Intranasal boosting with RBD-HR protein vaccine elicits robust mucosal and systemic immune responses

The emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants has decreased the efficacy of SARs-CoV-2 vaccines in containing coronavirus disease 2019(CoVID-19)over time,and booster vaccination strategies are urgently necessitated to achieve sufficient protection.Intranasal immunization can improvemucosal immunity,offer-ing protection against the infection and sustaining the spread of SARS-CoV-2.In this study,an intranasal booster of the RBD-HR vaccine after two doses of the mRNA vaccine significantly increased the levels of specific binding antibodies in serum,nasal lavage fluid,and bronchoal-veolar lavage fluid compared with only two doses of mRNA vaccine.After intranasal boosting with the RBD-HR vaccine,the levels of serum neutralizing antibodies against prototype and variant strains of SARS-Cov-2 pseudoviruses weremarkedly higher than those in mice receiving mRNA vaccine alone,and intranasal boosting with the RBD-HR vaccine also inhibited the bind-ing of RBD to hACE2 receptors.Furthermore,the heterologous intranasal immunization regimen promoted extensive memory T cell responses and activated CD103+dendritic cells in the respiratory mucosa,and potently enhanced the formation of T follicular helper cells and germinal center B cells in vital immune organs,including mediastinal lymph nodes,inguinal lymph nodes,and spleen.Collectively,these data infer that heterologous intranasal boosting with the RBD-HR vaccine elicited broad protective immunity against SARS-CoV-2 both locallyandsystemically.

Green manufacturing for achieving carbon neutrality goal requires innovative technologies:A bibliometric analysis from 1991 to 2022

Recent years have seen a significant increase in interest in green manufacturing as a key driver of global carbon-neutral efforts and sustainable development.To find the research hotspots of green manufacturing and reveal future research trends,this study reviewed and analyzed research articles from the Web of Science database on green manufacturing from1991 to 2022 using a bibliometric method.The findings indicate a significant rise in the number of articles related to green manufacturing since the 2010s.Moreover,there has been an increase in the involvement of scholars from developing countries such as China and India in this field.Based on the literature review and bibliometric cluster analysis on green manufacturing,we believed that future research may continue following the lines of intelligent technology integration,adoption of frontier engineering techniques,and industry development in line with carbon reduction targets.A framework for future green manufacturing development is proposed,with a focus on Chinese policies.The framework could provide policy implications for developing countries looking to pursue opportunities for development in green manufacturing.

A New Method for the Aging Evaluation of Oil-Paper Insulation Using η-Butanol and Methanol

To improve the accuracy and efficiency of the aging life prediction and assessment of transformer oil-paper insulation,and to make up for the deficiencies of traditional characterizers,such as 2-furfural,carbon monoxide,and carbon dioxide,a method for the simultaneous determination of methanol,ethanol,n−propanol,and η-butanol in oil with a single injection is established by headspace-gas chromatography-mass spectrometry.The measured results show that the determination limits of the four alcohol characterizers can be controlled to 10μg/kg level.Based on this method,the change patterns of the above four alcohols with thermal aging time and degree of polymerization are obtained through thermal aging experimental research.Ethanol,η-propanol,and η-butanol in oil indicate nearly linear correlations with thermal aging time and degree of polymerization,similar to that of methanol.By analyzing 52 sets of measured data of 500 kV EHV transformers in operation,η-butanol is found to have excellent performance,and a new method to evaluate the aging state of oil-paper insulation employing η-butanol and methanol is proposed along with the aging attention value model.The measured data of 500 kV EHV transformers in operation indicate that the combination of η-butanol and methanol as the preferred characterizers can effectively compensate for the shortcoming of traditional characterizers in the early stages of aging,and the feasibility of the method is verified.Two possible pathways for the generation of η-butanol by cellulose cleavage during the aging of oil-paper are proposed from the chemical structure of cellulose.

The incidence and mortality rates of cancer are increasing every year worldwide but the survival rate of cancer patients is still unsatisfactory.Therefore,it is necessary to further elucidate the molecular mechanisms involved in tumor development and drug resis-tance to improve cancer cure or survival rates.In recent years,autophagy has become a hot topic in the field of oncology research,which plays a double-edged role in tumorigenesis,pro-gression,and drug resistance.Meanwhile,long non-coding RNA(lncRNA)has also been shown to regulate autophagy,and the two-sided nature of autophagy determines the dual regulatory role of autophagy-related IncRNAs(ARlncRNAs).Therefore,ARlncRNAs can be effective ther-apeutic targets for various cancers.Furthermore,the high abundance and stability of ARlncR-NAs in tumor tissues make them promising biomarkers.In this review,we summarized the roles and mechanisms of ARlncRNAs in tumor cell proliferation,apoptosis,migration,invasion,drug resistance,angiogenesis,radiation resistance,and immune regulation.In addition,we described the clinical significance of these ARlncRNAs,including as biomarkers/therapeutic targets and their association with clinical drugs.

