Temporal allocation of metabolic tolerance to transgenic Bt cotton in beet armyworm,Spodoptera exigua(Hübner)

Digestive and detoxification enzyme activity and nutrient composition were examined in the body of fourth instar beet armyworms,Spodoptera exigua(Hübner),fed on transgenic Bacillus thuringiensis(Bt) and non-Bt cotton for different time *** composition and specific enzyme activities differed significantly between the *** fed Bt ***-Bt *** 1,6 and 24 h,free fatty acid and glucose levels were significantly lower in *** fed on Bt cotton than those fed on non-Bt *** fed on Bt cotton had significantly higher trypsin and total superoxide dismutase(T-SOD) activities and significantly lower lipase,carboxylesterase and acetylcholinesterase activities than non-Bt fed worms for all feeding time *** were also observed among feeding times within each cotton variety *** lower free fatty acid and total amino acid were observed in *** fed on Bt cotton for 24 h than in those fed for 1 *** lower activities of lipase and trypsin were detected in *** fed on Bt cotton for 24 h than those for 1 and 4 ***,carboxylesterase and acetylcholinesterase activities in *** fed on Bt cotton for 24 h were significantly higher than those for 1,4 and 6 *** interaction between cotton variety and feeding time significantly affected the activities of lipase,trypsin,acetylcholinesterase and T-SOD enzymes in *** the temporal allocation of protection and detoxification enzyme activities in the body of *** in response to *** can provide a meaningful evaluation on the metabolic tolerance of herbivorous insects under the continuous selection pressure of a toxic protein.

Effects of shrub-grass patterns on soil detachment and hydraulic parameters of slope in the Pisha sandstone area of Inner Mongolia,China

The characteristics of soil holding capacity for different shrub-grass patterns are important to research the mechanisms regulating vegetation on *** objective of this study was to describe the characteristics and mecha-nisms of soil erosion and hydraulic parameters under differ-ent vegetation patterns in the Pisha sandstone area of Inner Mongolia on lands of 8°slope *** carried out field scouring experiments on five different shrub-grass patterns as treatments,viz no shrubs(GL),shrubs on the upper part of the slope(SU),middle part of the slope(SM)and lower part of the slope(SL).We designated bare slope(BL)as the *** employed three different water flow rates(15,20,30 L·min^(−1)).Our results showed that the contribution of plant root systems to slope sediment reduction ranged from 64 to 84%.The root systems proved to be the main contributing factor to reduction of erosion by *** relationship between soil detachment rate,stream flow power,and flow unit stream power under different scouring discharge rates showed that soil detachment declined in rank order as:BL>GL>SU>SM>*** SL pat-tern had the lowest soil detachment rate(0.098 g·m^(−2)·s^(−1)),flow stream power(2.371 W·m^(−2)),flow unit stream power(0.165 m·s^(−1))and flow shear stress(16.986 Pa),and proved to be the best erosion combating *** results of decision coefficient and path analysis showed that stream power was the most important hydraulic parameter for describing soil detachment *** combination of stream power and shear stress,namely Dr=0.1ω−0.03τ−0.56(R^(2)=0.924),most accurately simulated the soil detachment characteristics on *** study suggests that the risk of soil ero-sion can be reduced by planting shrub-grass mixes on these *** the conditions of limited water resources and economy,the benefit of sediment reduction can be maxi-mized by planting shrubbery on the lower parts of slopes.

Geochemical Characteristics of the Upper Triassic Xujiahe Formation in Sichuan Basin, China and its Significance for Hydrocarbon Accumulation

The alternative development of coal-bearing hydrocarbon source rocks and low-porosity and low-permeability tight sandstone reservoirs of the Triassic Xujiahe Formation in the Sichuan Basin is favorable for near-source hydrocarbon accumulation. The natural gas composition of the Xujiahe Formation in the Sichuan Basin is dominated by hydrocarbon gases, of which the methane content is80.16%-98.67%. Typically, the C2^＋ content is larger than 5% in main wet gas. The dry gas is mainly distributed in the western and northern regions of the basin. The non-hydrocarbon gases mainly contain nitrogen, carbon dioxide, hydrogen, and helium, with a total content of 〈2%. The carbon isotope ranges of methane and its homologues in natural gas are： δ^13C1 of-43.8‰ to-29.6‰, δ^13C2 of-35.4‰ to-21.5‰, δ^13C3 of-27.6‰ to-19.8‰,and δ^13C4 of-27.7‰ to-18.8‰. δ^13C3〉δ^13C4 occurs in some natural gas with a low evolution degree; such gas is mainly coal-related gas from humic-type source rocks of the Xujiahe Formation. As for the natural gas, δ^2 HCH4 values ranged from-195‰ to-161‰,δ^2 HC2H6 values ranged from-154‰ to-120‰, and δ^2 HC2H8 values ranged from-151‰ to-108‰. The dry coefficient,δ^13C and δ^2 HC2H4 are all positively correlated with the maturity of source rocks. The higher the maturity of source rocks is, the larger the natural gas dry coefficient is and the larger the δ^13C and δ^2 HC2H4 values are, indicative of the characteristic of near-source accumulation. The δ^2 HC2H6 value of natural gas is influenced by paleosalinity to a relatively large extent; the higher the paleosalinity is, the larger the δ^2 HC2H4 value is. The Pr/Ph value of the condensate oil ranged from 1.60 to 3.43, illustrating light oxidization-light reduction and partial-oxidization characteristics of the depositional environment of coal-bearing source rocks of the Xujiahe Formation. Simulation experiment on hydrocarbon generating potential of various source rocks on the northern margin of the Qaidam Basin,Qinghai,China

