The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by china,fostered through extensive collaboration with international *** complex comprises four accelerators:a 30 GeV linac,a 1.1 GeV Damping Ring,a booster capable of achieving energies up to 180 GeV,and a Collider operating at varying energy modes(Z,W,H,and tt).The linac and Damping Ring are situated on the surface,while the subterranean booster and Collider are housed in a 100 km circumference underground tunnel,strategically accommodating future expansion with provisions for a potential super Proton Proton Collider(SPPC).The CEPC primarily serves as a Higgs *** its baseline design with synchrotron radiation(SR)power of 30 MW per beam,it can achieve a luminosity of 5×10^(34)cm^(-2)s^(-1)per interaction point(IP),resulting in an integrated luminosity of 13 ab^(-1)for two IPs over a decade,producing 2.6 million Higgs *** the SR power to 50 MW per beam expands the CEPC's capability to generate 4.3 million Higgs bosons,facilitating precise measurements of Higgs coupling at sub-percent levels,exceeding the precision expected from the HL-LHC by an order of *** Technical Design Report(TDR)follows the Preliminary Conceptual Design Report(Pre-CDR,2015)and the Conceptual Design Report(CDR,2018),comprehensively detailing the machine's layout,performance metrics,physical design and analysis,technical systems design,R&D and prototyping efforts,and associated civil engineering ***,it includes a cost estimate and a preliminary construction timeline,establishing a framework for forthcoming engineering design phase and site selection *** is anticipated to begin around 2027-2028,pending government approval,with an estimated duration of 8 *** commencement of experiments and data collection could potentially be initiated in the mid-2030s.

一个科学、规范、具有可行性的试验设计是获得可靠数据与保障临床试验质量的基础。本文通过收集和整理一家药物临床试验机构承接的注册类中药品种临床试验资料,归纳分析试验设计中存在的问题,将纳入分析的18项中药临床试验审查中发现的问题汇总分类为不符合法规要求、方案内容描述不清晰、试验方案和病例报告表等试验文件设计的可行性差、试验操作要求在不同研究文件或同一文件前后不一致、行文错误、其他等6类。针对以上问题,探讨申办者、研究者、临床试验机构三方为保障设计质量可以采取的措施,以期为提高临床试验质量提供参考。

Recent studies have shown that radiofrequency(RF) ablation therapy is a safe, feasible, and effective procedure for hepatic hemangiomas, even huge hepatic hemangiomas. RF ablation has the following advantages in the treatment of hepatic hemangiomas: minimal invasiveness, definite efficacy, high safety, fast recovery, relatively simple operation, and wide applicability. It is necessary to formulate a widely accepted consensus among the experts in china who have extensive expertise and experience in the treatment of hepatic hemangiomas using RF ablation, which is important to standardize the application of RF ablation for the management of hepatic hemangiomas, regarding the selection of patients with suitable indications to receive RF ablation treatment, the technical details of the techniques, therapeutic effect evaluations, management of complications, etc. A final consensus by a Chinese panel of experts who have the expertise of using RF ablation to treat hepatic hemangiomas was reached by means of literature review, comprehensive discussion, and draft approval.

AIM: To explore the effects of siRNA silencing of PIK3CA on proliferation, migration and invasion of gastric cancer cells and to investigate the underlying mechanisms. METHODS: The mutation of PIK3CA in exons 9 and 20 of gastric cancer cell lines HGC-27, SGC-7901, BGC-823, MGC-803 and MKN-45 was screened by polymerase chain reaction (PCR) followed by sequencing. BGC-823 cells harboring no mutations in either of the exons, and HGC-27 cells containing PIK3CA mutations were employed in the current study. siRNA targeting PIK3CA was chemically synthesized and was transfect- ed into these two cell lines in vitro . mRNA and protein expression of PIK3CA were detected by real-time PCR and Western blotting, respectively. We also measured phosphorylation of a serine/threonine protein kinase (Akt) using Western blotting. The proliferation, migration and invasion of these cells were examined separately by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltet-razolium bromide (MTT), wound healing and Transwell chambers assay. REsuLTS: The siRNA directed against PIK3CA effectively led to inhibition of both endogenous mRNA and protein expression of PIK3CA, and thus significantly down-regulated phosphorylation of Akt (P 3CA resulted in an obvious reduction in tumor cell proliferation activity, migration and invasion potential (P 27 cells exhibited stronger invasion ability than that shown by wild-type BGC-823 cells. Knockdown of PIK3CA in mutant HGC-27 cells contributed to a reduction in cell invasion to a greater extent than in non-mutant BGC-823 cells. CONCluSION: siRNA mediated targeting of PIK3CA may specifically knockdown the expression of PIK3CA in gastric cancer cells, providing a potential implication for therapy of gastric cancer.