Growth performance, serum parameters, inflammatory responses,intestinal morphology and microbiota of weaned piglets fed 18% crude protein diets with different ratios of standardized ileal digestible isoleucine to lysine

The present study was to explore the Ile requirement of piglets fed 18% crude protein(CP) diets. Two hundred and fifty 28-day-old Duroc × Landrace × Yorkshire piglets(8.37 ± 1.92 kg) were randomly divided into 5 dietary treatments(10 piglets per replicate, 5 barrows and 5 gilts per replicate) with 45%,50%, 55%, 60%, 65% standardized ileal digestible(SID) Ile-to-Lys ratios, and the SID Lys was formulated to1.19%. The experimental design consisted of two phases(d 1 to 14 and d 15 to 28). Results showed that average daily gain(ADG) had a tendency to quadratically increase as the SID Ile-to-Lys ratio increased(P = 0.09), and the optimum SID Ile-to-Lys ratios required to maximize ADG were 48.33% and 54.63% for broken-line linear model and quadratic polynomial model, respectively. Different SID Ile-to-Lys ratios had no significant effects on average daily feed intake and gain-to-feed ratio. Dry matter(P < 0.01), CP(P = 0.01), ether extract(P = 0.04), gross energy(P < 0.01) and organic matter(P < 0.01) digestibility increased quadratically. Serum total cholesterol levels decreased linearly(P = 0.01) and quadratically(P < 0.01); aspartate aminotransferase(P < 0.01), interleukin-1β(P = 0.01), and tumor necrosis factor-a(P < 0.01) levels decreased quadratically; immunoglobulin G(P = 0.03) and immunoglobulin M(P = 0.01)concentrations increased quadratically. Serum Ser levels decreased linearly(P < 0.01) and quadratically(P = 0.01); Glu(P = 0.02), Arg(P = 0.05), and Thr(P = 0.03) levels decreased quadratically; Gly(P < 0.01)and Leu(P = 0.01) levels decreased linearly; Ile(P < 0.01) concentration increased linearly. Duodenal villus height(P < 0.01) and villus height to crypt depth ratio(P < 0.01) increased quadratically. The deficiency or excess of Ile decreased short chain fatty acid-producing bacteria abundance and increased pathogenic bacteria abundance. Overall, taking ADG as the effect index, the optimum SID Ile-to-Lys ratios of piglets offered 18% CP diets were 48.33% and 54.63% based on two different statistical models

Hot Deformation Behavior and Microstructures Evolution of GNP-Reinforced Fine-Grained Mg Composites

Graphene nanoplates(GNPs)-reinforced magnesium matrix composites have been attracted great attention.However,knowledge is lack for the hot deformation behavior of GNP-reinforced magnesium(GNPs/Mg)composite.In this study,the fine-grained GNPs/Mg composite was fabricated by powder metallurgy process followed by extrusion.The hot deformation behavior,microstructure evolution and dynamic recrystallization(DRX)mechanism of fine-grained GNPs/Mg composite were investigated by hot compression test and electron back-scatter diffraction(EBSD).The hot compression tests of the composite were conducted at temperatures between 423 and 573 K with the strain rates from 0.001 to 1 s^(-1).The strain compensated power law equation was established to describe the hot deformation behavior of the composites.The stress exponent and activation energy of the composite are 7.76 and 83.23 kJ/mol,respectively,suggesting that the deformation mechanism is grain boundary slip controlled dislocation climb creep.The abnormally high stress exponent and activation energy are unattainable in the composite due to the fine grain size of the composites and the absence of Zener pinning and Orowan effects of GNPs reinforcement.The grain size increases with the decrease in Zener-Hollomn(Z)parameter,which can be well fitted by power-law relationship.With the increase in grain size and decrease in Z parameter,the geometrically necessary dislocation density decreases,which shows the approximately power-law relationship.A random and weak texture was formed after hot compression.The discontinuous dynamic recrystallization and continuous dynamic recrystallization mechanism dominated the DRX behavior at 473 K/0.001 s^(-1) and 573 K/0.001 s^(-1),respectively.

