Background: Regulatory B cells(Bregs) is an indispensable element in inducing immune tolerance after liver transplantation. As one of the microRNAs(miRNAs), mi R-29a-3p also inhibits translation by degrading the target mRNA, and yet the relationship between Bregs and mi R-29a-3p has not yet been fully explored. This study aimed to investigate the impact of miR-29a-3p on the regulation of differentiation and immunosuppressive functions of memory Bregs(m Bregs) and ultimately provide potentially effective therapies in inducing immune tolerance after liver transplantation. Methods: Flow cytometry was employed to determine the levels of Bregs in peripheral blood mononuclear cells. TaqMan low-density array miRNA assays were used to identify the expression of different miRNAs, electroporation transfection was used to induce mi R-29a-3p overexpression and knockdown, and dual luciferase reporter assay was used to verify the target gene of miR-29a-3p. Results: In patients experiencing acute rejection after liver transplantation, the proportions and immunosuppressive function of m Bregs in the circulating blood were significantly impaired. mi R-29a-3p was found to be a regulator of m Bregs differentiation. Inhibition of miR-29a-3p, which targeted nuclear factor of activated T cells 5(NFAT5), resulted in a conspicuous boost in the differentiation and immunosuppressive function of m Bregs. The inhibition of mi R-29a-3p in CD19~+ B cells was capable of raising the expression levels of NFAT5, thereby promoting B cells to differentiate into m Bregs. In addition, the observed enhancement of differentiation and immunosuppressive function of m Bregs upon mi R-29a-3p inhibition was abolished by the knockdown of NFAT5 in B cells. Conclusions: mi R-29a-3p was found to be a crucial regulator for m Bregs differentiation and immunosuppressive function. Silencing mi R-29a-3p could be a potentially effective therapeutic strategy for inducing immune tolerance after liver transplantation.

Reinforced concrete(RC)slabs are the primary load-carrying member of underwater *** can suffer severe local failures such as cratering,spalling,or breaching as a result of underwater close-in(UWCI)*** this study,we established a fully validated high-fidelity finite element analysis approach to precisely reproduce the local failures of RC slabs after a UWCI explosion.A recently proposed dynamic constitutive model is used to describe wet *** effects of free water content on the material properties,including the tensile/compressive strength,elastic modulus,strain rate effect,failure strength surface,and equation of state,are comprehensively calibrated based on existing test *** calibrated material parameters are then verified by a single-element test.A high-fidelity finite element analysis(FEA)approach of an RC slab subjected to a UWCI explosion is established using an arbitrary Lagrangian-Eulerian(ALE)*** previous UWCI explosion tests on RC orifice targets and underwater contact explosion tests on saturated concrete slabs showed that the established FEA approach could accurately reproduce the pressure-time history in water and damage patterns,including the cracking,cratering,and spalling,of the RC orifice target and saturated concrete ***,parametric studies conducted by simulating an RC slab subjected to a UWCI explosion showed that:(i)the local failure of an RC slab enlarges with increased charge weight,reduced standoff distance,and reduced structural thickness;(ii)compared to a water-backed RC slab,an air-backed RC slab exhibits much more obvious local and structural ***,to aid the anti-explosion design of relevant underwater facilities,based on over 90 simulation cases empirical formulae are summarized to predict local failure modes,i.e.,no spall,spall,and breach,of water-and air-backed RC slabs subjected to a UWCI explosion.

Feature Selection(FS)is an important data management technique that aims to minimize redundant information in a *** work proposes DENGO,an improved version of the Northern Goshawk Optimization(NGO),to address the FS *** NGO is an efficient swarm-based algorithm that takes its inspiration from the predatory actions of the northern *** order to overcome the disadvantages that NGO is prone to local optimum trap,slow convergence speed and low convergence accuracy,two strategies are introduced in the original NGO to boost the effectiveness of ***,a learning strategy is proposed where search members learn by learning from the information gaps of other members of the population to enhance the algorithm's global search ability while improving the population ***,a hybrid differential strategy is proposed to improve the capability of the algorithm to escape from the trap of the local optimum by perturbing the individuals to improve convergence accuracy and *** prove the effectiveness of the suggested DENGO,it is measured against eleven advanced algorithms on the CEC2015 and CEC2017 benchmark functions,and the obtained results demonstrate that the DENGO has a stronger global exploration capability with higher convergence performance and ***,the proposed DENGO is used for FS,and the 29 benchmark datasets from the UCL database prove that the DENGO-based FS method equipped with higher classification accuracy and stability compared with eight other popular FS methods,and therefore,DENGO is considered to be one of the most prospective FS ***'s code can be obtained at https://***/matlabcentral/fileexchange/158811-project1.

