Targeting epigenetic mechanisms in amyloid-β-mediated Alzheimer’s pathophysiology:unveiling therapeutic potential

Alzheimer’s disease is a prominent chronic neurodegenerative condition characterized by a gradual decline in memory leading to *** evidence suggests that Alzheimer’s disease is associated with accumulating various amyloid-βoligomers in the brain,influenced by complex genetic and environmental *** memory and cognitive deficits observed during the prodromal and mild cognitive impairment phases of Alzheimer’s disease are believed to primarily result from synaptic *** life,environmental factors can lead to enduring changes in gene expression and the emergence of brain *** changes,known as epigenetic modifications,also play a crucial role in regulating the formation of synapses and their adaptability in response to neuronal *** this context,we highlight recent advances in understanding the roles played by key components of the epigenetic machinery,specifically DNA methylation,histone modification,and microRNAs,in the development of Alzheimer’s disease,synaptic function,and activity-dependent synaptic ***,we explore various strategies,including enriched environments,exposure to non-invasive brain stimulation,and the use of pharmacological agents,aimed at improving synaptic function and enhancing long-term potentiation,a process integral to epigenetic ***,we deliberate on the development of effective epigenetic agents and safe therapeutic approaches for managing Alzheimer’s *** suggest that addressing Alzheimer’s disease may require distinct tailored epigenetic drugs targeting different disease stages or pathways rather than relying on a single drug.

Stroke is classified as ischemic or hemorrhagic,and there are few effective treatments for either *** mechanisms play a critical role in secondary brain injury following a stroke,which manifests as cytokine release,blood–brain barrier disruption,neuronal cell death,and ultimately behavioral *** the inflammatory response has been shown to mitigate this cascade of events in experimental stroke ***,in clinical trials of anti-inflammatory agents,longterm immunosuppression has not demonstrated significant clinical benefits for *** may be attributable to the dichotomous roles of inflammation in both tissue injury and repair,as well as the complex pathophysiologic inflammatory processes in *** acute harmful inflammatory responses or inducing a phenotypic shift from a pro-inflammatory to an anti-inflammatory state at specific time points after a stroke are alternative and promising therapeutic *** agents that can modulate inflammation requires a detailed understanding of the inflammatory processes of ***,epigenetic reprogramming plays a crucial role in modulating post-stroke inflammation and can potentially be exploited for stroke *** this review,we summarize current findings on the epigenetic regulation of the inflammatory response in stroke,focusing on key signaling pathways including nuclear factor-kappa B,Janus kinase/signal transducer and activator of transcription,and mitogen-activated protein kinase as well as inflammasome *** also discuss promising molecular targets for stroke *** evidence to date indicates that therapeutic targeting of the epigenetic regulation of inflammation can shift the balance from inflammation-induced tissue injury to repair following stroke,leading to improved post-stroke outcomes.

Metal-organic framework(MOF)and its derivatives have low-cost,controllable structure,and good catalytic performance,which are often used in the electrochemical *** this work,SnO_(2)in situ modified graphite felt(SnO_(2)/GF)is prepared by hydrothermal method then simple thermal treatment using Sn-based MOF(Sn-MOF)as ***_(2)is uniformly and firmly distributed on the GF surface rather than the common agglomeration and poor bonding of metal oxides on carbon-based electrodes,providing stable active centers for the VO^(2+)/VO_(2)^(+)and V^(2+)/V^(3+)redox ***250 mA·cm^(-2),the energy efficiency of the battery with SnO_(2)/GF remains at 63.2%,while the blank one has *** former battery,at 100 mA·cm^(-2),has higher energy efficiency and good cycle stability(over 200 cycles).The battery performance of this study is better than that of most previous report in metal oxide-related *** work obtains high-performance composite electrode by simple treatment of MOF,which provides a reference for the application of MOF in vanadium redox flow battery.

