Success rate of current human-derived gastric cancer organoids establishment and influencing factors:A systematic review and meta-analysis

BACKGROUND Human-derived gastric cancer organoids(GCOs)are widely used in gastric cancer research;however,the culture success rate is generally *** To explore the potential influencing factors,and the literature on successful culture rates of GCOs was reviewed using *** PubMed,Web of Science,and EMBASE were searched for *** trained researchers selected the studies and extracted *** 17.0 software was used for meta-analysis of the incidence of each outcome *** adjusted Methodological Index for Non-Randomized Studies scale was used to assess the quality of the included *** plots and Egger’s test were used to detect publication *** analyses were conducted for sex,tissue source,histo-logical classification,and the pathological tumor-node-metastasis(pTNM)cancer staging *** Eight studies with a pooled success rate of 66.6%were *** derived from women and men had success rates of 67%and 46.7%,*** from surgery or biopsy/endoscopic submucosal dissection showed success rates of 70.9%and 53.7%,*** of poorly-differentiated,moderately-differentiated and signet-ring cell cancer showed success rates of 64.6%,31%,and 32.7%,*** with pTNM stages I-II and III-IV showed success rates of 38.3%and 65.2%,respectively.Y-27632 and non-Y-27632 use showed success rates of 58.2%and 70%,*** generated with collagenase were more successful than those constructed with Liberase TH and TrypLE(72.1%vs 71%,respectively).EDTA digestion showed a 50%lower success rate than other methods(P=0.04).CONCLUSION GCO establishment rate is low and varies by sex,tissue source,histological type,and pTNM *** Y-27632,and using Liberase TH,TrypLE,or collagenase yields greater success than EDTA.

Fertility-preserving treatment and pregnancy outcomes in the early stage of endometrial carcinoma

Objective This study aimed to review the available literature on fertility-preserving treatment and pregnancy outcomes in patients with early-stage endometrial carcinoma who desired to preserve their fertility. Data sources The PubMed database （1992-2012） was searched for the words ＂conservative ＂OR＂ fertility sparing ＂OR＂ fertility preserving＂ AND ＂endometrial neoplasms＂（MESH）. All relevant articles in English and the relevant references were collected. Study selection Data from published articles about fertility-preserving treatment of endometrial cancer, including the response and recurrence rate of conservative treatment, strategies of infertility treatment, pregnancy, and obstetric outcomes, were selected. Data were mainly extracted from 41 studies, which are listed in the reference section of this review. Results Hormone therapy was the most common method used for early-stage endometrial carcinoma in patients who wished to preserve fertility. Sixty percent of the patients became pregnant after remission of the carcinoma. The percentage of patients who conceived in the assisted reproductive technology group was higher than that of the natural pregnancy group （80.0% vs. 43.2%, P〈0.01）. A higher rate of preterm labor and multiple pregnancies was observed in the assisted reproductive technology group than that in the natural pregnancy group. The majority of pregnancies （71.4%） in the assisted reproductive technology group were achieved by in vitro fertilization-embryo transfer. The clinical pregnancy rate of transfer cycles in patients with endometrial carcinoma was 34.1%. Conclusions Assisted reproductive technology is a good option in well-selected patients with early-stage endometrial carcinoma who have completed conservative treatment. In vitro fertilization-embryo transfer offers an opportunity to achieve an immediate pregnancy.

Development of a new instrument for measurement of high temperature mechanical properties of resin-bonded sand

The mechanical properties of resin-bonded sand mixtures at high temperatures significantly affect the quality of casting. However, the existing instruments for high-temperature performances testing mainly focus on inorganic binder-bonded sands no matter the test items or the atmospheric protection, while the instrumentss specially designed for resin-bonded sand are not yet available. A new instrument for testing the hightemperature performance of resin sand was designed including the confirmation of the testing parameters, loading, measurement and control systems, and the design of the frame shape and heating furnace. This instrument can test the compressive strength, heat tolerance time and restraining load of phenol-formaldehyde resin coated sand, self-hardened furan resin sand, and trimethylamine(TEA)-based resin bonded sand at high temperatures. The developed instrument has a high accuracy offering smaller than 0.3% deviation at a full scale in the measurement of the high temperature compressive strength and the restraining load over the range of 0-6.8 MPa and 0-2,000 N, respectively. The high temperature heat tolerance time range is 0-300 s and its measurement accuracy is ±1 s.

