Repurposing lansoprazole to alleviate metabolic syndrome via PHOSPHO1 inhibition

Drug repurposing offers an efficient approach to therapeutic *** this study,our bioinformatic analysis first predicted an association between obesity and lansoprazole(LPZ),a commonly prescribed drug for gastrointestinal *** went on to show that LPZ treatment increased energy expenditure and alleviated the high-fat diet-induced obesity,insulin resistance,and hepatic stea-tosis in *** with LPZ elicited thermogenic gene expression and mitochondrial respiration in primary adipocytes,and induced cold tolerance in cold-exposed mice,suggesting the activity of LPZ in promoting adipose thermogenesis and energy ***,LPZ is an efficient inhibitor of adipose phosphocholine phosphatase 1(PHOSPHOI)and produces metabolic benefits in a PHOS-PHO1-dependent *** results suggested that LPZ may stimulate adipose thermogenesis by inhi-biting the conversion of 2-arachidonoylglycerol-lysophosphatidic acid(2-AG-LPA)to 2-arachidonoylglycerol(2-AG)and reduce the activity of the thermogenic-suppressive cannabinoid recep-tor *** summary,we have uncovered a novel therapeutic indication and mechanism of LPZ in managing obesity and its related metabolic syndrome,and identified a potential metabolic basis by which LPZ improves energy metabolism.

Prophylactic effects of Edible Lilium-Egg yolk compound powder on depression-like behaviors in rats exposed to chronic unpredictable mild stress: A role of gut-brain axis

The incidence rate and harmfulness of depression are increasing yearly， and the research on the antidepressant effect of regulating the intestinal microecology balance has attracted widespread attention. The purpose of this research was to study the preventive effects and the regulatory mechanism of edible Lilium-egg yolk compound powder(Chinese abbreviation， BHT) on gut-brain crosstalk in depressive rats induced by chronic unpredictable mild stress(CUMS) through the "microbiota-gut-brain" axis. Results showed that infusion of BHT for 21 days ameliorated the morphology of the hippocampus and colon， increased the rate of sucrose preference， reduced the immobility time of forced swimming， increased the expression level of anti-inflammatory cytokine Interleukin-10 (IL-10) in serum， up-regulate the expression level of plasma neurotransmitter 5-hydroxy tryptamine (5-HT)， down-regulate the expression of inflammatory factors tumor necrosis factor-α (TNF-α)， interleukin-6 (IL-6)， and interleukin-1β (IL-1β) and neurotransmitter glutamic acid (Glu) in serum， and decrease the expression levels of lipopolysaccharide (LPS)， diamine oxidase (DAO) and lactate dehydrogenase (LD) in plasma. The underlying mechanism may be to reduce the activation of microglia (lower levels of CD11b， IBa1 protein and gene expression)， regulate hippocampal GR/BDNF/NPY signal pathway (higher levels of GR， BDNF， NPY protein and gene expression)， enhance intestinal mucosal barrier function (higher levels of ZO-1， Occludin protein and gene expression)， regulate the composition and structure of intestinal flora(Analysis results of LEfSe combined with random forest showed that Prevotella and Paraprevotella might be biomarkers of BHT intervention in depression)， and it is related to the improvement of glutathione metabolism， and other metabolic pathways. Dynamics of information diffusion and disease transmission in time-varying multiplex networks with asymmetric activity levels

While the interaction between information and disease in static networks has been extensively investigated,many studies have ignored the characteristics of network *** this study,we construct a new two-layer coupling model to explore the interactions between information and *** upper layer describes the diffusion of disease-related information,and the lower layer represents the disease *** then use power-law distributions to examine the influence of asymmetric activity levels on dynamic propagation,revealing a mapping relationship characterizing the interconnected propagation of information and diseases among partial nodes within the ***,we derive the disease outbreak threshold by using the microscopic Markov-chain approach(MMCA).Finally,we perform extensive Monte Carlo(MC)numerical simulations to verify the accuracy of our theoretical *** findings indicate that the activity levels of individuals in the disease transmission layer have a more significant influence on disease transmission compared with the individual activity levels in the information diffusion ***,reducing the damping factor can delay disease outbreaks and suppress disease transmission,while improving individual quarantine measures can contribute positively to disease *** study provides valuable insights into policymakers for developing outbreak prevention and control strategies.