Competitive interactions and coexistence of sympatric flagship carnivores in Asia

Understanding the competition and coexistence of flagship carnivores is key to creating strategies for their conservation in the face of global carnivore declines.Although studies exploring the dynamics and competition between tigers(Panthera tigris)and leopards(P.pardus)span decades,there is a lack of understanding regarding the factors that influence their coexistence mechanisms on a broad scale,as well as the drivers determining their exploitative and interference competition.We gathered a comprehensive list of research papers among which 36 papers explored the interspecific interactions between tigers and leopards and tested the influence of biotic and abiotic factors on the coexistence mechanisms along three dimensions using multiple response variables regression models;we also tested the influence of ecological drivers determining the exploitative or interference competition between tigers and leopards.Elevation and ungulate density were the most important predictors in regulating the coexistence mechanisms.Tigers and leopards exhibited more positive relations/higher overlaps as elevation increased in the spatial niche.In addition,they showed a higher dietary overlap in the prey-rich regions.We determined that interference competition between tigers and leopards was less frequently observed in habitats with dense tree cover and homogeneous vegetation structures.Meanwhile,studies with multiple metrics would promote the detection of interference competition.Our study provides new insight into the competitive interactions and coexistence mechanisms of tigers and leopards on a broad scale.Policy-makers and managers should pay more attention to the factors of elevation,prey abundance,and habitat structures for the conservation of tigers and leopards.

Slippery hydrogel with desiccation-tolerant‘skin’for high-precision additive manufacturing

Hydrogels inevitably undergo dehydration,structural collapse,and shrinkage deformation due to the uninterrupted evaporation in the atmosphere,thereby losing their flexibility,slipperiness,and manufacturing precision.Here,we propose a novel bioinspired strategy to construct a spontaneously formed‘skin’on the slippery hydrogels by incorporating biological stress metabolites trehalose into the hydrogel network,which can generate robust hydrogen bonding interactions to restrain water evaporation.The contents of trehalose in hydrogel matrix can also regulate the desiccation-tolerance,mechanical properties,and lubricating performance of slippery hydrogels in a wide range.Combining vat photopolymerization three-dimensional printing and trehalose-modified slippery hydrogels enables to achieve the structural hydrogels with high resolution,shape fidelity,and sophisticated architectures,instead of structural collapse and shrinkage deformation caused by dehydration.And thus,this proposed functional hydrogel adapts to manufacture large-scale hydrogels with sophisticated architectures in a long-term process.As a proof-of-concept demonstration,a high-precision and sophisticated slippery hydrogel vascular phantom was easily fabricated to imitate guidewire intervention.Additionally,the proposed protocol is universally applicable to diverse types of hydrogel systems.This strategy opens up a versatile methodology to fabricate dry-resistant slippery hydrogel for functional structures and devices,expanding their high-precision processing and broad applications in the atmosphere.

Dynamics of an impurity ion transport in oil-paper insulation under various electric fields

The ionic transport process is an important counterpart to charge transport in oil-paper insulation,and it significantly impacts oil flow electrification at the oil-paper interface.Despite this,the dynamics of this phenomenon and the underlying mechanisms remain unclear,particularly at the molecular level.To understand this fundamental aspect,we conduct Molecular Dynamics study on the transport behaviour of an impurity ion in different oil-paper insulation models under various external electric fields.Different influence factors,such as external electric fields,temperatures,and local structural characteristics,are investigated in relation to the corresponding ionic mobility in different polymer models.According to the simulations,ionic mobility and its response to electric fields are higher in weaker electrostatic models.As a result,mineral oil exhibits the highest ionic mobility and the most substantial enhancement of ionic mobility by external electric fields,followed by vegetable oil,oil-paper blends,and insulating paper.This significant deviation in ionic mobility between oil and paper leads to the formation of an electric double layer near the oil-paper interface.The underlying physical mechanisms of different ionic mobility and its response to the electric field in different polymer models could be explained by the different polymer structural influences in terms of interaction energy and coordination numbers in a static manner,as well as by the interactions between the impurity ion and its surrounding atoms in terms of lifetime correlation functions and velocity autocorrelation functions in a dynamic manner.In addition,the influence of temperature on ionic mobility in mineral and vegetable oil is examined,and their activation energies are calculated.Advancing in the fundamental understanding of the dynamics of the ion transport process in oil-paper insulation is vital to improving their insulating properties for oil-impregnated power transformers.

Mycoplasma pneumoniae(M.pneumoniae),primarily transmitted through respiratory droplets when infected individuals cough or sneeze,is a common cause of communityacquired pneumonia,especially among school-age children and adolescents.The infection occurs endemically with an epidemic peak every few years.The worldwide incidence confirmed by direct test methods was reported to be 8.61%between 2017 and 2020 across all age groups[1].
