Helicobacter pylori and 17β-estradiol induce human intrahepatic biliary epithelial cell abnormal proliferation and oxidative DNA damage

BACKGROUND: Biliary cancers are more common in females, and previous studies have suggested that Helicobacter pylori (H. pylori) exists in the biliary system. However, the effects of H. pylori infection and estrogen on the biological behaviors of human biliary epithelium mucosa remain unknown. The present study aimed to clarify their effects on the proliferation, apoptosis, migration and oxidative DNA damage of a human intrahepatic biliary epithelial cell (HIBEC) line in vitro. METHODS: HIBECs were co -cultured with 17 beta-estradiol (at 10(-9) mol/L, 10(-7) mol/L, and 10(-5) mol/L) and H. pylori (at MOI=0.5:1, 1:1, and 2:1) and continuously passaged until the 15th generation (approximately 45 days). Then, the following assays were performed. HIBEC proliferation was measured using the CCK-8 assay, plate clone-formation assay and by determining Ki-67 expression with immunocytochemistry; cell apoptosis and migration were investigated using Annexin-V/PI and transwell assays, respectively; and reactive oxygen species (ROS) and 8-hydroxy-2'-deoxyguanosine (8-OHdG) production were detected by flow cytometry and immunofluorescence staining combined with confocal laser scanning microscopy, respectively. The results were the basis for evaluating the level of oxidative stress and the related DNA damage in HIBECs. RESULTS: HIBECs maintained a normal morphology and vitality when treated with 17 beta-estradiol (at 10(-9) mol/L) and H. pylori (at MOI=0.5:1 and 1:1). 17 beta-estradiol at 10(-7) mol/L and 10(-5) mol/L and H. pylori at MOI=2:1, by contrast, caused cell death. Compared with controls, HIBECs treated with 17 beta-estradiol (10(-9) mol/L) and H. pylori (MOI=1:1) had a higher up-regulation of proliferation, Ki-67 expression, clone formation, migration activity and the expression of ROS and 8-OHdG and exhibited a down-regulation of apoptosis. The above effects were further increased when 17 beta-estradiol and H. pylori were combined (P<0.05). CONCLUSIONS: H. pylori and 17

An efficient method for the synthesis of novel benzo[b]pyrido[3＇,2＇：4,5]thieno[2,3-e][l,6]naphthyridine-8-one derivatives has been developed using a Pictet-Spengler reaction between 4-（3-aminothieno[2,3-b]pyridin-2-yl）quinoline-2-ones,which could be obtained from the alkylation of4-bromomethylquinoline-2-ones with 3-cyanopyridine-2-thione followed by a Thorpe-Ziegler isomerization,and aromatic aldehydes under p-TsOH as catalysis in good yields.

An efficient method for the synthesis of novel benzo[b]pyrimido[4′,5′：5,4]thieno[2,3e]-[1,6]naphthyridine-8-one derivatives via Pictet-Spengler cyclization is reported. The reaction of 4-（3-aminopyrimido[4,5-d]thieno-2-yl）quinoline-2-ones, which could be obtained from Thorpe-Ziegler isomerization of 4-bromomethylquinoline-2-ones and 5-cyano-1,6-dihydro-4-methyl-2-phenyl-6-thioxopyrimidine,with aromatic aldehydes in the presence of BF3·OEt2 gives pyrimidothieno[1,6]naphthyridines in good yields.

Based on a comprehensive analysis of the mineral composition of major lunarrocks (highland anorthosite, lunar mare basalt and KREEP rock), we investigate the reflectancespectral characteristics of the lunar rock-forming minerals, including feldspar, pyroxene andolivine. The affecting factors, the variation of the intensity of solar radiation with wavelengthand the reflectance spectra of the lunar rocks are studied. We also calculate the reflectivity oflunar mare basalt and highland anorthosite at 300 nm, 415 nm, 750 nm, 900 nm, 950 nm and 1000 nm. Itis considered that the difference in composition between lunar mare basalt and highland anorthositeis so large that separate analyses are needed in the study of the reflectivity of lunar surfacematerials in the two regions covered by mare basalt and highland anorthosite, and especially in theregion with high Th contents, which may be the KREEP-distributed region.

This paper addresses a distributed real-time optimal power flow(RTOPF) strategy for DC microgrids. In this paper, we focus on the scenarios where local information sharing is conducted in stochastic communication networks subject to random failures. Most existing real-time optimal power flow(OPF) algorithms for the DC microgrid require all controllers to work in concert with a fixed network topology to maintain a zero gap between estimated global constraint violations. Thus, the high reliability of communication is required to ensure their convergence. To address this issue, the proposed RTOPF strategy tolerates stochastic communication failures and can seek the optimum with a constant step size considering the operation limitations of the microgrid. These aspects make the strategy suitable for real-time optimization, particularly when the communication is not reliable. In addition, this strategy does not require information from non-dispatchable devices, thereby reducing the number of sensors and controllers in the system. The convergence of the proposed strategy and the optimal equilibrium points are theoretically proven. Finally, simulations of a 30-bus DC microgrid are performed to validate the effectiveness of the proposed designs.

For 0 〈β 〈 1, the author once wrote a paper to deal with the representation problem of the conjugate cone [L^β[0, 1]β^＊ of complex β-nanach space L^β[0, 1]. In this paper, replacing [0, 1] with a Borel finite measure space （Ω,M,μ） and replacing the complex field C with a Banach space X, we study the representation problem of the conjugate cone [L^β（μ, X）]β^＊ of L^β（μ, X）, and obtain [L^β（μ, X）]β^＊≌L^∞ M^＋（μ, S）, called the Quasi-Representation Theorem of [L^β（μ, X）]β^＊.

The condensation of benzaldehyde, urea, and ethyl acetoacetate according to the procedure described by Biginelli was investigated at the B3LYP/6-31G（d）, B3LYP/6-31 ＋G（d,p）, and B3LYP/6-311＋G（3df,2p）//B3LYP/6-31＋G（d,p） levels to explore the reaction mechanism. According to the mechanism proposed by Kappe, structures of five intermediates were optimized and four transition states were found. The calculation results proved that the mechanism proposed by Kappe is right.

This paper presents an improved gravitational search algorithm （IGSA） as a hybridization of a relatively recent evolutionary algorithm called gravitational search algorithm （GSA）, with the free search differential evolution （FSDE）. This combination incorporates FSDE into the optimization process of GSA with an attempt to avoid the premature convergence in GSA. This strategy makes full use of the exploration ability of GSA and the exploitation ability of FSDE. IGSA is tested on a suite of benchmark functions. The experimental results demonstrate the good performance of IGSA.