Self-organized titanium dioxide(TiO_(2))nanotubes,which are prepared by electrochemical anodizing,have been widely researched as promising anodes for liion *** nanotubular morphology and bulk structure of TiO_(2)nanotubes can be easily changed by adjusting the anodizing and annealing *** is provided to investigate different phenomena by selectively adjusting a specific parameter of the li^(+)insertion *** this paper,we reviewed how the morphology and crystallography of TiO_(2)nano tubes influence the electrochemical performance of li^(+)*** particular,electrochemical performances of amorphous and anatase titanium dioxide nanotube anodes were compared in *** we all know,TiO_(2)nanotube anodes have the advantages of nontoxicity,good stability,high safety and large specific surface area,in lithium-ion ***,they suffer from poor electronic conductivity,inferior ion diffusivity and low theoretical capacity(335 mAh·g^(-1)),which limit their practical ***,there are two ways to overcome the shortcomings of titanium dioxide nanotube anodes,including doping and synthesis *** achievements and existing problems associated with doped TiO_(2)nanotube anodes and composite material anodes are summarized in the present *** on the analysis of lithium insertion mechanism of titanium dioxide nanotube electrodes,the prospects and possible research directions of TiO_(2)anodes in lithiumion batteries are discussed.

Joint Bayesian and Greedy Recovery for Compressive Sensing

Greedy algorithms are widely used for sparse recovery in compressive sensing. Conventional greedy algorithms employ the inner product vector of signal residual and sensing matrix to determine the support, which is based on the assumption that the indexes of the larger-magnitude entries of the inner product vector are more likely to be contained in the correct ***, this assumption may be not valid when the number of measurements is not sufficient, leading to the selection of an incorrect support. To improve the accuracy of greedy recovery, we propose a novel greedy algorithm to recover sparse signals from incomplete *** entries of a sparse signal are modelled by the type-II Laplacian prior, such that the k indexes of the correct support are indicated by the largest k variance hyperparameters of the entries. Based on the proposed model, the supports can be recovered by approximately estimating the hyperparameters via the maximum a posteriori process. Simulation results demonstrate that the proposed algorithm outperforms the conventional greedy algorithms in terms of recovery accuracy, and it exhibits satisfactory recovery speed.

Keyword Query over Error-Tolerant Knowledge Bases

With more and more knowledge provided by WWW, querying and mining the knowledge bases have attracted much research attention. Among all the queries over knowledge bases, which are usually modelled as graphs, a keyword query is the most widely used one. Although the problem of keyword query over graphs has been deeply studied for years, knowledge bases, as special error-tolerant graphs, lead to the results of the traditional defined keyword queries out of users＇ satisfaction. Thus, in this paper, we define a new keyword query, called confident r-clique, specific for knowledge bases based on the r-clique definition for keyword query on general graphs, which has been proved to be the best one. However, as we prove in the paper, finding the confident r-cliques is #P-hard. We propose a filtering-and-verification framework to improve the search efficiency. In the filtering phase, we develop the tightest upper bound of the confident r-clique, and design an index together with its search algorithm, which suits the large scale of knowledge bases well. In the verification phase, we develop an efficient sampling method to verify the final answers from the candidates remaining in the filtering phase. Extensive experiments demonstrate that the results derived from our new definition satisfy the users＇ requirement better compared with the traditional r-clique definition, and our algorithms are efficient.

Towards practical lean-electrolyte li-S batteries:Highly solvating electrolytes or sparingly solvating electrolytes?

lithium-sulfur(li-S)batteries hold great promise to be the next-generation candidate for high-energy-density secondary batteries but in the prerequisite of using low electrolyte-to-sulfur(E/S)*** solvating electrolytes(HSEs)and sparingly solvating electrolytes(SSEs),with opposite nature towards the dissolution of polysulfides,have recently emerged as two effective solutions to decrease the E/S ratio and increase the overall practical energy density of li-S *** featuring with high polysulfide solvation ability have the potential to reduce the E/S ratio by dissolving more polysulfides with less electrolyte,while SSEs alter the sulfur reaction pathway from a dissolution-precipitation mechanism to a quasi-solid mechanism,thereby independent on the use of electrolyte *** HSEs and SSEs show respective effectiveness in lean-electrolyte li-S batteries,but encounter different challenges to bring li-S batteries into practical *** review aims to present a comparative discussion on their unique features and basic electrochemical reaction mechanisms in practical lean-electrolyte li-S *** is focused on the current technical challenges and possible solutions for their future development.

