fully sequenced. The sequences revealed two completed gap filling and seven sequence extensions, which can be used for further selection of BAC clones confirming that the fosmid library will facilitate the sequence completion of B. rapa.

Superhydrophobic and antireflective nanograss-coated glass for high performance solar cells

We present a facile method for producing superhydrophobic nanograss-coated （SNGC） glass surfaces that possess both reduced reflectivity and self-cleaning properties at the air/glass interface. The refractive index of a CaF2 nanograss （NG） layer on a glass substrate, deposited by glancing angle vapor deposition, is 1.04 at 500 nm, which is the second-lowest value ever reported so far. The fluorinated NG layer gives rise to a high water contact angle （〉150°） and very efficient cleaning out of dust with water drops. Using the dual functionalities of the SNGC glass, we demonstrate superhydrophobic and antireflective organic photovoltaic cells with excellent power conversion efficiency.
