Construction of a New Molecular Barcode for Discriminating Plants and Animals with a Close Genetic Relationship

DNA barcodes have been proposed as a shortcut to provide species identification and as a way to accelerate the discovery of new species. A number of candidate gene regions have been suggested as possible barcodes for animals and plants, but for the identification of recently diverged species and/or varieties with only a few genetic differences it has been reported to be problematic in some cases. This study selected widely cultivated cruciferous vegetables as the primary samples, after failure of discrimination of each species using current DNA barcodes, we performed the fluorescent amplified fragment length polymorphism （F-AFLP） and successfully discriminated each species, subspecies, variety and their cultivar in 74 samples. Then the non-qualitative results obtained from F-AFLP were transformed into two-dimensional barcodes image file of each cultivar via the PDF417 software. This method was also successfully applied to the discrimination of 17 Chinese indigenous pig breeds. The barcode we constructed which greatly reduces the information storage space is genotypes-specific, and can be conveniently decoded into the original data and thereby be conveniently shared and referred to. We believe that it is possible to construct a new data sharing molecular barcode system that could discriminate the subspecies, varieties, cultivars and even individuals with close genetic relationships.

Response of soil N_2O emissions to precipitation pulses under different nitrogen availabilities in a semiarid temperate steppe of Inner Mongolia, China

Short-term nitrous oxide（N2O） pulse emissions caused by precipitation account for a considerable portion of the annual N2O emissions and are greatly influenced by soil nitrogen（N） dynamics. However, in Chinese semiarid temperate steppes, the response of N2O emissions to the coupling changes of precipitation and soil N availability is not yet fully understood. In this study, we conducted two 7-day field experiments in a semiarid temperate typical steppe of Inner Mongolia, China, to investigate the N2O emission pulses resulting from artificial precipitation events（approximately equivalent to 10.0 mm rainfall） under four N addition levels（0, 5, 10 and 20 g N/（m2·a）） using the static opaque chamber technique. The results show that the simulated rainfall during the dry period in 2010 caused greater short-term emission bursts than that during the relatively rainy observation period in 2011（P〈0.05）. No significant increase was observed for either the N2O peak effluxes or the weekly cumulative emissions（P〉0.05） with single water addition. The peak values of N2O efflux increased with the increasing N input. Only the treatments with water and medium（WN10） or high N addition（WN20） significantly increased the cumulative N2O emissions（P〈0.01） in both experimental periods. Under drought condition, the variations in soil N2O effluxes were positively correlated with the soil NH4-N concentrations in the three N input treatments（WN5, WN10, and WN20）. Besides, the soil moisture and temperature also greatly influenced the N2O pulse emissions, particularly the N2O pulse under the relatively rainy soil condition or in the treatments without N addition（ZN and ZWN）. The responses of the plant metabolism to the varying precipitation distribution and the length of drought period prior to rainfall could greatly affect the soil N dynamics and N2O emission pulses in semiarid grasslands.

The aliphatic hydrocarbon distributions of terrestrial plants around an alpine lake： a pilot study from Lake Ximencuo, Eastern Qinghai-Tibet Plateau

As part of an investigation of the sources of aliphatic hydrocarbons to the sediments of alpine Lake Ximencuo, leaves of the eight dominant vascular plants were collected and their hydrocarbon contents were analyzed. A series of unsaturated aliphatic hydrocarbons were identified in the plant leaves; in particular, Festuca sp. contain a series of n-alkadienes that have rarely been reported in previous studies. The comparison of n-alkane proxies （ACL27 33, ACLT, Paq, and CPI） and δ13Corg among plant leaves, surface soils, and lake sediments suggests that organic proxies have been altered to varying degrees during the transport and burial process of organic materials. It is believed that microbial reworking and source changes have great impacts on organic proxies in the alpine lake system. In addition, the cluster analysis for plant leaves depending on n-alkane compositions and the ACLT proxy generates similar results. Accordingly, we postulate that the average chain length of plant waxes might be a potential indicator of plant classification in regions such as the Qinghai-Tibet Plateau.

Grain-size reduction of feldspar and flow of deformed granites within the Gaoligong shear zone, southwestern Yunnan, China

Mineral deformation and rock flow mechanism in the lithosphere are related to the rheological behavior and weakening mechanism of the continent. Natural deformation behaviors of feldspars are not well understood due to the complexity of their mineral compositions, crystal structures, as well as changing deformation conditions. The refined microstructure,fabric and composition of major minerals in the deformed granitic rocks within the Gaoligong shear zone(GLGSZ), southwestern Yunnan, China, were studied. With increasing mylonitization, two fabric types of end-members have been distinguished(type-I banded granitic mylonite and type-II banded ultramylonite). The two types of deformed granitic rocks have the same mineral assemblage, but different mineral modes. The type-I banded granitic mylonite has a greater proportion of K-feldspar(mostly present as porphyroclasts)>plagioclase>quartz±biotite, however, the type-II banded ultramylonite has a greater proportion of fine-grained plagioclase>K-feldspar>quartz±biotite. The crystallographic preferred orientation(CPO) patterns of quartz combined with two-feldspar geothermometer, confirm that the quartz grains in the type-I and type-II granitic rock have undergone high-temperature dislocation creep deformation. The K-feldspar grains in the matrix of type-II banded ultramylonite show a dominant(100)[010] slip system with dislocation creep recrystallization, while the fine-grained plagioclase grains present a weak CPO pattern with superplastic flow. The K-feldspar porphyroclasts show grain-size reduction associated with mineral composition and fabric transformation. The myrmekite formation with the fine-grained neocrystallization of plagioclase and quartz significally replaced the K-feldspar porphyroclasts. Finally, the fine-grained neocrystallization plagioclases were formed further into the high strain localized ultramylonites with superplastic flow.

