Potentilla sericea is a heavy metal hyperaccumulator landscaping plant.MYB transcription factors play an important role in regulating plant stress response to adversity.However,there are few studies on MYB transcription factors in stress tolerance in Potentilla sericea.In this study,the PsMYB62 gene was successfully cloned from Potentilla sericea.Methods:Bioinformatic analysis and real-time quantitative PCR(qPCR)methods were used to evaluate this gene.The transgenic A.thaliana were obtained by flower dipping and the gene function was identified by determining physiological indicators under cadmium stress.Results:The open reading frame of PsMYB62 is 942 bp,which encodes 313 amino acids(aa)and belongs to the R2R3 MYB transcription factor.The plant overexpression vector PBI121-PsMYB62-GFP was constructed and successfully transferred into A.thaliana.The relative expression level of PsMYB62 was significantly increased by CdCl_(2),NaCl,ABA,and mannitol treatments.The germination rate of transgenic seeds was higher than those of wild type(WT)and empty vector(EV)under different concentrations of cadmium treatment.Upon treatment with 100μmol·L^(−1)of CdCl_(2)·2.5H_(2)O,the activities of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)in the transgenic plants were significantly higher than those in the WT and EV.The contents of H_(2)O_(2),O_(2)·−and malondialdehyde(MDA)in transgenic lines were increased,but lower than those in WT and EV.The expression levels of AtGSH,AtPCS,and AtNAS4 that were related to the regulation of cadmium were increased,but the expression levels of transgenic lines were higher than those of WT and EV.Conclusion:The above results showed that PsMYB62 could be induced by cadmium and could improve the cadmium resistance of plants.

Stripe (yellow) rust caused by Puccinia striiformis f. sp. tritici occurs in almost allwheat-producing regions of the world. Severe countrywide epidemics in Chinahave caused substantial yield losses. Growing resistant cultivars is the beststrategy to control this disease but the pathogen can overcome resistance inwheat cultivars. The high variation in the virulence of the pathogen combinedwith the large areas of susceptible wheat cultivars enables the pathogenpopulation to increase rapidly and disperse over long distances under favorableenvironmental conditions, resulting in severe pandemics within croppingseasons. Current stripe rust control measures are based on many years ofresearch including the underlying epidemiology regarding year-to-year survivalof the pathogen, pathways of pathogen dispersal within seasons and years, therole of P. striiformis sexual hybridization, the use of resistance sources inbreeding programs, and year-round surveillance of national wheat crops that arepresent in different parts of the country throughout the year. All these strategiesdepend on accurate prediction of epidemics, more precise use of fungicides tomeet national requirements and better deployment of resistance genes. Newideas with potential application in sustainable protection of stripe rust includenegative regulatory gene editing, resistance gene overexpression and biologicalcontrol based on microbiomes.

Desmoid-type fibromatosis (DF) is a rare monoclonal fibroblastic proliferation that is characterized by locally infiltrative but rarely metastatic lesions. Tyrosine kinase and γ-secretase inhibitors are primarily used in the targeted therapy of DF. The use of these drugs, however, is mainly based on the recommendations of retrospective studies with small sample sizes. Previous studies that focused on the mechanism, efficacy, and safety of targeted therapy for DF were reviewed to provide references for clinical applications and research. The efficacy and safety of targeted therapy were compared with those of other systemic therapy options. Targeted therapy does not provide considerable advantages in efficacy and safety over other medical treatments and is usually applied after the failure of antihormonal therapies, nonsteroidal anti-inflammatory drugs, and chemotherapy. Further studies are required to explore the mechanism, indications, and appropriate drug dosage of the targeted therapy of DF.

The development and deployment of diverse resistance sources in new wheat cultivars underpin the durable control of stripe rust.In the present study,two loci for adult plant resistance(APR),QYr SM155.1 and QYr SM155.2,were identified in the Chinese wheat breeding line Shaanmai 155.QYr SM155.1 was mapped to a 3.0-c M interval between the single-nucleotide polymorphism(SNP)markers AX-109583610 and AX-110907562 on chromosome arm 2 BL.QYr SM155.2 was mapped to a 2.1-c M interval flanked by the SNP markers AX-110378556 and AX-86173526 on chromosome arm 7 AS.A genome-wide association study was used to identify markers associated with APR in a panel of 411 spring wheat lines.Thirteen and 11 SNPs were significantly associated with QYr SM155.1 and QYr SM155.2,respectively,corresponding to physical intervals of 653.75–655.52 Mb on 2 BL and 81.63–83.93 Mb on7 AS.To characterize the haplotype variation and the distribution of these QTL,haplotype analysis was performed based on these SNPs in an independent panel of 1101 worldwide wheat accessions.Three major haplotypes(2 B_h1,2 B_h2,and 2 B_h3)for QYr SM155.1 and four major haplotypes(7 A_h1,7 A_h2,7 A_h3,and 7 A_h4)for QYr SM155.2 were identified.Accessions individually harboring QYr SM155.1_h1 and QYr SM155.2_h1 haplotypes and their combination displayed resistance.Additional assays of 1306 current Chinese cultivars and breeding lines using markers flanking QYr SM155.1 and QYr SM155.2 indicated that the resistance haplotypes of the two QTL were present in respectively 1.45%and 14.16%of lines.Increasing resistance haplotype frequencies at these two loci using marker-assisted selection should benefit wheat production in China.

