Clinical significance of tumor-associated macrophage infiltration in supraglottic laryngeal carcinoma

Tumor-associated macrophages(TAMs) can elicit contrasting effects on tumor progression,depending on different tumor *** study aimed to explore the correlation between TAM infiltration and clinicopathologic characteristics,metastasis,and prognosis of supraglottic laryngeal *** in intratumoral and peritumoral regions of 84 specimens of supraglottic laryngeal carcinoma tissues were detected by immunohistochemical staining with monoclonal CD68 *** density of peritumoral CD68+ TAMs in recurrence cases(9/11) and in dead cases(17/23) were significantly higher than those in non-recurrence cases(33/73) and in survival cases(25/61),with significant differences(P = 0.024 and 0.007,respectively).The Kaplan-Meier survival analysis showed a significant relationship between the infiltration of both intratumoral and peritumoral CD68 + TAMs and the overall survival of *** 5year survival rate was significantly lower in the group with a high density of intratumoral CD68+ TAMs than in the group with a low density(39.6% vs.82.5%,P < 0.05).Similarly,the 5-year survival rate was significantly lower in the group with a high density of peritumoral CD68+ TAMs than in the group with a low density(50.6% vs.73.1%,P < 0.05).Cox regression analysis revealed that T classification,distant metastasis,and intratumoral or peritumoral CD68 + TAMs were independent factors for disease-free survival,whereas T classification and intratumoral CD68 + TAMs were independent factors for overall *** results indicate that TAM infiltration in supraglottic laryngeal carcinoma can be used to predict metastasis and prognosis and is an independent factor for prognosis.

Antiproliferative effects of cinobufacini on human hepatocellular carcinoma HepG2 cells detected by atomic force microscopy

AIM:To investigate the antiproliferative activity of cinobufacini on human hepatocellular carcinoma HepG2 cells and the possible mechanism of its ***:HepG2 cells were treated with different concentrations of *** viability was measured by methylthiazolyl tetrazolium(MTT) *** cycledistribution was analyzed by flow cytometry(FCM).Cytoskeletal and nuclear alterations were observed by fluorescein isothiocyanate-phalloidin and DAPI staining under a laser scanning confocal *** in morphology and ultrastructure of cells were detected by atomic force microscopy(AFM) at the nanoscale ***:MTT assay indicated that cinobufacini significantly inhibited the viability of HepG2 cells in a dosedependent *** the concentration of cinobufacini increasing from 0 to 0.10 mg/m L,the cell viability decreased from 74.9% ± 2.7% to 49.41% ± 2.2% and 39.24% ± 2.1%(P < 0.05).FCM analysis demonstrated cell cycle arrest at S phase induced by *** immunofluorescence studies of cytoskeletal and nuclear morphology showed that after cinobufacini treatment,the regular reorganization of actin filaments in HepG2 cells become chaotic,while the nuclei were not damaged ***,high-resolution AFM imaging revealed that cell morphology and ultrastructure changed a lot after treatment with *** appeared as significant shrinkage and deep pores in the cell membrane,with larger particles and a rougher cell ***:Cinobufacini inhibits the viability of HepG2 cells via cytoskeletal destruction and cell membrane toxicity.

p53-dependent Fas expression is critical for Ginsenoside Rh2 triggered caspase-8 activation in HeLa cells

We have recently reported that Ginsenoside Rh2 （G- Rh2） induces the activation of two initiator caspases, caspase-8 and caspase-9 in human cancer cells. How- ever, the molecular mechanism of its death-inducing function remains unclear. Here we show that G-Rh2 stimulated the activation of both caspase-8 and cas- pase-9 simultaneously in HeLa cells. Under G-Rh2 treatment, membrane death receptors Fas and TNFR1 are remarkably upregulated. However, the induced expression of Fas but not TNFR1 was contributed to the apoptosis process, Moreover, significant increases in Fas expression and caspase-8 activity temporally coin- cided with an increase in p53 expression in p53-non- mutated HeLa and SK-HEP-1 cells upon G-Rh2 treat- ment. In contrast, Fas expression and caspase-8 activity remained constant with G-Rh2 treatment in p53-mutated SW480 and PC-3 cells. In addition, siRNA-mediated knockdown of p53 diminished G-Rh2-induced Fas expression and caspase-8 activation, These results indicated that G-Rh2-triggered extrinsic apoptosis relies on p53-mediated Fas over-expression. In the intrinsic apoptotic pathway, G-Rh2 induced strong and immedi- ate translocation of cytosolic BAK and BAX to the mitochondria, mitochondrial cytochrome c release, and subsequent caspase-9 activation both in HeLa and in SW480 ceils, p53-mediated Fas expression and sub- sequent downstream caspase-8 activation as well as p53-independent caspase-9 activation all contribute to the activation of the downstream effector caspase-3/-7, leading to tumor cell death. Taken together, we suggest that G-Rh2 induces cancer cell apoptosis in a multi-path manner and is therefore a promising candidate for anti- tumor drug development.