MXene Hybridized Polymer with Enhanced Electromagnetic Energy Harvest for Sensitized Microwave Actuation and Self-Powered Motion Sensing

Polymeric microwave actuators combining tissue-like softness with programmable microwave-responsive deformation hold great promise for mobile intelligent devices and bionic soft robots. However, their application is challenged by restricted electromagnetic sensitivity and intricate sensing coupling. In this study, a sensitized polymeric microwave actuator is fabricated by hybridizing a liquid crystal polymer with Ti3C2Tx (MXene). Compared to the initial counterpart, the hybrid polymer exhibits unique space-charge polarization and interfacial polarization, resulting in significant improvements of 230% in the dielectric loss factor and 830% in the apparent efficiency of electromagnetic energy harvest. The sensitized microwave actuation demonstrates as the shortened response time of nearly 10 s, which is merely 13% of that for the initial shape memory polymer. Moreover, the ultra-low content of MXene (up to 0.15 wt%) benefits for maintaining the actuation potential of the hybrid polymer. An innovative self-powered sensing prototype that combines driving and piezoelectric polymers is developed, which generates real-time electric potential feedback (open-circuit potential of similar to 3 mV) during actuation. The polarization-dominant energy conversion mechanism observed in the MXene-polymer hybrid structure furnishes a new approach for developing efficient electromagnetic dissipative structures and shows potential for advancing polymeric electromagnetic intelligent devices.

Spatial distribution of ion polytropic index joint-modulated by temperature anisotropy and MHD disturbances in the southern high latitude magnetosheath

Using the Cluster data from 2001 to 2010, we studied spatial distribution of effective ion polytropic index in the southern high latitude magnetosheath, and joint-modulation of ion polytropic index by temperature anisotropy and MHD disturbances. The magnetosheath ions generally experience various polytropic processes with different polytropic index. The median polytropic indexes of magnetosheath ions in the GSE X-Y plane decrease toward the bow shock. Near the magnetopause, the median polytropic indexes are basically between isothermal and adiabatic except in the duskside flank close to the terminator. The analysis of correlation coefficient of perturbed ion number density with parallel magnetic field CC_δnδB_‖ and ion temperature anisotropy parameter A_T, indicates that the dominant MHD disturbance near magnetopause is slow mode with larger ion temperature anisotropy, and there are various modes of MHD disturbances with insignificant ion temperature anisotropy near the bow *** polytropic index modulated by slow mode disturbances is generally larger than that modulated by fast mode disturbances, and the larger ion temperature anisotropy, the larger polytropic index. The median polytropic indexes modulated jointly by slow mode disturbances and the strong ion temperature anisotropy can be larger than 1.0, while those modulated by fast mode disturbances and weak temperature anisotropy can be even possibly close to zero. Moreover, because of pronounced dusk-favored asymmetry of ion temperature anisotropy, the median polytropic index in the dawnside flank of the magnetosheath near the terminator is smaller than that in the duskside flank of the magnetosheath. The good correspondence between the distributions of median polytropic indexes and ion temperature anisotropy and MHD disturbances indicates that the ion temperature anisotropy and MHD disturbances determine the distribution of the polytropic index in the magnetosheath.

Catalytic steam reforming of biomass over Ni-based catalysts: Conversion from poplar leaves to hydrogen-rich syngas

Hepatoprotective and antioxidant effects of dietary Glycyrrhiza polysaccharide against TCDD-induced hepatic injury and RT-PCR quantification of AHR2,ARNT2,CYPIA mRNA in jian Carp(Cyprinus carpio ***)

To evaluate the protective effects of Glycyrrhiza polysaccharide（GPS） against 2,3,7,8-tetrachlorodibenzo-p-dioxin（TCDD）-induced hepatotoxicity in jian carp,the fish were fed diets containing GPS at doses of 0.1,0.5 and 1.0 g/kg for 60 days before an intraperitoneal injection of 0.6 μg/kg TCDD at a volume of 0.05 mL/10 g body *** 72 hr post-injection,blood and liver samples were taken for biochemical analysis and the fish liver samples were used for the preparation of pathological *** results showed that increases in alanine aminotransferase（GPT）,aspartate aminotransferase（GOT）,lactate dehydrogenase（LDH）,and alkaline phosphatase（AKP） in serum induced by TCDD were significantly inhibited by pre-treatment with 1.0 g/kg *** the 1.0 g/kg GPS pre-treatment,total protein（TP）,albumin（Alb）,catalase（CAT）,glutathione peroxidase（GPx）,total antioxidant capacity（T-AOC） and superoxide dismutase（SOD） activities in liver tissue increased significantly,malondialdehyde（MDA） formation（P ＆lt; 0.05 or P ＆lt; 0.01） was significantly inhibited,and the expression of cytochrome P4501A（CYP1A）,aryl hydrocarbon receptor 2（AHR2） and aryl hydrocarbon receptor nuclear translocator 2（ARNT2） mRNA（P ＆lt; 0.05） was significantly *** observations on fish liver were obtained by preparing paraffin tissue sections via HE staining,and the results showed that histological changes were obviously reduced by 0.5 and 1.0 g/kg *** significantly reduced liver tissue damage caused by ***,these results proved the hepatoprotective effect of GPS in protecting against fish liver injury induced by TCDD,and supported the use of GPS（1.0 g/kg） as a hepatoprotective and antioxidant agent in fish.
