Antiviral drug design based on structural insights into the N-terminal domain and C-terminal domain of the SArS-CoV-2 nucleocapsid protein

Nucleocapsid(N) protein plays crucial roles in the life cycle of severe acute resp.r.tory syndrome coronavirus 2(SArS-CoV-2), including the formation of ribonucleoprotein(rNp) complex with the viral *** we rep.r.ed the crystal structures of the N-terminal domain(NTD) and C-terminal domain(CTD) of the N protein and an NTD-rNA complex. Our structures reveal a unique tetramer organization of NTD and identify a distinct rNA binding mode in the NTD-rNA complex, which could contribute to the formation of the rNp complex. We also screened small molecule inhibitors of N-NTD and N-CTD and discovered that ceftriaxone sodium, an antibiotic, can block the binding of rNA to NTD and inhibit the formation of the rNp complex. These results together could facilitate the further research of antiviral drug design targeting N protein.

discovery of pulsars is one of the main goals for large radio *** Five-hundredmeter Ap.r.ure Spherical radio Telescope(FAST),that incorp.r.tes an L-band 19-beam receiver with a system temp.r.ture of about 20 K,is the most sensitive radio telescope utilized for discovering *** designed the snapshot observation mode for a FAST key science project,the Galactic plane pulsar Snapshot(GppS)survey,in which every four nearby pointings can observe a cover of a sky patch of 0.1575 square degrees through beam-switching of the L-band 19-beam *** integration time for each pointing is 300 seconds so that the GppS observations for a cover can be made in 21 *** goal of the GppS survey is to discover pulsars within the Galactic latitude of±10∘from the Galactic plane,and the highest priority is given to the inner Galaxy within±5∘.Up to now,the GppS survey has discovered 201 pulsars,including currently the faintest pulsars which cannot be detected by other telescopes,pulsars with extremely high disp.r.ion measures(DMs)which challenge the currently widely used models for the Galactic electron density distribution,pulsars coincident with sup.r.ova remnants,40 millisecond pulsars,16 binary pulsars,some nulling and mode-changing pulsars and rotating radio transients(rrATs).The follow-up observations for confirmation of new pulsars have polarization-signals recorded for polarization profiles of the ***-detection of previously known pulsars in the survey data also leads to significant improvements in p.r.meters for 64 *** GppS survey discoveries are published and will be updated at http://***/GppS/.

Background::Coronavirus disease 2019(COVID-19),which is caused by severe acute resp.r.tory syndrome coronavirus 2(SArS-CoV-2),has rapidly spread throughout the *** this study,we aimed to identify the risk factors for severe COVID-19 to improve treatment ***::A multicenter,cross-sectional study was conducted on 313 patients hospitalized with *** were classified into two groups based on disease severity(nonsevere and severe)according to initial clinical *** test results and epidemiological and clinical characteristics were analyzed using descriptive *** and multivariate logistic regression models were used to detect potential risk factors associated with severe ***::A total of 289 patients(197 nonsevere and 92 severe cases)with a median age of 45.0(33.0,61.0)years were included in this study,and 53.3%(154/289)were ***(192/286,67.1%)and cough(170/289,58.8%)were commonly observed,followed by sore throat(49/289,17.0%).Multivariate logistic regression analysis suggested that patients who were aged≥65 years(Or:2.725,95%confidence interval[CI]:1.317-5.636;p=0.007),were male(Or:1.878,95%CI:1.002-3.520,p=0.049),had comorbid diabetes(Or:3.314,95%CI:1.126-9.758,p=0.030),cough(Or:3.427,95%CI:1.752-6.706,pr diarrhea(Or:2.629,95%CI:1.109-6.231,p=0.028)on admission had a higher risk of severe ***,stratification analysis indicated that male patients with diabetes were more likely to have severe COVID-19(71.4%vs.28.6%,χ2=8.183,p=0.004).Conclusions::The clinical characteristics of those with severe and nonsevere COVID-19 were significantly *** elderly,male patients with COVID-19,diabetes,and presenting with cough and/or diarrhea on admission may require close monitoring to prevent deterioration.

We present the observational results from a detailed timing analysis of the black hole candidate EXO 1846-031 during its outburst in 2019 with the observations of Insight-HXMT,NICEr and *** outburst can be classified roughly into four different ***-C quasi-p.r.odic oscillations(QpOs)observed by NICEr(about 0.1-6 Hz)and Insight-HXMT(about 0.7-8 Hz)are also rep.r.ed in this ***,we study various physical quantities related to QpO *** QpO rms-frequency relationship in the energy band 1-10 keV indicates that there is a turning pointing in frequency around2 Hz,which is similar to that of GrS 1915+105.A possible hypothesis for the relationship above may be related to the inclination of the source,which may require a high inclination to explain *** relationships between QpO frequency and QpO rms,hardness,total fractional rms and count rate have also been found in other transient sources,which can indicate that the origin of type-C QpOs is non-thermal.

