Uncovering Chemical Interactions between Danshen and Danggui Using Liquid Chromatography–Mass Spectrometry and Network Pharmacology-Based Research on Stroke

Objective: The objective of this study was to decipher chemical interactions between Danshen and Danggui using liquid chromatography–mass spectrometry(LC-MS) and explore the mechanisms of Danshen–Danggui against stroke using network pharmacology and molecular docking. Materials and Methods: First, the chemical compounds of Danshen–Danggui were profiled using ultra-high-performance liquid chromatography(HPLC)-quadrupole time-of-flight MS. Accurately characterized compounds in various proportions of Danshen–Danggui were quantified using HPLC combined with triple quadrupole electrospray tandem MS. Network pharmacology was used to uncover the essential mechanisms of action of Danshen–Danggui against stroke. Discovery Studio Software was used for the molecular docking verification of key active chemicals and stroke-related targets. Results: A total of 53 compounds were characterized, and 22 accurately identified constituents(10 phenolic acids, 8 phthalides, and 4 tanshinones) were quantified in 15 proportions of Danshen–Danggui. The quantification results showed that Danggui significantly increased the dissolution of most phenolic acids(compounds from Danshen), whereas Danshen promoted the dissolution of most phthalides(compounds from Danggui). Overall, the combination of Danshen and Danggui at a 1:1 ratio resulted in the maximum total dissolution rate. Further network pharmacology and molecular docking results indicated that Danshen–Danggui exerted anti-stroke effects mainly by regulating inflammation-related(tumor necrosis factor, hypoxia-inducible factor, and toll-like receptor) signaling pathways, which ranked among the top three pathways based on Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis. Conclusion: The chemical compounds in Danshen–Danggui could interact with each other to increase the dissolution of the most active compounds, which could provide a solid basis for uncovering the compatibility mechanisms of Danshen–Danggui and Danshen–Danggui-based formulae.

OXCT1 regulates NF-κB signaling pathway through β-hydroxybutyrate-mediated ketone body homeostasis in lung cancer

Metabolic reprogramming is one of the hallmarks of cancer.1 Ketone bodies behave as alternative fuel for cancer cells to support survival and proliferation.2 3-Oxoacid CoA-transferase 1 (OXCT1) is a key enzyme that catalyzes the first and rate-limiting step of ketolysis. Recently, several studies have revealed the significance of OXCT1 in cancer development, though the underlying mechanisms remain largely unknown.3 In this study, we revealed a novel regulatory mechanism for tumorigenesis that OXCT1 regulated SREBP1-TRIM21-p65 axis through ketone body homeostasis in non-small cell lung cancer (NSCLC). In terms of mechanism, we found that OXCT1 could activate NF-κB signaling pathway by suppressing transcriptional activity of the sterol regulatory element binding protein 1 (SREBP1). As a transcription factor, SREBP1 could bind to the promoter of E3 ubiquitin ligase TRIM21, which mediated the ubiquitination of p65. Furthermore, we demonstrated that OXCT1 could maintain the homeostasis of β-hydroxybutyrate (β-HB), which acted as a signaling metabolite to activate SREBP1. Thus, β-HB connected OXCT1 with SREBP1 to activate NF-κB signaling pathway and promoted tumor initiation and progression. Taken together, these findings highlight a previously unappreciated mechanism for activation of NF-κB signaling by OXCT1 and ketone body, and demonstrate that targeting OXCT1 can inhibit NSCLC tumorigenesis.

Formaldehyde(HCHO)is considered one of the most abundant gas-phase carbonyl compounds in the atmosphere,which can be directly emitted through transportation sources.Long-Path Differential Optical Absorption Spectroscopy(LP-DOAS)was used to observe HCHO in the river channel of Wusong Wharf in Shanghai,China for the whole year of 2019.Due to the impact of ship activity,the annual average HCHO level in the channel is about2.5 times higher than that in the nearby campus environment.To explain the sources of HCHO under different meteorological conditions,the tracer-pair of CO and O_(x)(NO_(2)+O_(3))was used on the clustered air masses.The results of the source appointment show that primary,secondary and background account for 24.14%(3.34±1.19 ppbv),44.78%(6.20±2.04ppbv)and 31.09%(4.31±2.33 ppbv)of the HCHO in the channel when the air masses were from the mixed direction of the city and channel,respectively.By performing background station subtraction at times of high primary HCHO values and resolving the plume peaks,directly emitted HCHO/NO_(2)in the channel environment and plume were determined to be mainly distributed between 0.2 and 0.3.General cargo ships with higher sailing speeds or main engine powers tend to have higher HCHO/NO_(2)levels.With the knowledge of NO_(2)(or NO_(x))emission levels from ships,this study may provide data support for the establishment of HCHO emission factors.