Size distribution and chemical characteristics of particles from crop residue open burning in North China

Crop residue open burning is an important emission source of ambient particles in China.This study analyzed the particle emission characteristics of crop residue open burning through combustion experiments with a novel open combustion simulation device using three typical crop straws in north China(corn,wheat,and rice).Particle samples size ranging from 0.006–9.890μm were collected by an Electrical Low Pressure Impactor plus,a high size-resolution instrument capable of dividing particles into 14 size stages.The size distributions of organic carbon(OC),elemental carbon(EC),water-soluble ions,and elements were analyzed,and source chemical profiles were constructed for PM0.1,PM1,PM2.5,and PM10.The number concentration of particles was concentrated in the Aiken nuclei mode(0.006–0.054μm),accounting for 75%of the total number,whereas the mass concentration was concentrated in the accumulation mode(0.054–0.949μm),accounting for 85.43%of the mass loading.OC,EC,Cl−,and K(include total K and water-soluble K)were the major chemical components of the particles,whose mass percentage distributions differed from those of other components.These fivemain components exhibited a bell-shaped size distribution in the 0.006–9.890μm range,whereas the other components exhibited a U-shaped distribution.Among the chemical profiles for PM0.1–PM10,OC was the most important component at 10–30%,followed by EC at 2%–8%.The proportions of K^(+),Cl^(−),and K varied substantially in different experimental groups,ranging from 0–15%,and K+and Cl−were significantly correlated(r=0.878,α=0.000).

To examine the features of heavy metal pollution of PM2.5 （particulate matter less than 2.5 μm） in Tianjin, China, as well as the exposure risk of PM2.5 to human health, we analyzed ambient PM2.5samples collected from a campus of Nankai University in June, August, and October 2012. The concentrations of PM2.5 and heavy metals （Ni, Cu, Pb, Zn, Cr, Cd, Hg, As and Mn） in PM2.5 were analyzed by gravimetric analysis and inductively coupled plasma-mass spectrometry, respectively. The results show that the heavy metals contained in PM2.5 were, in descending order, Cu, Zn, Pb, Mn, Cr, Ni, Cd, As, and Hg. The proportion of Cd exceeded the secondary level of National Ambient Air Quality Standard of China （GB 3095-2012） by 1.3 times, while others were within the limit. Enrichment factor analysis indicated that Cu, Zn, Cd, Pb, and Hg are mainly from anthropogenic sources. Principal component analysis indicated that the main sources of the heavy metals are vehicle exhaust, chemical waste, and coal-burning activities. The nine heavy metals which may cause health issues by exposure through the human respiratory system and should be further examined are Cr, Cd, As, Ni, Cu, Pb, Mn, Zn, and Hg, in the order of decreasing risk levels. With reference to the U.S. EPA standard the risk levels of all nine metals were below the acceptable level （10 6/year）.

To investigate the levels of exposure to particulate-bound polycyclic aromatic hydrocarbon （PAH） and to estimate the risk these levels pose to traffic assistants （TAs） in Tianjin （a rnegacity in North China）, a measurement campaign （33 all-day exposure samples, 25 occupational-exposure samples and 10 indoor samples） was conducted to characterize the TAs＇ exposure to PAHs, assess the cancer risk and identify the potential sources of exposure. The average total exposure concentration of 14 PAHs was approximately 2871 ＋ 928 ng/rn3 （on-duty）, and 1622 ＋ 457 ng/m3 （all-day）. The indoor PAHs level was 1257 ＋ 107 ng/m3. After 8000 Monte Carlo simulations, the cancer risk resulting from exposure to PAHs was found to be approximately 1.05 x 10-4. A multivariate analysis was applied to identify the potential sources, and the results showed that, in addition to vehicle exhaust, coal combustion and cooking fumes were also another two important contributors to personal PAH exposure. The diagnostic ratios of PAH compounds agree with the source apportionment results derived from principal component analysis.

钢-ECC组合梁高温冷却后受剪性能研究

为研究受热温度、冷却方式对钢-高延性水泥基材料(ECC)组合梁高温后受剪性能的影响,通过4根高温后钢-ECC组合梁和1根常温对比组合梁的静力加载试验,分析了钢-ECC组合梁在高温后的裂缝扩展规律、破坏模式、剩余承载力及变形。试验结果表明:高温作用后,钢-ECC组合梁裂缝呈现多缝开裂、密集分布的特征,随温度升高,裂缝数量增多、宽度增大,湿水冷却后裂缝深度加深;钢-ECC组合梁在常温下及高温后的破坏形态均为剪切破坏,经历200℃高温后钢-ECC组合梁仍具有较好的延性,而当温度高于400℃时,采用湿水冷却后延性下降显著;高温后钢-ECC组合梁承载力降低,200℃内冷却方式对承载力影响较小,400℃时湿水冷却后承载力显著小于自然冷却的,自然冷却和湿水冷却后承载力分别降低8.7%、25.7%;冷却方式对钢-ECC组合梁初始抗剪刚度影响显著,湿水冷却后钢-ECC组合梁初始抗剪刚度提高,而自然冷却后降低。基于ABAQUS软件中瞬态热传导分析模块模拟钢-ECC组合梁的温度场,并采用塑性损伤模型对钢-ECC组合梁进行热-力耦合全过程分析,建立了有限元模型,经比较,模型的预测结果与试验得到的荷载-位移曲线吻合良好。进一步利用有限元模型开展参数分析,发现与自然冷却相比,钢-ECC组合梁湿水冷却后受剪承载力随翼板厚度及强度增加而增长的幅度较小。
