Prediction of gastric cancer risk by a polygenic risk score of Helicobacter pylori

BACKGROUND Genetic variants of Helicobacter pylori(H. pylori) are involved in gastric cancer occurrence. Single nucleotide polymorphisms(SNPs) of H. pylori that are associated with gastric cancer have been reported. The combined effect of H. pylori SNPs on the risk of gastric cancer remains *** To assess the performance of a polygenic risk score(PRS) based on H. pylori SNPs in predicting the risk of gastric *** A total of 15 gastric cancer-associated H. pylori SNPs were selected. The associations between these SNPs and gastric cancer were further validated in 1022 global strains with publicly available genome sequences. The PRS model was established based on the validated SNPs. The performance of the PRS for predicting the risk of gastric cancer was assessed in global strains using quintiles and random forest(RF) methods. The results of RF analyses indicated that the area under the curve(AUC) value for classifying gastric cancer and non-gastric cancer was 0.75, suggesting that the PRS based on H. pylori SNPs was capable of predicting the risk of gastric cancer. Reference genes for quantitative real-time PCR analysis and quantitative expression of P5CS in Agropyron mongolicum under drought stress

Reference genes, stably expressing in different tissues and cells, are commonly used as the references in expression analysis. Selecting the optimum reference gene is crucial to the success of experiments. In this study, the expression stabilities of nine common reference genes, including ACT2, 18 S r RNA, APRT, EF-1α, RNA POL II, TUBα, TUBβ, GAPDH and TLF of Agropyron mongolicum, were studied under drought condition. Among them, 18 S r RNA was found to be the most optimum reference gene under drought stress by the analyzing of ge Norm and Norm Finder software. quantitative expression levels of P5 CS using 18 S r RNA as the reference gene, and proline contents under drought stress in A. mongolicum were further operated, and we found the expression level of P5 CS gene and proline content had a significantly positive relationship（R^2=0.7763, P〈0.05）. This study established and validated 18 S r RNA as the reference genes in A. mongolicum under drought stress, providing a powerful tool for the quantitative expression analysis of drought genes in A. mongolicum.

Visual Detection of Vibrio parahaemolyticus using Combined CRISPR/Cas12a and Recombinase Polymerase Amplification

Objective To establish an ultra-sensitive,ultra-fast,visible detection method for Vibrio parahaemolyticus(VP).Methods We established a new method for detecting the tdh and trh genes of VP using clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 12a(CRISPR/Cas12a)combined with recombinase polymerase amplification and visual detection(CRISPR/Cas12a-VD).Results CRISPR/Cas12a-VD accurately detected target DNA at concentrations as low as 10^(-18)M(single molecule detection)within 30 min without cross-reactivity against other *** detecting pure cultures of VP,the consistency of results reached 100%compared with real-time *** method accurately analysed pure cultures and spiked shrimp samples at concentrations as low as 10^(2)CFU/*** The novel CRISPR/Cas12a-VD method for detecting VP performed better than traditional detection methods,such as real-time PCR,and has great potential for preventing the spread of pathogens.

Influences of soil moisture and salt content on loess shear strength in the Xining Basin, northeastern qinghai-Tibet Plateau

Moisture and salt content of soil are the two predominant factors influencing its shear strength. This study aims to investigate the effects of these two factors on shear strength behavior of loess in the Xining Basin of Northeast qinghai-Tibet Plateau, where such geological hazards as soil erosion, landslides collapse and debris flows are widespread due to the highly erodible loess. Salinized loess soil collected from the test site was desalinized through salt-leaching in the laboratory. The desalinized and oven-dried loess samples were also artificially moisturized and salinized in order to examine how soil salinity affects its shear strength at different moisture levels. Soil samples prepared in different ways(moisturizing, salt-leaching, and salinized) were measured to determine soil cohesion and internal friction angle. The results show that salt-leaching up to 18 rounds almost completely removed the salt content and considerably changed the physical components of loess, but the soil type remained unchanged. As salt content increases from 0.00% to 12.00%, both the cohesion and internal friction angle exhibit an initial decrease and then increase with salt content. As moisture content is 12.00%, the salt content threshold value for both cohesion and internal friction angle is identified as 3.00%. As the moisture content rises to 16.0% and 20.00%, the salt content threshold value for cohesion is still 6.00%, but 3.00% for internal friction angle. At these thresholds soil shear strength is the lowest, below which it is inversely related to soil salinity. Beyond the thresholds, however, the relationship is positive. Dissimilar to salinity, soil moisture content exerts an adverse effect on shear strength of loess. The findings of this study can provide a valuable guidance on stabilizing the engineering properties of salinized loess to prevent slope failures during heavy rainfall events.

Using Stroh＇s formalism the simple explicit expressions of Green＇s functions for a 2D piezoelectric body with two semi-infinite fixed conductor cracks subjected to a generalized line force were given. The Coulomb force acting on the free line charge aroused by the piezoelectricity and the distributed boundary polarization charges was discussed at first. Interactions between two singularities with free charge （s） located in a region with external cracks were studied, too. The numerical results show that the Coulomb force for two or more singularities with one free charge at least will have much influence on the electromechanical fields in piezoelectric media when these singularities move closely with each other and therefore cannot be overlooked again. The solutions obtained are valid not only for plane and anti-plane problems but also for coupled problems between in-plane and out-of-plane deformations.

A new organocatalytic double annulation cascade involving scission/recombination of N-O bonds of nitrones is reported for the first time, and used to produce a range of hitherto unprecedented tricyclic bridged-fused benzo[d]azepines bearing three stereogenic centers with moderate to good yields and complete diastereoselectivity. A quinine-catalyzed reaction of yne–allenone esters with nitrones worked well and provided a convergent and regioselective pathway to access these three-dimensional scaffolds from the planar conjugated system. Density functional theory(DFT) calculations have been applied to understand the key process for forming diradical intermediates.

Structural features and thermoelectric performance of Sb- and Bi-doped Cu_(2)SnSe_(3) compounds

In this paper, a series of Sb-doped and Bi-doped Cu_(2)Sn_(1-x)M_(x)Se_(3) samples(M = Sb, Bi) are prepared by vacuum melting combined with the spark plasma sintering process. The effects of different atomic doping amounts on their properties are discussed. Structural studies indicate that all obtained samples comprise a single Cu_(2)SnSe_(3) phase. Sb and Bi atoms are experimentally demonstrated to be efficient cation dopants for increasing the transport performance. Compared with that doping on the cation site,Bi doping is much more efficient in increasing the electron concentration of the Cu_(2)SnSe_(3) system. Ultimately, a high figure of merit of 0.36 is achieved in the Cu_(2)Sn_(0.94)Sb_(0.06) Se_(3) sample at 773 K due to the enhanced power factor and lowered lattice thermal conductivity,which are 1.73 times higher than those of the pure *** results provide an efficient approach to enhance thermoelectric performance via other doping atoms, which could also be applied to copper-based chalcogenide materials.