Mammalian target of rapamycin(m TOR) inhibitor as an attractive drug target with promising antitumor effects has been widely investigated. High quality clinical trial has been conducted in liver transplant(LT) recipients in Western countries. However, the pertinent studies in Eastern world are paucity. Therefore, we designed a clinical trial to test whether sirolimus can improve recurrence-free survival(RFS) in hepatocellular carcinoma(HCC) patients beyond the Milan criteria after LT. This is an open-labeled, single-arm, prospective, multicenter, and real-world study aiming to evaluate the clinical outcomes of early switch to sirolimus-based regimens in HCC patients after LT. Patients with a histologically proven HCC and beyond the Milan criteria will be enrolled. The initial immunosuppressant regimens are center-specifc for the frst 4-6 weeks. The following regimens integrated sirolimus into the regimens as a combination therapy with reduced calcineurin inhibitors based on the condition of patients and centers. The study is planned for 4 years in total with a 2-year enrollment period and a 2-year follow-up. We predict that sirolimus conversion regimen will provide survival benefts for patients particular in the key indicator RFS as well as better quality of life. If the trial is conducted successfully, we will have a continued monitoring over a longer follow-up time to estimate indicator of overall survival. We hope that the outcome will provide better evidence for clinical decision-making and revising treatment guidelines based on Chinese population data.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

采前喷施茉莉酸甲酯对火龙果采后品质和生理特性的影响

【目的】分析采前喷施茉莉酸甲酯(MeJA)对火龙果采收品质、耐贮性及生理特性的影响,为生产上应用MeJA提升火龙果采收品质和耐贮性提供技术支持。【方法】以紫红龙火龙果为试验材料,采用5种质量浓度(0、50、100、200和400μmol/L)的MeJA采前8 d喷施火龙果,调查采收时产量、单果重、一级果率、商品果率和病果率,观察比较采后腐烂时间和腐烂率变化,测定对照(0μmol/L)和最佳处理贮藏过程中品质和生理特性的变化。【结果】采前喷施MeJA对火龙果公顷产量和单果重无显著影响(P>0.05),但采前喷施100μmol/LMeJA处理能明显提升火龙果采收时的商品性,其一级果率和商品果率分别较对照显著提高52.37%和104.38%(P3-葡聚糖酶(Glu)、几丁质酶(CHT)和苯丙氨酸解氨酶(PAL)活性,酶活性最大差值发生在果实开始腐烂前后,PPO、POD和CHT活性于贮藏第6 d差值最大,其酶活性分别较对照提高33.13%、140.38%和207.16%,PAL和Glu活性于贮藏第9 d差值最大,其酶活性分别较对照提高83.38%和25.29%。【结论】采前喷施适宜浓度的MeJA可减少生育期火龙果的感病率,提升商品果率,维持贮藏品质,提高贮藏期间的抗氧化能力和抗病能力,从而延缓果实衰老和控制贮藏期间腐烂的发生,延长贮藏期,以100μmol/L的处理效果最佳。

对北京平原区南部大兴榆垡镇420 m深的PGZ01孔岩心进行了岩石地层、磁性地层、14C测年、介形类组合和孢粉组合研究。结果表明:(1)Matuyama(松山)/Gauss(高斯)和Brunhes(布容)/Matuyama(松山)极性界线分别位于280 m和78.35 m处,钻孔底部已经进入Gauss正极性带;取280 m处Matuyama/Gauss极性界线作为第四系下限,显示可能的永定河泥石流—冲积扇在第四纪之前即到达了北京平原南部,但是华北平原上新统标志岩石地层单元——厚层棕红色黏土在Olduvai(奥尔都维)极性亚带之上即已出现。(2)结合AMS14C测年,尝试将约60 m以上地层的氧化—还原状况,与对古季风影响反映灵敏的沿海平原钻孔同样深度地层进行对比,继而确定上更新统和全新统的底界深度分别位于56.60 m和18.95 m。(3)MIS5(海洋同位素阶段)以来的特征沉积物,与沿海平原钻孔地层有很好的可比性,同时也有8~4 ka期间史前大洪水的影响。本研究为廊(坊)固(安)凹陷西北边缘区第四纪地层划分对比、古地理环境演化、活动断裂以及区域差异构造沉降研究,提供了重要的年代地层格架,对理解晚更新世以来古季风变化的环境效应有重要的启迪。