The northern margin of the Qaidam Basin is one of the main oil-gas exploration areas in western China,where source rocks are composed mostly of Middle and Lower Jurassic dark mud shale,carbargillite and coal. A large number of subsurface and outcrop samples differing in lithology with different types of organic matter were selected for resource evaluation,research and calculation. And among them,13 samples were used for simulation experiment on hydrocarbon generating potential of various source rocks. At first,two kinds of heating modes were compared through simulation experiment,including single temperature-step heating and continual heating. Perhaps,the process of hydrocarbon generation and expulsion occurred naturally between a close system and an open system. In addition,the first heating mode was convenient,and all its reaction products were involved in the whole thermal evolution,and the final simulation experimental results were basically in consistency with the natural evolution trend. Therefore,the first heating mode was adopted and the hydrocarbon yield of every sample was worked out. Accord-ing to the type and lithology of organic matter and the hydrocarbon yield of samples for simulation experiment,hy-drocarbon generation and expulsion mode with three kinds of lithology and five types of source rock has been estab-lished to provide the basis for hydrocarbon generation evaluation,research and resource calculation.

An inducible CRISPR-ON system for controllable gene activation in human pluripotent stem cells

Human pluripotent stem cells （hPSCs） are an important system to study early human development, model human diseases, and develop cell replacement therapies. However, genetic manipulation of hPSCs is challenging and a method to simultaneously activate multiple genomic sites in a controllable manner is sorely needed. Here, we constructed a *** system to efficiently upregulate endogenous genes in hPSCs. A doxycycline （Dox） inducible dCasg-VP64-p65-Rta （dCas9-VPR） transcription activator and a reverse Tet transactivator （rtTA） expression cassette were knocked into the two alleles of the AAVS1 locus to generate an iVPR hESC line. We showed that the dCas9-VPR level could be precisely and reversibly controlled by the addition and withdrawal of Dox. Upon transfection of multiplexed gRNA plasmid targeting the NANOG pro- moter and Dox induction, we were able to control NANOG gene expression from its endogenous locus. Interestingly, an elevated NANOG level promoted naive pluripotsnt gene expression, enhanced cell survival and clonogenicity, and enabled hESCs to integrate with the inner cell mass （ICM） of mouse blastocysts in vitro. Thus, iVPR cells provide a convenient platform for gene function studies as well as high-throughput screens in hPSCs.

Porcine reproductive and respiratory syndrome virus(PRRSV) continues to cause significant economic loss worldwide and remains a serious threat to the pork industry. Currently, vaccination strategies provide limited protection against PRRSV infection, and consequently, new antiviral strategies are urgently required. Andrographolide(Andro) and its derivative potassium dehydrographolide succinate(PDS) have been used clinically in China and other Asian countries as therapies for inflammation-related diseases, including bacterial and viral infections, for decades. Here, we demonstrate that Andro and PDS exhibit robust activity against PRRSV replication in Marc-145 cells and primary porcine alveolar macrophages(PAMs). The two compounds exhibited broad-spectrum inhibitory activities in vitro against clinically circulating type 2 PRRSV GD-HD, XH-GD, and NADC30-like HNhx strains in China. The EC_(50)values of Andro against three tested PRRSV strain infections in Marc-145 cells ranged from 11.7 to 15.3 lmol/L, with selectivity indexes ranging from 8.3 to10.8, while the EC_(50)values of PDS ranged from 57.1 to 85.4 lmol/L, with selectivity indexes ranging from 344 to ***, the anti-PRRSV activity of the two compounds is closely associated with their potent suppression on NFj B activation and enhanced oxidative stress induced by PRRSV infection. Further mechanistic investigations revealed that PDS, but not Andro, is able to directly interact with PRRSV particles. Taken together, our findings suggest that Andro and PDS are promising PRRSV inhibitors in vitro and deserves further in vivo studies in swine.
