Identification of differentially expressed mRNAs as novel predictive biomarkers for gastric cancer diagnosis and prognosis

BACKGROUND Gastric cancer(GC)has a high mortality rate *** significant progress in GC diagnosis and treatment,the prognosis for affected patients still remains *** To identify important candidate genes related to the development of GC and iden-tify potential pathogenic mechanisms through comprehensive bioinformatics *** The Gene Expression Omnibus database was used to obtain the GSE183136 dataset,which includes a total of 135 GC *** limma package in R software was employed to identify differentially expressed genes(DEGs).Thereafter,enrichment analyses of Gene Ontology(GO)terms and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways were performed for the gene modules using the clusterProfile package in R *** protein-protein interaction(PPI)networks of target genes were constructed using STRING and visualized by Cytoscape *** common hub genes that emerged in the cohort of DEGs that was retrieved from the GEPIA database were then screened using a Venn *** expression levels of these overlapping genes in stomach adenocarcinoma samples and non-tumor samples and their association with prognosis in GC patients were also obtained from the GEPIA database and Kaplan-Meier ***,real-time quantitative polymerase chain reaction(RT-qPCR)and western blotting were performed to determine the mRNA and protein levels of glutamic-pyruvic transaminase(GPT)in GC and normal immortalized cell *** addition,cell viability,cell cycle distribution,migration and invasion were evaluated by cell counting kit-8,flow cytometry and transwell ***,we also conducted a retrospective analysis on 70 GC patients diagnosed and surgically treated in Wenzhou Central Hospital,Dingli Clinical College of Wenzhou Medical University,The Second Affiliated Hospital of Shanghai University between January 2017 to December *** tumor and adjacent normal samples were collected from the patients to determine the

Two-grid Method of Expanded Mixed Finite Element Approximations for Parabolic Integro-differential Optimal Control Problems

This paper aims to construct a two-grid scheme of fully discretized expanded mixed finite element methods for optimal control problems governed by parabolic integro-differential equations and discuss a priori error estimates. The state variables and co-state variables are discretized by the lowest order Raviart-Thomas mixed finite element, and the control variable is approximated by piecewise constant functions. The time derivative is discretized by the backward Euler method. Firstly, we define some new mixed elliptic projections and prove the corresponding error estimates which play an important role in subsequent convergence ***, we derive a priori error estimates for all variables. Thirdly, we present a two-grid scheme and analyze its convergence. In the two-grid scheme, the solution of the parabolic optimal control problem on a fine grid is reduced to the solution of the parabolic optimal control problem on a much coarser grid and the solution of a decoupled linear algebraic system on the fine grid and the resulting solution still maintains an asymptotically optimal accuracy. At last, a numerical example is presented to verify the theoretical results.

Unexpected difficult airway due to severe upper tracheal distortion:A case report

BACKGROUND Difficult airway is a significant cause of anesthesia-associated death and ***,physical examinations of thyromental distance,mouth opening,Mafampaii classification,*** with X-ray,computed tomography(CT),and other imaging technologies are mainly used to evaluate difficult ***,in many special cases,i.e.,emergency surgery,imaging examinations cannot be completed *** patients’airway can only be evaluated through general physical examination,which inevitably increases the likelihood of an unexpected difficult airway during *** SUMMARY We report a rare case of difficult intubation because of severe upper trachea distortion after *** holmium laser lithotripsy was performed under transurethral ureteroscopy because the patient had anuria for 4 d and a creatinine level of 890μmol/*** to the urgency of the condition,chest radiography or chest CT was not examined before surgery and the anesthesiologist did not evaluate the airway adequately,resulting in an unexpected difficult *** The incidence of tracheal malformation and tracheal stenosis is extremely low,but the risk of hypoxia and even death due to difficult airways is extremely high for such *** is recommended to complete preoperative imaging examinations of the *** life-threatening emergency patients,a pre-anesthesia reassessment should be performed and surgeons should be prepared to prevent and manage the difficult airway.

