RNA-based nanomedicines encompass a range of therapeutic approaches that utilize RNA molecules or molecules that target RNAs for the treatment or prevention of *** include antisense oligonucleotides(ASOs),small interfering RNAs(siRNAs),endogenous microRNAs(miRNAs),messenger RNAs(mRNAs),clustered regularly interspersed short palindromic repeats-associated protein 9(CRISPR/Cas9),single guide RNAs(sgRNAs),as well as RNA *** therapeutic agents exert their effects through various mechanisms such as gene inhibition,addition,replacement,and *** advancement of RNA biology and the field of RNA therapy has paved the way for the development and utilization of RNA-based nanomedicine in human *** remarkable example of RNA-based nanomedicine is the mRNA-based vaccines including mRNA-1273(Moderna)and BNT162b2(Pfizer/BioNTech)that have been successfully employed in response to the coronavirus disease 2019(COVID-19)*** review aims to highlight the advantages of RNA-based nanomedicines,provides an overview of significant developments in delivery systems,elucidates the molecular mechanisms of action underlying RNA-based nanomedicines,and discusses their clinical ***,the review will address the existing challenges and innovations in delivery platforms while exploring the future possibilities for these promising RNA-based nanomedicines.

Stress-assisted corrosion mechanism of 3Ni steel by using gradient boosting decision tree machining learning method

Traditional 3Ni weathering steel cannot completely meet the requirements for offshore engineering development,resulting in the design of novel 3Ni steel with the addition of microalloy elements such as Mn or Nb for strength enhancement becoming a *** stress-assisted corrosion behavior of a novel designed high-strength 3Ni steel was investigated in the current study using the corrosion big data *** information on the corrosion process was recorded using the galvanic corrosion current monitoring *** gradi-ent boosting decision tree(GBDT)machine learning method was used to mine the corrosion mechanism,and the importance of the struc-ture factor was *** exposure tests were conducted to verify the calculated results using the GBDT *** indic-ated that the GBDT method can be effectively used to study the influence of structural factors on the corrosion process of 3Ni ***-ferent mechanisms for the addition of Mn and Cu to the stress-assisted corrosion of 3Ni steel suggested that Mn and Cu have no obvious effect on the corrosion rate of non-stressed 3Ni steel during the early stage of *** the corrosion reached a stable state,the in-crease in Mn element content increased the corrosion rate of 3Ni steel,while Cu reduced this *** the presence of stress,the increase in Mn element content and Cu addition can inhibit the corrosion *** corrosion law of outdoor-exposed 3Ni steel is consistent with the law based on corrosion big data technology,verifying the reliability of the big data evaluation method and data prediction model selection.

Very high-energy electrons(VHEEs)are potential candidates for FLASH radiotherapy for deep-seated *** proposed a compact VHEE facility based on an X-band high-gradient high-power *** this study,we investigated and realized the first X-band backward traveling-wave(BTW)accelerating structure as the buncher for a VHEE facility.A method for calculating the parameters of single cell from the field distribution was introduced to simplify the design of the BTW ***-domain circuit equations were applied to calculate the transient beam parameters of the buncher in the unsteady state.A prototype of the BTW structure with a thermionic cathode-diode electron gun was designed,fabricated,and tested at high power at the Tsinghua X-band high-power test *** structure successfully operated with 5-MW microwave pulses from the pulse compressor and outputted electron bunches with an energy of 8 MeV and a pulsed current of 108 mA.

Contribution of high-latitude permafrost regions in the Northern Hemisphere to global wildfire carbon emissions

Wildfires are major disturbances in permafrost ecosystems, with increasing frequency and intensity in recent years. In permafrost regions, wildfires not only burn surface and subsurface organic matter but also accelerate permafrost thawing,releasing significant amounts of greenhouse gases such as carbon dioxide and methane into the atmosphere. However, the contribution of high-latitude permafrost regions in the Northern Hemisphere to global wildfire carbon emissions remains poorly understood. This study integrates remote sensing data and ground observations to analyze the contributions of aboveground and belowground fuel combustion in high-latitude permafrost regions to global wildfire carbon emissions from 2002 to 2020, as well as the spatiotemporal variations in these contributions. Our findings indicate that permafrost regions contribute approximately11.96% of global wildfire carbon emissions, with aboveground emissions accounting for approximately 3.94% of global aboveground emissions and belowground emissions contributing approximately 63.57% of global belowground emissions. The contribution of high-latitude permafrost regions to global emissions peaked in July and August, whereas the continuous permafrost zones(areas with more than 90% permafrost coverage) showed the most significant increase in June and July. The contributions of both aboveground and belowground emissions from high-latitude permafrost regions to global wildfire emissions have been increasing, primarily due to the reduction in global wildfire emissions, in contrast with the rising emissions from wildfires in high-latitude permafrost regions. This study highlights the significant role of wildfires, particularly the combustion of belowground biomass in high-latitude permafrost regions, in global carbon emissions. Design method and experimental study of a cathode tool with an extremely high leveling ratio for electrochemical machining of blisk

To obtain final parts with the desired dimensional accuracy and repeatability via electrochemical machining(ECM), the machining process must enter an ECM balanced state. However,for the ECM processing of blisk, a key component of aerospace engines, the surface of the blade blank often has an uneven allowance distribution due to the narrow passage of the cascade. It is difficult to remedy this issue in subsequent processing steps, which is necessary to ensure the dimensional accuracy and repeatability of the final blade profile. To solve this problem, electrolytic machining must be preceded by electrolytic shaping, which requires cathode tools with large leveling ratios to quickly homogenize the blank surface of the blade. In this study, to obtain a cathode tool with an extremely high leveling ratio, a design method based on the variation in the electrode gap in the non-equilibrium electrolytic state is proposed, and a dissolution model based on the nonequilibrium state is established. In this design method, the allowance on the blank to be machined is first divided into many discrete allowances with the normal direction. The initial machining clearance, feed rate, and total machining time are then calculated using classical ECM equilibrium state theory based on the maximum allowance. Meanwhile, the point coordinates of the cathode tool at maximum allowance can be determined. The non-equilibrium model can then be used to calculate the relative coordinate positions corresponding to the remaining discrete allowances. Finally, the entire cathode tool profile is designed. Simulations, fundamental experiments, and blisk unit workpiece experiments were carried out to validate the design approach. In the simulated processing of the plane workpiece, the leveling ratio of the cathode tool designed by the proposed method(0.77)was 83% higher than that of the cathode tool designed using the traditional method. Improving the Short-Range Precipitation Forecast of Numerical Weather Prediction through a Deep Learning-Based Mask Approach

Due to various technical issues,existing numerical weather prediction(NWP)models often perform poorly at forecasting rainfall in the first several *** correct the bias of an NWP model and improve the accuracy of short-range precipitation forecasting,we propose a deep learning-based approach called UNet Mask,which combines NWP forecasts with the output of a convolutional neural network called *** UNet Mask involves training the UNet on historical data from the NWP model and gridded rainfall observations for 6-hour precipitation *** overlap of the UNet output and the NWP forecasts at the same rainfall threshold yields a *** UNet Mask blends the UNet output and the NWP forecasts by taking the maximum between them and passing through the mask,which provides the corrected 6-hour rainfall *** evaluated UNet Mask on a test set and in real-time *** results showed that UNet Mask outperforms the NWP model in 6-hour precipitation prediction by reducing the FAR and improving CSI *** tests also showed that different small rainfall thresholds applied to the UNet and the NWP model have different effects on UNet Mask's forecast *** study shows that UNet Mask is a promising approach for improving rainfall forecasting of NWP models.