Brassinosteroids(BRs)are a class of phytohormones that regulate plant growth and *** previous studies,we cloned and identified PROTEIN PHOSPHATASE WITH KELCH-liKE1(OsPPKL1)as the causal gene for the quantitative trait locus GRAIN LENGTH3(qGL3)in rice(Oryza sativa).We also showed that qGL3/OsPPKL1 is mainly located in the cytoplasm and nucleus and negatively regulates BR signaling and grain *** qGL3 is a negative regulator of BR signaling,its turnover is critical for rapid response to changes in ***,we demonstrate that qGL3 interacts with the WD40-domain-containing protein WD40-REPEAT PROTEIN48(OsWDR48),which contains a nuclear export signal(NES).The NES signal is crucial for the cytosolic localization of OsWDR48 and also functions in the self-turnover of *** show that OsWDR48 physically interacts with and genetically acts through qGL3 to modulate BR ***,qGL3 may indirectly promote the phosphorylation of OsWDR48 at the Ser-379 and Ser-386 *** of both phosphorylation sites in OsWDR48 to non-phosphorylatable alanine enhanced the strength of the OsWDR48-qGL3 ***,we found that brassinolide can promote the accumulation of non-phosphorylated OsWDR48,leading to strong interaction intensity between qGL3 and *** together,our results show that OsWDR48 facilitates qGL3 retention and induces degradation of qGL3 in the *** findings suggest that qGL3 self-modulates its turnover by binding to OsWDR48 to regulate its cytoplasmic localization and stability,leading to efficient orchestration of BR signal transduction in rice.

liver fibrosis is a dynamic wound-healing response characterized by the agglutination of the extracellular matrix(ECM).Si-wu-Tang(SWT),a traditional Chinese medicine(TCM)formula,is known for treating gynecological diseases and liver *** previous studies demonstrated that long non-coding RNA H19(H19)was markedly upregulated in fibrotic livers while its deficiency markedly reversed ***,the mechanisms by which SWT influences H19 remain ***,we established a bile duct ligation(BDL)-induced liver fibrosis model to evaluate the hepatoprotective effects of SWT on various cells in the *** results showed that SWT markedly improved ECM deposition and bile duct reactions in the ***,SWT relieved liver fibrosis by regulating the transcription of genes involved in the cytoskeleton remodeling,primarily in hepatic stellate cells(HSCs),and influencing cytoskeleton-related angiogenesis and hepatocellular *** modulation collectively led to reduced ECM *** extensive bioinformatics analyses,we determined that H19 acted as a miRNA sponge and mainly inhibited miR-200,miR-211,and let7b,thereby regulating the above cellular regulatory ***,SWT reversed H19-related miRNAs and signaling pathways,diminishing ECM deposition and liver ***,these protective effects of SWT were diminished with the overexpression of H19 in *** conclusion,our study elucidates the underlying mechanisms of SWT from the perspective of H19-related signal networks and proposes a potential SWT-based therapeutic strategy for the treatment of liver fibrosis.

Photocatalytic aerobic oxidation reactions are largely governed by the efficiency of charge separation and subsequent reactive oxygen species(ROS) generation. Herein, we report a polarization engineering strategy to promote the charge separation and ROS generation efficiency by substituting the benzene unit with furan/thiophene in porous organic polymers(POPs). Benefiting from the extent of local polarization, the thiophene-containing POP(JNU-218) exhibits the best photocatalytic performance in aerobic oxidation reactions, with a yield much higher than those for the furan-containing POP(JNU-217) and the benzenecontaining POP(JNU-216). Experimental studies and theoretical calculations reveal that the increase of local polarization can indeed reduce the exciton binding energy, and therefore facilitate the separation of electron-hole pairs. This work demonstrates a viable strategy to tune charge separation and ROS generation efficiency by modulating the dipole moments of the building blocks in porous polymeric organic semiconductors.