Postoperative accurate pain assessment of children and artificial intelligence: A medical hypothesis and planned study

Although the pediatric perioperative pain management has been improved in recent years,the valid and reliable pain assessment tool in perioperative period of children remains a challenging task.Pediatric perioperative pain management is intractable not only because children cannot express their emotions accurately and objectively due to their inability to describe physiological characteristics of feeling which are different from those of adults,but also because there is a lack of effective and specific assessment tool for children.In addition,exposure to repeated painful stimuli early in life is known to have short and long-term adverse sequelae.The short-term sequelae can induce a series of neurological,endocrine,cardiovascular system stress related to psychological trauma,while long-term sequelae may alter brain maturation process,which can lead to impair neurodevelopmental,behavioral,and cognitive function.Children’s facial expressions largely reflect the degree of pain,which has led to the developing of a number of pain scoring tools that will help improve the quality of pain mana-gement in children if they are continually studied in depth.The artificial inte-lligence(AI)technology represented by machine learning has reached an unprecedented level in image processing of deep facial models through deep convolutional neural networks,which can effectively identify and systematically analyze various subtle features of children’s facial expressions.Based on the construction of a large database of images of facial expressions in children with perioperative pain,this study proposes to develop and apply automatic facial pain expression recognition software using AI technology.The study aims to improve the postoperative pain management for pediatric population and the short-term and long-term quality of life for pediatric patients after operational event.

Exact Box-Constrained Economic Operating Region for Power Grids Considering Renewable Energy Sources

The growing integration of renewable energy sources manifests as an effective strategy for reducing carbon emissions. This paper strives to efficiently approximate the set of optimal scheduling plans(OSPs) to enhance the performance of the steady-state adaptive cruise method(SACM) of power grid, improving the ability of dealing with operational uncertainties. Initially, we provide a mathematical definition of the exact boxconstrained economic operating region(EBC-EOR) for the power grid and its dispatchable components. Following this, we introduce an EBC-EOR formulation algorithm and the corresponding bi-level optimization models designed to explore the economic operating boundaries. In addition, we propose an enhanced big-M method to expedite the computation of the EBCEOR. Finally, the effectiveness of the EBC-EOR formulation, its economic attributes, correlation with the scheduling plan underpinned by model predictive control, and the significant improvement in computational efficiency(over twelvefold) are verified through case studies conducted on two test systems..

Atomic layer deposition in advanced display technologies:from photoluminescence to encapsulation

Driven by the growing demand for next-generation displays,the development of advanced luminescent materials with exceptional photoelectric properties is rapidly accelerating,with such materials including quantum dots and phosphors,etc.Nevertheless,the primary challenge preventing the practical application of these luminescent materials lies in meeting the required durability standards.Atomic layer deposition(ALD)has,therefore,been employed to stabilize luminescent materials,and as a result,flexible display devices have been fabricated through material modification,surface and interface engineering,encapsulation,cross-scale manufacturing,and simulations.In addition,the appropriate equipment has been developed for both spatial ALD and fluidized ALD to satisfy the low-cost,high-efficiency,and high-reliability manufacturing requirements.This strategic approach establishes the groundwork for the development of ultra-stable luminescent materials,highly efficient light-emitting diodes(LEDs),and thin-film packaging.Ultimately,this significantly enhances their potential applicability in LED illumination and backlighted displays,marking a notable advancement in the display industry.

Effect of Nozzle Inclination Angle on Fuel-Air Mixing and Combustion in a Heavy Fuel Engine

Heavy-fuel engines are widely used in UAVs(Unmanned Autonomous Vehicles)because of their reliability and high-power density.In this study,a combustion model for an in-cylinder direct injection engine has been imple-mented using the AVL FIRE software.The effects of the angle of nozzle inclination on fuel evaporation,mixture distribution,and combustion in the engine cylinder have been systematically studied at 5500 r/min and consider-ing full load cruise conditions.According to the results,as the angle of nozzle inclination increases,the maximum combustion explosion pressure in the cylinderﬁrst increases and then it decreases.When the angle of nozzle incli-nation is less than 45°,the quality of the mixture in the cylinder and the combustion performance can be improved by increasing the angle.When the angle of nozzle inclination is greater than 45°,however,the mixture unevenness increases slightly with the angle,leading to a deterioration of the combustion performances.When the angle of nozzle inclination is between 35°and 55°,the overall combustion performance of the engine is rela-tively good.When the angle of nozzle inclination is 45°,the combustion chamber’s geometry and the cylinder’s airﬂow are well matched with the fuel spray,and the mixture quality is the best.Compared with 25°,the peak heat release rate increases by 20%,and the maximum combustion burst pressure increases by 5.5%.

