Network pharmacology-based analysis of effective components and mechanism of Rhizoma coptidis in treating diabetes

Objective:To identify the active ingredients,potential targets,and mechanism of Rhizoma coptidis by bioinformatics method,and to explore the hypoglycemic effect of Rhizoma coptidis by in vitro ***:The chemical components of Rhizoma coptidis were collected through database search,and oral bioavailability and drug-likeness were used for preliminary *** targets of Rhizoma coptidis and diabetes-related targets were collected by database retrieval and reverse docking techniques,and the biological process of cross-set proteins was *** inhibitory effects of Rhizoma coptidis onα-glucosidase,α-amylase activity,and advanced glycation end products(AGEs)were determined via in vitro *** addition,the effects of Rhizoma coptidis on preadipocyte differentiation,absorption of glucose by adipocytes,and the level of intracellular triglyceride were investigated using the adipocyte differentiation ***:There were 11 potentially active ingredients in Rhizoma ***-6,caspase-3,epidermal growth factor receptor(EGFR),MYC,and estrogen receptor 1 were considered as the key *** bioinformatics analysis showed that Rhizoma coptidis played an antidiabetic role mainly via biological processes and signaling pathways including hormone receptor activity,glutathione binding,steroid binding,*** vitro experiments showed that the extract of Rhizoma coptidis inhibited the activities ofα-glucosidase andα-amylase,and the generation of AGEs;meanwhile,the extract promoted the absorption of glucose by *** addition,the extract of Rhizoma coptidis decreased triglyceride ***:Our network pharmacology and in vitro experiments demonstrate the anti-diabetic effects and possible underlying mechanisms of Rhizoma coptidis extract.

In this study,a series of Mn-Ce/Al2O3catalysts was prepared by different methods of depositionprecipitation（MnCeAl-DP）,impregnation（MnCeAl-IM） and citric acid（MnCeAl-CA）,and the distinct effect of preparation methods on NOxremoval performance at low temperature was *** show that MnCeAl-DP exhibits not only the best activity but also the highest resistance against SO2/*** the assistance of comprehensive characterizations from scanning electron microscopy（SEM）,Brunauer-Emmett-Teller（BET）,X-ray diffraction（XRD）,H2-temperature programmed reduction（H2-TPR）,NH3-te mperature programmed deso rption（NH3-TPD）,and X-ray photoelectron spectroscopy（XPS）,it is revealed that the MnCeAl-DP sample owns admired features of large surface area and pore volume,enriched Mn4+and chemisorbed oxygen species originating from enhanced interaction between MnOxand CeO2,as well as improved adsorption capacity to NH3and *** these factors contribute to activity *** in-situ DRIFTS studies reveal that the improvement of NH3-SCR performance over MnCeAI-DP is related to the formation of abundant nitrate species,which is beneficial to the "NH4NO3" reaction pathway and thus enhances low-temperature activity.

Polyampholyte gels,which have hierarchical structures,exhibit excellent self-healing properties and have great promise for biomaterials and *** investigated the relationship between microscopic structures and macroscopic viscoelastic properties of polyampholyte gels and found three factors influencing their viscoelastic properties,including the chemical crosslinking bonds,topological entanglements controlled by monomer concentration,and the ionic *** strength plays a major role on the strength of ionic bonds.A crossover point of elastic modulus and loss modulus was observed in the dynamic frequency sweeps at low monomer concentration or low chemical crosslinking density for gels with intermediate strength of ionic *** solid-liquid transition signaled by the crossover point is a typical feature of dynamic associated gels,representing the dynamical association-dissociation of the ionic bonds and full relaxation of the topological entanglements in the gel *** the crossover point disappears when the ionic bonds are too weak or too strong to form“permanent”***,in the non-linear yielding measurement,gels with intermediate strength of the ionic bonds are ductile and yield at very large shear strain due to the self-healing properties and the dynamic association-dissociation of the ionic *** the self-healing properties disappear when the ionic bond strength is too weak or too *** work reveals the mechanism of how the dynamic association-dissociation of ionic bonds influences both the linear and non-linear viscoelastic properties of the polyampholyte gels.

In acidic paddy fields of South China,rice(Oryza sativa L.)faces the dual challenges of cadmium(Cd)toxicity and silicon(Si)*** previous studies have highlighted the functions of Si application timing and strategies in mitigating Cd-stressed rice,the precise mechanisms underlying the health restoration of Cd-toxic rice and the assurance of grain safety remain *** study explored Cd translocation and detoxification in the shoots of rice regulated by various Si fertilization regimes:Si(T)(all Si added before transplanting),Si(J)(all Si added at jointing),and Si(TJ)(half Si added both before transplanting and at jointing).The findings revealed that the regime of Si(TJ)was more beneficial to rice health and grain safety than Si(T)and Si(J).The osmotic regulators such as proline,soluble sugars,and soluble proteins were significantly boosted by Si(TJ)compared to other Si treatments,and which enhanced membrane integrity,balanced intracellular pH,and increased Cd tolerance of ***,Si(TJ)was more effective than Si(T)and Si(J)on the Cd sequestration in the cell wall,Cd bio-passivation,and the down-regulated expression of the Cd transport *** concentrations of Cd in the xylem and phloem treated with Si(TJ)were reduced ***,Si(TJ)facilitated much more Cd bound with the outer layer proteins of grains,and promoted Cd chelation and complexation by phytic acid,phenolics,and ***,Si(TJ)outperformed Si(T)and Si(J)in harmonizing the phycological processes,inhibiting Cd translocation,and enhancing Cd detoxification in rice *** the split Si application strategy offers potential for reducing Cd toxicity in rice grain.