Conventional thermosetting polymers,mostly derived from nonrenewable petroleum resources,are not reprocessable and recyclable due to their highly cross-linked three-dimensional networks and face the disadvantage of high *** solve these issues,in this study,we synthesized a novel Schiff base covalent adaptable thermoset from a furan-derived tri-aldehyde monomer(TMFP)and a furan-derived di-amine monomer(DFDA).The as-prepared TMFP-DFDA-Vitrimer exhibited superior anti-flammability with a high limiting oxygen index(LOI)of 35.0%and a UL-94 V-0 rating,which was attributed to the excellent charring ***,TMFPDFDA-Vitrimer could also be conveniently recycled by chemical decomposition under a mixed hydrochloric acid/tetrahydrofuran(HCl/THF)*** recycling for 5 times,the thermal,mechanical,and flame retardant properties of the recycled TMFP-DFDA-Vitrimer retained almost unchanged compared to the original *** work provides a prime instance to develop advanced thermosetting polymers from abundant furan-based compounds.

Postpartum quality of life and mental health in women with heart disease:Integrated clinical communication and treatment

The welding arc,as a carrier for the conversion of electrical energy to thermal energy,has a direct impact on the quality of welding by its properties and *** the tungsten inert gas(TIG)welding process under the condition of Ar-He alternating gas supply,the arc is alternately converted between Ar arc and He arc with an alternating gas supply cycle,which has obvious arc change *** FLUENT software was used to numerically simulate the characteristics of the TIG arc under the condition of alternating gas supply,and the arc temperature field,arc pressure,electric potential and current density distribution under the condition of alternating gas supply were *** with the real-time data of arc pressure measured by the water-cooled copper plate with holes,it is proved that the TIG arc has obvious dynamic characteristics under the condition of Ar-He alternating gas *** unique dynamic TIG arc acts on the 5A06 aluminum alloy weld,causing the molten pool to stir,resulting in uniform microstructure and grain refinement at the weld,and thereby improving the mechanical properties of the welded joint.

Machine Learning Security Defense Algorithms Based on Metadata Correlation Features

With the popularization of the Internet and the development of technology,cyber threats are increasing day by *** such as malware,hacking,and data breaches have had a serious impact on *** network security environment in the era of big data presents the characteristics of large amounts of data,high diversity,and high real-time *** security defense methods and tools have been unable to cope with the complex and changing network security *** paper proposes a machine-learning security defense algorithm based on metadata association *** control over unauthorized users through privacy,integrity,and *** user model is established and the mapping between the user model and the metadata of the data source is *** analyzing the user model and its corresponding mapping relationship,the query of the user model can be decomposed into the query of various heterogeneous data sources,and the integration of heterogeneous data sources based on the metadata association characteristics can be *** and classify customer information,automatically identify and perceive sensitive data,build a behavior audit and analysis platform,analyze user behavior trajectories,and complete the construction of a machine learning customer information security defense *** experimental results show that when the data volume is 5×103 bit,the data storage integrity of the proposed method is 92%.The data accuracy is 98%,and the success rate of data intrusion is only 2.6%.It can be concluded that the data storage method in this paper is safe,the data accuracy is always at a high level,and the data disaster recovery performance is *** method can effectively resist data intrusion and has high air traffic control *** can not only detect all viruses in user data storage,but also realize integrated virus processing,and further optimize the security defense effect of user big data.