Mitogenome-based phylogeny of mosquitoes(Diptera:Culicidae)

Mosquitoes are of great medical significance as vectors of many deadly diseases.Mitogenomes have been widely used in phylogenetic studies,but mitogenome knowledge within the family Culicidae is limited,and Culicidae phylogeny is far from resolved.In this study,we surveyed the mitogenomes of 149 Culicidae species,including 7 newly sequenced species.Comparative analysis of 149 mosquito mitogenomes shows gene composition and order to be identical to that of an ancestral insect,and the AT bias,length variation,and codon usage are all consistent with that of other reported Dipteran mitogenomes.Phylogenetic analyses based on the DNA sequences of the 13 protein-coding genes from the 149 species robustly support the monophyly of the subfamily Anophelinae and the tribes Aedini,Culicini,Mansonini,Sabethini,and Toxorhynchitini.To resolve ambiguous relationships between clades within the subfamily Culicinae,we performed topological tests and show that Aedini is a sister to Culicini and that Uranotaenini is a sister to(Mansonini)+(Toxorhynchitini+Sabethini).In addition,we estimated divergence times using a Bayesian relaxation clock based on the sequence data and 3 fossil calibration points.The results show mosquitoes diverged during the Early Jurassic with massive Culicinae radiations during the Cretaceous,coincident with the emergence of angiosperms and the burst of mammals and birds.Overall,this study,which uses the largest number of Culicidae mitogenomes sequenced to date,comprehensively reveals the mitogenome characteristics and mitogenome-based phylogeny and divergence times of Culicidae,providing information for further studies on the mitogenome,phylogeny,evolution,and taxonomic revision of Culicidae.

Improving mathematical model of burden distribution and correcting chute angle to cope with fluctuation of stock line

Accurate evaluations of the burden distribution are of critical importance to stabilize the operation of blast furnace.The mathematical model and discrete element method(DEM)are two attractive methods for predicting burden distribution.Based on DEM,the initial velocities of the pellet,sinter,and coke were calculated,and the velocity attenuations of the above three particles between the burden and the chute were analyzed.The initial velocity and velocity attenuation were applied to a mathematical model for improving the accuracy.Additionally,based on the improved model,a scheme for rectifying the chute angles was proposed to address the fluctuation of the stock line and maintain a stable burden distribution.The validity of the scheme was confirmed via a stable burden distribution under different stock lines.The mathematical model has been successfully applied to evaluate the online burden distribution and cope with the fluctuation of the stock line.

Effect of screening colonoscopy frequency on colorectal cancer mortality in patients with a family history of colorectal cancer

BACKGROUND Colorectal cancer is a common malignant tumor in China,and its incidence in the elderly is increasing annually.Inflammatory bowel disease is a group of chronic non-specific intestinal inflammatory diseases,including ulcerative colitis and Crohn’s disease.We included the clinicopathological and follow-up data of patients with colorectal cancer who underwent laparoscopic colectomy or open colectomy at our Gastrointestinal Department between January 2019 and December 2022.Surgical indicators,oncological indicators,and survival rates were compared between the groups.The results of 104 patients who met the above criteria were extracted from the database(laparoscopic colectomy group=63,open colectomy group=41),and there were no statistically significant differences in the baseline data or follow-up time between the two groups.RESULTS Intraoperative blood loss,time to first ambulation,and time to first fluid intake were significantly lower in the laparoscopic colectomy group than in the open colectomy group.The differences in overall mortality,tumor-related mortality,and recurrence rates between the two groups were not statistically significant,and survival analysis showed that the differences in the cumulative overall survival,tumor-related survival,and cumulative recurrence-free rates between the two groups were not statistically significant.CONCLUSION In elderly patients with colorectal cancer,laparoscopic colectomy has better short-term outcomes than open colectomy,and laparoscopic colectomy has superior long-term survival outcomes compared with open colectomy.