In this paper,we consider the semilinear elliptic equation systems{△u+u=αQ_(n)(x)|u|^(α-2)|v|^(β)u in R^(N),-△v+v=βQ(x)|u|^(α)|v|^(β-2)v in R^(N),where N≥3,α,β>1,α+β0} shrink to a set with finitely many points as n→∞.Motivated by the work of Fang and Wang[13],we use variational methods to study the limiting profile of ground state solutions which are concentrated at one point of the set with finitely many points,and we build the localized concentrated bound state solutions for the above equation systems.

Distribution characteristics and removal rate of antibiotics and antibiotic resistance genes in different treatment processes of two drinking water plants

Emerging pollutants,such as antibiotics and antibiotic-resistance genes,are becoming increasingly important sources of safety and health *** water safety,which is closely related to human health,should receive more attention than natural water body ***,minimal research has been performed on the efficacy of existing treatment processes in water treatment plants for the removal of antibiotics and antibiotic resistance *** address this research gap,this study detected and analyzed six main antibiotics and nine antibiotic resistance genes in the treatment processes of two drinking water plants in *** were collected over three months and then detected and analyzed using ultra-high-performance liquid chromatography-tandem mass spectrometry and fluorescence *** total concentrations of antibiotics and antibiotic resistance genes in the influent water of the two water plants were characterized as December>March>*** precipitation and filtration processes of the Zou Maling Water Plant and Yu Shidun Water Plant successfully removed the *** ozone-activated carbon process increased the removal rate of most antibiotics to 100%.However,a large amount of antibiotic resistance gene residues remained in the effluents of the two water *** experiments demonstrated that the existing ozone-activated carbon processes could not effectively remove antibiotic resistance *** study provides a reference for the optimization of drinking water treatment processes for antibiotics and antibiotic resistance gene removal.

Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and ***,its tumor-suppressive mechanisms remain largely ***,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its *** expression and its association with clinicopathologic characteristics of PDA patients were analyzed using *** mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot *** biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal *** expression was reduced in human PDA cell lines and *** UTX expression was associated with poor differentiation and prognosis in *** UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the ***,UTX expression was trans-activated by GATA6 ***6-mediated PDA progression could be blocked,at least partially,by silencing UTX *** conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and ***,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

在“逆全球化”因素扰动下,中国面临的全球供应链风险加剧,如何更好维护中国制造业全球供应链安全成为重要议题。文章构建了2003—2021年中国18个制造业细分行业的全球供应链风险指数,并基于中间品贸易的视角探究中国制造业OFDI对其全球供应链风险的影响。研究发现:OFDI在畅通贸易渠道、促进母国产业技术升级的同时,降低了中国中间品进出口的对外依赖程度,提升了对国际市场依赖的多元化程度,进而有助于降低中国制造业的全球供应链风险。其中,中低技术制造业、低流动性制造业以及OFDI流向欠发达地区对防范全球供应链风险的作用更明显。

Modulation of host N6-methyladenosine modification by gut microbiota in colorectal cancer

As a research hotspot in the field of molecular biology,N6-methyladenosine(m6A)modification has made progress in the treatment of colorectal cancer(CRC),leukemia and other *** studies have demonstrated that the tumour microenvironment(TME)regulates the level of m6A modification in the host and activates a series of complex epigenetic signalling pathways through interactions with CRC cells,thus affecting the progression and prognosis of ***,with the diversity in the composition of TME factors,this action is reci-procal and ***,some studies have experimentally revealed that the intestinal flora can alter CRC cell proliferation by directly acting on m6A and thereby altering CRC cell *** review summarizes the data,supporting the idea that the intestinal flora can influence host m6A levels through pathways such as methyl donor metabolism and thus affect the progression of *** also review the role of m6A modification in the diagnosis,treatment,and prognostic assessment of CRC and discuss the current status,limitations,and potential clinical value of m6A modification in this *** propose that additional in-depth research on m6A alterations in CRC patients and their TME-related targeted therapeutic issues will lead to better therapeutic outcomes for CRC patients.