New insight into Ca^(2+)-permeable channel in plant immunity

Calcium ions(Ca^(2+)) are crucial intracellular second messengers in eukaryotic cells. Upon pathogen perception, plants generate a transient and rapid increase in cytoplasmic Ca^(2+)levels, which is subsequently decoded by Ca^(2+)sensors and effectors to activate downstream immune responses. The elevation of cytosolic Ca^(2+)is commonly attributed to Ca^(2+)influx mediated by plasma membranelocalized Ca^(2+)–permeable channels. However, the contribution of Ca^(2+)release triggered by intracellular Ca^(2+)-permeable channels in shaping Ca^(2+)signaling associated with plant immunity remains poorly understood. This review discusses recent advances in understanding the mechanism underlying the shaping of Ca^(2+)signatures upon the activation of immune receptors, with particular emphasis on the identification of intracellular immune receptors as non-canonical Ca^(2+)-permeable channels. We also discuss the involvement of Ca^(2+)release from the endoplasmic reticulum in generating Ca^(2+)signaling during plant immunity.

In situ embedded bismuth nanoparticles among highly porous carbon fibers for efficient carbon dioxide reduction

Electrocatalysis provides an optimal approach for the conversion of carbon dioxide(CO2) into high-value chemicals, thereby presenting a promising avenue toward achieve carbon neutrality. However, addressing the selectivity and stability challenges of metal catalysts in electrolytic reduction remains a daunting task. In this study, the electrospinning method is employed to fabricate porous carbon nanofibers loaded with bismuth nanoparticles with the help of in situ pyrolysis. The porous carbon nanofibers as conductive support would facilitate the dispersion of bismuth active sites while inhibiting their aggregation and promoting the mass transfer, thus enhancing their electrocatalytic activity and stability. Additionally, nitrogen doping induces electron delocalization in bismuth atoms through metal-support interactions, thus enabling efficient adsorption of intermediates for improving selectivity based on the theoretical calculation. Consequently, Bi@PCNF-500 exhibits the exceptional selectivity and stability across a wide range of potential windows. Notably, its faradaic efficiency(FE) of formate reaches 92.7% in H-cell and94.9% in flow cell, respectively, with good electrocatalytic stability. The in situ characterization and theoretical calculations elucidate the plausible reaction mechanism to obtain basic rules for designing efficient electrocatalyst.

The nucleocapsid protein(NP)plays a crucial role in SARS-CoV-2 replication and is the most abundant structural protein with a long *** its vital role in severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)assembly and host inflammatory response,it remains an unexplored target for drug *** this study,we identified a small-molecule compound(ciclopirox)that promotes NP degradation using an FDA-approved library and a drug-screening cell *** significantly inhibited SARS-CoV-2 replication both in vitro and in vivo by inducing NP *** induced abnormal NP aggregation through indirect interaction,leading to the formation of condensates with higher viscosity and lower *** condensates were subsequently degraded via the autophagy-lysosomal pathway,ultimately resulting in a shortened NP half-life and reduced NP *** results suggest that NP is a potential drug target,and that ciclopirox holds substantial promise for further development to combat SARS-CoV-2 replication.

Carbon-based nanomaterials cause toxicity by oxidative stress to the liver and brain in Sprague-Dawley rats

Carbon-based nanomaterials have important research significance in various disciplines,such as composite materials,nanoelectronic devices,biosensors,biological imaging,and drug ***,the human and ecological risks associated with carbon-based nanomaterials have received increasing ***,the biological safety of carbon based nanomaterials has not been systematically *** this study,we used different types of carbon materials,namely,graphene oxide(GO),single-walled carbon nanotubes(SWCNTs),and multiwalled carbon nanotubes(MWCNTs),as models to observe their distribution and oxidative damage in *** results of Histopathological and ultrastructural examinations indicated that the liver and lungs were the main accumulation targets of these ***-μ-XRF analysis revealed that SWCNTs and MWCNTs might be present in the *** shows that the three types of carbon-based nanomaterials could cross the gas-blood barrier and eventually reach the liver *** addition,SWCNTs and MWCNTs could cross the blood-brain barrier and accumulate in the cerebral *** increase in ROS and MDA levels and the decrease in GSH,SOD,and CAT levels indicated that the three types of nanomaterials might cause oxidative stress in the *** suggests that direct instillation of these carbon-based nanomaterials into rats could induce ROS *** addition,iron(Fe)contaminants in these nanomaterials were a definite source of free ***,these nanomaterials did not cause obvious damage to the rat brain *** deposition of selenoprotein in the rat brain was found to be related to oxidative stress and Fe *** information may support the development of secure and reasonable applications of the studied carbon-based nanomaterials.
