Cell softness reveals tumorigenic potential via ITGB8/AKT/glycolysis signaling in a mice model of orthotopic bladder cancer

Background:Bladder cancer,characterized by a high potential of tumor recurrence,has high lifelong monitoring and treatment costs.To date,tumor cells with intrinsic softness have been identified to function as cancer stem cells in several cancer types.Nonetheless,the existence of soft tumor cells in bladder tumors remains elusive.Thus,our study aimed to develop a microbarrier microfluidic chip to efficiently isolate deformable tumor cells from distinct types of bladder cancer cells.Methods:The stiffness of bladder cancer cells was determined by atomic force microscopy(AFM).The modified microfluidic chip was utilized to separate soft cells,and the 3D Matrigel culture system was to maintain the softness of tumor cells.Expression patterns of integrinβ8(ITGB8),protein kinase B(AKT),and mammalian target of rapamycin(mTOR)were determined by Western blotting.Double immunostaining was conducted to examine the interaction between F-actin and tripartite motif containing 59(TRIM59).The stem-cell-like characteristics of soft cells were explored by colony formation assay and in vivo studies upon xenografted tumor models.Results:Using our newly designed microfluidic approach,we identified a small fraction of soft tumor cells in bladder cancer cells.More importantly,the existence of soft tumor cells was confirmed in clinical human bladder cancer specimens,in which the number of soft tumor cells was associated with tumor relapse.Furthermore,we demonstrated that the biomechanical stimuli arising from 3D Matrigel activated the F-actin/ITGB8/TRIM59/AKT/mTOR/glycolysis pathways to enhance the softness and tumorigenic capacity of tumor cells.Simultaneously,we detected a remarkable up-regulation in ITGB8,TRIM59,and phospho-AKT in clinical bladder recurrent tumors compared with their non-recurrent counterparts.Conclusions:The ITGB8/TRIM59/AKT/mTOR/glycolysis axis plays a crucial role in modulating tumor softness and stemness.Meanwhile,the soft tumor cells become more sensitive to chemotherapy after stiffening,that offers new insights fo

Thorium anomaly on the lunar surface and its indicative meaning

The Moon has been divided into three terranes:Procellarum KREEP Terrane(PKT),Feldspathic Highland Terrane(FHT),and South Pole-aitken Terrane(SPAT),using globally measured Th and FeO.Many lunar evolu-tion models have predicted that a lunar magma ocean will produce a residual layer enriched in incompatible elements such as K,REE,and P(i.e.,KREEP)in the late age of crys-tallization;and that the distribution of thorium can be used as a proxy for determining the global distribution of KREEP.The thorium distribution in these three terranes is inhomo-geneous.The highest concentration of thorium is in PKT,the medium concentration of thorium is in SPAT,and almost none in FHT.Then what is the specific distribution in each of the terrane and what enlightenment can it tell us?Here we present and describe the detailed thorium distribution in PKT,SPAT,and FHT and provide some information for the origin of asymmetries on the lunar surface.

Control of highly pathogenic avian influenza through vaccination

The stamping-out strategy has been used to control highly pathogenic avian influenza viruses in many countries,driven by the belief that vaccination would not be successful against such viruses and fears that avian influenza virus in vaccinated birds would evolve more rapidly and pose a greater risk to humans.In this review,we summarize the successes in controlling highly pathogenic avian influenza in China and make suggestions regarding the requirements for vaccine selection and effectiveness.In addition,we present evidence that vaccination of poultry not only eliminates human infection with avian influenza virus,but also significantly reduces and abolishes some harmful characteristics of avian influenza virus.

Nonnegative Matrix Factorization(NMF)is one of the most popular feature learning technologies in the field of machine learning and pattern recognition.It has been widely used and studied in the multi-view clustering tasks because of its effectiveness.This study proposes a general semi-supervised multi-view nonnegative matrix factorization algorithm.This algorithm incorporates discriminative and geometric information on data to learn a better-fused representation,and adopts a feature normalizing strategy to align the different views.Two specific implementations of this algorithm are developed to validate the effectiveness of the proposed framework:Graph regularization based Discriminatively Constrained Multi-View Nonnegative Matrix Factorization(GDCMVNMF)and Extended Multi-View Constrained Nonnegative Matrix Factorization(ExMVCNMF).The intrinsic connection between these two specific implementations is discussed,and the optimization based on multiply update rules is presented.Experiments on six datasets show that the effectiveness of GDCMVNMF and ExMVCNMF outperforms several representative unsupervised and semi-supervised multi-view NMF approaches.

A novel adaptive coordinated tracking control scheme for a morphing aircraft with telescopic wings

Inspired by flight biology,morphing flight technology has great potential to improve the adaptability and maneuverability of aircraft.This paper is devoted to the flight control problem of morphing aircraft,and aimed at safe and fuel-saving flight through morphing actively.Specifically,the longitudinal dynamics of a morphing aircraft with telescopic wings is modelled as a strict-feedback nonlinear system.Through fitting the expression of aerodynamic parameters by the mor-phing ratio,the model uncertainties induced by morphing errors are embedded in the dynamics.To meet the safety and fuel-saving requirements,an Adaptive Coordinated Tracking Control Scheme(ACTCS)is then proposed,which consists of a morphing control module and a tracking control module.For the morphing control module,an on-line morphing decision model is given in an optimization process with respect to the morphing ratio,and a second-order tracking filter is introduced to smooth the decision output and ensure the physical realizability.For the tracking control module,the novel adaptive controllers for the velocity and altitude subsystems are proposed based on the dynamic surface control method,in which adaptive mechanisms are designed to com-pensate for the model uncertainties.Finally,the proposed ACTCS is simulated in nine different cases of the test flight mission,to verify its effectiveness,robustness and fuel-saving effect.