Heparanase expression,degradation of basement membrane and low degree of infi ltration by immunocytes correlate with invasion and progression of human gastric cancer

AIM： To disclose the mechanisms that accelerate or limit tumor invasion and metastasis in gastric cancer patients. METHODS： The heparanase expression, continuity of basement, degree of infiltration by dendritic cells and lymphocytes in gastric cancer tissues from 33 the early and late stage patients were examined by immunohistochemistry, in situ hybridization and transmission electron microscopy. RESULTS： Heparanase mRNA expression in the late stage patients with gastric cancer was stronger than that in the early stage gastric cancer patients. In the early stage gastric cancer tissues, basement membrane （BM） appeared intact, whereas in the late stage, discontinuous BM was often present. The density of Sl00 protein positive tumor infiltrating dendritic cells （TIDC） in the early stage gastric cancer tissues was higher than that in the late stage. The infiltrating degree of tumor infiltrating lymphocytes （TIL） in the early stage patients whose tumor tissues contained a high density of TIDC was significantly higher than that in the late stage gastric cancer tissues patients with a low density of TIDC. There were few cancer cells penetrated through the continuous BM of cancer nests in the early stage gastric cancers, but many cancer cells were found outside of the defective BM of cancer nests in the late stage. CONCLUSION： Our results suggest that strongheparanase expression is related with the degradation of BM which allows or accelerates tumor invasion and metastasis. However, high density of TIDC and degree of infiltration by TIL are associated with tumor progression in human gastric cancers.

Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and *** on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target *** has led to restrictions on current therapeutic *** this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target *** peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal *** nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury *** damage to target organs can occur,including skeletal muscle atrophy and sensory receptor *** then provide a brief review of the research advances in therapeutic approaches to peripheral nerve *** main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical ***,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ ***,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.

Normalized glandular dose coefficients for digital breast tomosynthesis using detailed Chinese breast models

The rise in breast cancer diagnoses among Chinese women has necessitated the use of X-ray breast screening,which carries a radiation *** study aimed to provide a dosimetry protocol for the Chinese female population to replace the traditional standard that utilizes simplified breast models,for the accurate estimation of the mean glandular dose of a patient undergoing digital breast tomosynthesis(DBT).The first set of detailed Chinese female breast models and representative breast parameters was *** backscatter radiation and computational efficiency,we improved the combination of these models and the Chinese reference adult female whole-body voxel *** acquisition for four commercial DBT systems that are widely employed in China was simulated using the Monte Carlo method to obtain the normalized glandular dose coefficients of DBT(D_(gN)^(DBT))and the glandular depth dose(D_(g)^(dep)(z))for different breast characteristics and X-ray *** calculated a series of D_(gN)^(DBT) values for breasts with different percentage mass glandularities(5%,25%,50%,75%,and 100%)and compressed breast thicknesses(2,3,4,5,6,and 7 cm)at various tube potentials(25,28,30,32,35,and 49 kV)and target/filter combinations(W/Rh,W/Al,Mo/Mo,Rh/Rh,and Rh/Ag).The parameter dependence of the breast characteristics and beam conditions on D_(gN)^(DBT) in detailed breast models was *** D_(gN)^(DBT) results were 14.6-51.0%lower than those of the traditional dosimetry standard in *** difference in D_(gN)^(DBT) was mainly due to a decrease in the depth of the main energy deposition area caused by the glandular distribution along the depth *** results obtained in this study may be used to improve breast dosimetry in China and provide more detailed information on risk assessment during DBT.