An overview and perspective of identifying lunar craters

The spatiotemporal evolution of lunar impact craters is crucial for investigating lunar interior structure,internal and external dynamic processes,and interplanetary impact *** in lunar crater identification are reviewed based on topography and gravity data,and the features and mechanisms of topographic or buried craters are analyzed regarding morphology,gravity anomalies,gravity gradients,and the underlying crust-mantle interface *** on the compiled crater catalog,the early lunar impact flux and thermal evolution are further discussed according to the basin ages and their interior *** some ancient impact basins,the crater size-frequency distribution measurements revealed age discrepancies from previous studies,suggesting that the lunar late heavy bombardment event started at~3.95 *** degraded bulge structures of the crust-mantle interface beneath mare basins reveal that these basins formed on the lunar crust surface under high-temperature conditions and underwent prolonged relaxation compared to highland ***,we reveal that the up-to-date identification of lunar buried craters remains inaccurate and incomplete,preventing us from accurately reconstructing lunar and interplanetary impact ***,we propose that a gravity model constructed using localized orthogonal basis functions can be useful for identifying lunar craters.

Optimized the vanadium electrolyte with sulfate-phosphoric mixed acids to enhance the stable operation at high-temperature

Herein, the influence of the concentration design and comprehensive performance of the sulfate-phosphoric mixed acid system electrolyte is investigated to realize an electrolyte that maintains high energy density and stable operation at high temperatures. Static stability tests have shown that VOPO4 precipitation occurs only with vanadium(V) electrolyte. The concentration of vanadium ion of 2.0–2.2 mol·L^(–1), phosphoric acid of 0.10–0.15 mol·L^(–1), and sulfuric acid of 2.5–3.0 mol·L^(–1) are suitable for a vanadium redox flow battery in the temperature range from –20 to 50 ℃. The equations for predicting the viscosity and conductivity of electrolytes are obtained by the response surface method. The optimized electrolyte overcomes precipitation generation. It has 2.8 times higher energy density than the non-phosphate electrolyte, and a coulomb efficiency of 94.0% at 50 ℃. The sulfate-phosphoric mixed acid system electrolyte promotes the electrode reaction process, increases the current density, and reduces the resistance. This work systematically optimizes the concentrations of composition of positive and negative vanadium electrolytes with mixed sulfate-phosphoric acid. It provides a basis for the different valence states and comprehensive properties of sulfate-phosphoric mixed acid system vanadium electrolytes under extreme environments, guiding engineering applications.

Cisplatin-based miRNA delivery strategy inspired by the circCPNE1/miR-330-3p pathway for oral squamous cell carcinoma

Circular RNAs(circRNAs)are ideal biomarkers of oral squamous cell carcinoma(OSCC)because of their highly stable closed-loop structure,and they can act as microRNA(miRNA)sponges to regulate OSCC *** analyzing clinical samples,we identified circCPNE1,a dysregulated circRNA in OSCC,and its expression level was negatively correlated with the clinical stage of OSCC ***-of-function assays revealed the tumor-suppressive effect of circCPNE1,which was then identified as a miR-330-3p ***-330-3p was recognized as a tumor promoter in multiple studies,consistent with our finding that it could promote the proliferation,migration,and invasion of OSCC *** results indicated that selective inhibition of miR-330-3p could be an effective strategy to inhibit OSCC ***,we designed cationic polylysine-cisplatin prodrugs to deliver antagomiR-330-3p(a miRNA inhibitory analog)via electrostatic interactions to form PP@miR nanoparticles(NPs).Paratumoral administration results revealed that PP@miR NPs effectively inhibited subcutaneous tumor progression and achieved partial tumor elimination(2/5),which confirmed the critical role of miR-330-3p in OSCC *** findings provide a new perspective for the development of OSCC treatments.