Benefit of aerosol reduction to winter wheat during China's clean air action:A case study of Henan Province

A strongly declining aerosol radiative effect has been observed in China since 2013 after implementing the clean air action,yet its impact on wheat(Triticum aestivum L.)production remains unclear.We use satellite measures and a biophysical crop model to assess the impact of aerosol-induced radiative perturbations on winter wheat production in the agricultural belt of Henan province from 2013 to 2018.After calibrating parameters with the extended Fourier Amplitude Sensitivity Test(EFAST)and the generalized likelihood uncertainty estimation(GLUE)method,the DSSAT CERES-Wheat model was able to simulate crop biomass and yield more accurately.We found that the aerosol negatively impacted wheat biomass by 21.87%and yield by 22.48%from 2006 to 2018,and the biomass effects from planting to anthesis were more significant compared to anthesis to maturity.Due to the strict clean air action,under all-sky conditions,the surface solar shortwave radiation(SSR)in 2018 increased by about 7.08%over 2006-2013 during the wheat growing seasons.As a result of the improvement of crop photosynthesis,winter wheat biomass and yield increased by an average of 5.46%and 2.9%,respectively.Our findings show that crop carbon uptake and yield will benefit from the clean air action in China,helping to ensure national food and health security.

Cellular mechanotransduction in health and diseases:from molecular mechanism to therapeutic targets

Cellular mechanotransduction,a critical regulator of numerous biological processes,is the conversion from mechanical signals to biochemical signals regarding cell activities and metabolism.Typical mechanical cues in organisms include hydrostatic pressure,fluid shear stress,tensile force,extracellular matrix stiffness or tissue elasticity,and extracellular fluid viscosity.Mechanotransduction has been expected to trigger multiple biological processes,such as embryonic development,tissue repair and regeneration.However,prolonged excessive mechanical stimulation can result in pathological processes,such as multi-organ fibrosis,tumorigenesis,and cancer immunotherapy resistance.Although the associations between mechanical cues and normal tissue homeostasis or diseases have been identified,the regulatory mechanisms among different mechanical cues are not yet comprehensively illustrated,and no effective therapies are currently available targeting mechanical cue-related signaling.This review systematically summarizes the characteristics and regulatory mechanisms of typical mechanical cues in normal conditions and diseases with the updated evidence.The key effectors responding to mechanical stimulations are listed,such as Piezo channels,integrins,Yes-associated protein(YAP)/transcriptional coactivator with PDZ-binding motif(TAZ),and transient receptor potential vanilloid 4(TRPV4).We also reviewed the key signaling pathways,therapeutic targets and cutting-edge clinical applications of diseases related to mechanical cues.

DOK3 promotes proliferation and inhibits apoptosis of prostate cancer via the NF-κB signaling pathway

Background: DOK3 (Downstream of kinase 3) is involved primarily with immune cell infiltration. Recent research reported the role of DOK3 in tumor progression, with opposite effects in lung cancer and gliomas;however, its role in prostate cancer (PCa) remains elusive. This study aimed to explore the role of DOK3 in PCa and to determine the mechanisms involved. Methods: To investigate the functions and mechanisms of DOK3 in PCa, we performed bioinformatic and biofunctional analyses. Samples from patients with PCa were collected from West China Hospital, and 46 were selected for the final correlation analysis. A lentivirus-based short hairpin ribonucleic acid (shRNA) carrier was established for silencing DOK3. A series of experiments involving the cell counting kit-8, bromodeoxyuridine, and flow cytometry assays were performed to identify cell proliferation and apoptosis. Changes in biomarkers from the nuclear factor kappa B (NF-κB) signaling pathway were detected to verify the relationship between DOK3 and the NF-κB pathway. A subcutaneous xenograft mouse model was performed to examine phenotypes after knocking down DOK3 in vivo . Rescue experiments with DOK3 knockdown and NF-κB pathway activation were designed to verify regulating effects. Results: DOK3 was up-regulated in PCa cell lines and tissues. In addition, a high level of DOK3 was predictive of higher pathological stages and worse prognoses. Similar results were observed with PCa patient samples. After silencing DOK3 in PCa cell lines 22RV1 and PC3, cell proliferation was significantly inhibited while apoptosis was promoted. Gene set enrichment analysis revealed that DOK3 function was enriched in the NF-κB pathway. Mechanism experiments determined that knockdown of DOK3 suppressed activation of the NF-κB pathway, increased the expressions of B-cell lymphoma-2 like 11 (BIM) and B-cell lymphoma-2 associated X (BAX), and decreased the expression of phosphorylated-P65 and X-linked inhibitor of apoptosis (XIAP). As a common precursor for supercritical CO_(2)(scCO_(2))deposition techniques,solubility data of organometallic complexes in scCO_(2)is crucial for the preparation of nanocomposites.Recently,metal acetylacetonates have shown great potential for the preparation of single-atom catalytic materials.In this study,the solubilities of iron(Ⅲ)acetylacetonate(Fe(acac)3)and nickel(Ⅱ)acetylacetonate(Ni(acac)2)were measured at the temperature from 313.15 to 333.15 K and in the pressure range of 9.5–25.2 MPa to accumulate new solubility data.Solubility was measured using a static weight loss method.The semi-empirical models proposed by Chrastil and Sung et al.were used to correlate the solubility data of Fe(acac)3 and Ni(acac)2.The equations obtained can be used to predict the solubility of the same system in the experimental range.