EVALUATION OF EFFICACY AND SAFETY OF diACErEIN IN KNEE OSTEOArTHrITIS IN CHINESE pATIENTS

Objective To evaluate the efficacy and safety of diacerein in patients with knee osteoarthritis (OA). Methods A total of 223 patients satisfying the American College of rheumatology criteria for knee OA were chosen for this 17-week, randomized, double-dummy, diclofenac sodium-controlled trial, with diacerein dosage of 100 mg/d and diclofenac sodium of 75mg/d. Efficacy and safety of both drugs were evaluated. results Totally 106 patients in the diacerein group and 107 patients in the diclofenac group were considered qualified for the evaluation. After 12 weeks of treatment, the total effective rates of patients/physicians’ overall assessment in diacerein and diclofenac groups were 65.4%/61.6% and 61.2%/61.2%, respectively (p>0.05). The primary efficacy p.r.meter [visual analog scale (VAS) assessment of pain on 20 metres walking] and the secondary efficacy p.r.meters [tenderness on palpation, Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC), and 36-item Short-Form (SF-36) Health Survey] significantly improved comp.r.d with baseline in both groups (pp p.r.od, there were no obvious changes in above p.r.meters in diacerein group. However, in diclofenac group, pain on 20 metres walking, tenderness on palpation, and WOMAC became aggravated after withdrawing the drug for 4 weeks (preover, the consumption of p.r.cetamol was significantly lower in diacerein group than in diclofenac group during follow-up (prelated adverse events were 35.7% in diacerein and 45.1% in diclofenac group, respectively. Mild-to-moderate gastrointestinal disorders were the most frequent adverse events. Conclusions diacerein is as effective as diclofenac sodium in treating patients with knee OA. Furthermore, it has better extended effect and a good safety profile. It is generally well tolerated and has no severe adverse effect.

The full array of the Large High Altitude Air Shower Observatory(LHAASO)has been in op.r.tion since July *** its kilometer-square array(KM2A),we optimized the selection criteria for very high and ultrahigh energyγ-rays using data collected from August 2021 to August 2022,resulting in an improvement in significance of the detection in the Crab Nebula of ap.r.ximately 15%,comp.r.d with that of previous *** the implementation of these new selection criteria,the angular resolution was also significantly improved by ap.r.ximately 10%at tens of *** aspects of the full KM2A array p.r.ormance,such as the pointing error,were also calibrated using the Crab *** resulting energy spectrum of the Crab Nebula in the energy range of 10-1000 TeV are well fitted by a log-p.r.bola model,which is consistent with the previous results from LHAASO and other exp.r.ments.

Monkeypox is a zoonotic *** the first human monkeypox case was detected in 1970,it has been prevalent in some countries in central and western *** May 2022,monkeypox cases have been rep.r.ed in more than 96 non-endemic countries and regions *** of September 14,2022,there have been more than 58,200 human monkeypox cases,and there is community *** cessation of smallpox vaccination in 1980,which had some cross-protection with monkeypox,resulted in a general lack of immunity to monkeypox,which caused global concern and *** of Sep-tember 14,2022,there are four monkeypox cases in china,including three in Taiwan province and one in Hong Kong *** foreign studies have shown that children are vulnerable to monkeypox and are also at high risk for severe disease or *** order to improve pediatricians'understanding of monkeypox and achieve early detection,early diagno-sis,early treatment,and early disposal,we have organized national authoritative exp.r.s in pediatric infection,resp.r.tory,dermatology,critical care medicine,infectious diseases,and public health and others to formulate this exp.r. consensus,on the basis of the latest"Clinical management and infection prevention and control for monkeypox"released by The World Health Organization,the"guidelines for diagnosis and treatment of monkeypox(version 2022)"issued by National Health Commission of the people's republic of china and other relevant *** the development of this consensus,multidisciplinary exp.r.s have repeatedly demonstrated the etiology,epidemiology,transmission,clinical manifestations,laboratory examinations,diagnosis,differential diagnosis,treatment,discharge criteria,prevention,disposal process,and key points of prevention and control of suspected and confirmed cases.

A 48-year-old woman was admitted with 15-mo history of abdominal pain, diarrhea and hematochezia, and 5-mo history of defecation difficulty. She had been successively admitted to nine hospitals, with an initial diagnosis of inflammatory bowel disease with stenotic sigmoid colon. Findings from computed tomography virtual colonoscopy, radiography with meglumine diatrizoate, endoscopic balloon dilatation, metallic stent implantation and later overall colonoscopy, coupled with the newfound knowledge of compound Qingdai pill-taking, led to a subsequent diagnosis of ischemic or toxic bowel disease with sigmoid colon stenosis. The patient was successfully treated by lap.r.scopic sigmoid colectomy, and postop.r.tive pathological examination revealed ischemic or toxic injury of the sigmoid colon, providing a final diagnosis of druginduced sigmoid colon stenosis. This case highlights that adequate awareness of drug-induced colon stenosis has a decisive role in avoiding misdiagnosis and mistreatment. The diagnostic and therapeutic exp.r.ences learnt from this case suggest that endoscopic balloon expansion and colonic metallic stent implantation as bridge treatments were demonstrated as crucial for the differential diagnosis of benign colonic stenosis. Skillful surgical technique and ap.r.priate p.r.op.r.tive management helped to ensure the safety of our patient in subsequent surgery after long-term use of glucocorticoids.

老年髋部骨折具有高发生率、高致残率、高病死率的特点,已成为威胁老年人健康的公共卫生问题。手术是老年髋部骨折的首选治疗手段,但围术期下肢深静脉血栓(DVT)的发生率极高,一旦进展为肺栓塞将显著增加患者死亡风险。针对这一问题,临床指南和专家共识均强调对此类患者应积极采取基础预防、物理预防、药物预防的全面预防措施。在此预防体系中,基础预防是物理和药物预防的基石。目前,临床上对于基础预防措施缺乏统一明确的标准。为此,中华护理学会骨科专业委员会与中国医疗保健国际交流促进会骨科分会护理学部组织相关护理专家,结合循证医学证据和专家临床经验,制订《老年髋部骨折患者围术期下肢深静脉血栓基础预防专家共识(2024版)》,共提出10条推荐意见,以规范老年髋部骨折患者围术期下肢DVT的基础预防措施,促进患者康复。
