Integrating well logs, 3D seismic, and earthquake data for comprehensive prediction of 3D in-situ stress orientations: A case study from the Weiyuan area in the Sichuan Basin, China

Determining the orientation of in-situ stresses is crucial for various geoscience and engineering applications. Conventional methods for estimating these stress orientations often depend on focal mechanism solutions(FMSs) derived from earthquake data and formation micro-imager(FMI) data from well ***, these techniques can be costly, depth-inaccurate, and may lack spatial coverage. To address this issue, we introduce the use of three-dimensional(3D) seismic data(active sources) as a lateral constraint to approximate the 3D stress orientation field. Recognizing that both stress and fracture patterns are closely related to seismic velocity anisotropy, we derive the orientation of azimuthal anisotropy from multi-azimuth 3D seismic data to compensate for the lack of spatial stress orientation information. We apply our proposed workflow to a case study in the Weiyuan area of the Sichuan Basin, China, a region targeted for shale gas production. By integrating diverse datasets, including 3D seismic, earthquakes, and well logs, we develop a comprehensive 3D model of in-situ stress(orientations and magnitudes). Our results demonstrate that the estimated anisotropy orientations from 3D seismic data are consistent with the direction of maximum horizontal principal stress(SHmax) obtained from FMIs. We analyzed 12earthquakes(magnitude > 3) recorded between 2016 and 2020 for their FMSs and compressional axis(Paxis) orientations. The derived SHmax direction from our 3D stress model is 110°ES(East-South), which shows excellent agreement with the FMSs(within 3.96°). This close alignment validates the reliability and precision of our integrated method for predicting 3D SHmax orientations.

Alpine plants possess unique traits to adapt alpine *** leaf trait relationships of alpine plants can be captured by the two trait dimensions of organ size and resource economics is *** hypothesized that,beyond the trait dimensions of leaf size and resource economics,nonstructured carbohydrates(NSC)would reflect a dimension of cold-tolerance in alpine *** test this hypothesis,we measured 12 leaf traits critical to leaf construction and growth in 143 species across 7 sites ranging from alpine steppes to alpine meadows along an environmental gradient on the Tibetan ***,a cold resistance experiment was conducted at one of these sites to estimate the lethal temperature causing 50%frost damage(LT_(50))of 11 alpine *** majority of variations in 12 leaf traits of alpine plants were captured by three trait axes,in which leaf carbon(LCC)and NSC(including leaf starch;LSC and leaf soluble sugars;LSS)were clustered in a new dimension(PC3)beyond leaf size and structure,and resource *** LCC,LSC and LSS all showed negative correlations with mean annual temperature,a significant negative correlation was only found between LSS and LT_(50).It indicated that PC3 was able to reflect the cold-tolerance of alpine plants to some extent,in which LSS was the most critical *** storage and transformation of NSC under stressful conditions could reflect a dimension of long-term metabolic adaptation and cold-tolerance,which is an extension of the resource-utilization strategy beyond construction cost and growth.

Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

Single-dose daclizumab induction therapy in patients with liver transplantation

AIM: To investigate the efficacy and safety of a singledose daclizumab induction therapy in orthotopic liver transplantation (OLTx).METHODS: A retrospective study was made for 54 cases of OLTx in recent three years. The daclizumab group consisted of 23 cases of OLTx who received single-dose of 2 mg/kg intravenously after postoperative 24 hours. The control group consisted of the remaining 31 patients. Additional immunosuppressors included steroids, mycomphenolate mofetil, facrolimus or microemulsion cyclosporine used in all patients. Meta-statistical analysis was made for general data,incidence of acute rejection and infection, postoperative clinical course, complications and prognosis between two ***: Pretransplant demographies were not significantly different between two groups. In the induction group there were significantly less acute rejection episodes (5 of 23, 21.74 %) than those in the control group (12 of 31, 38.71%), which were proved by pathologic diagnosis (P＜0.05). The incidence of infection at the early stage was not significantly different between two ***: Induction therapy with single-dose of daclizumab is safe and effective and appears to be able to reduce the incidence of acute rejection.

Lamivudine prophylaxis of liver allograft HBV reinfection in HBV related cirrhotic patients after liver transplantation

BACKGROUND: Liver allograft hepatitis B virus (HBV) reinfection and hepatitis B (HB) recurrence jeopardize the long-term survival of recipient and liver allograft. Lamivu- dine has been referred as a novel antiviral agent against HBV in HBV cirrhotic patients even in liver transplantation setting. We assessed the prophylatic effect of lamivudine on liver allograft HBV reinfection and clarified the dynamic changes of HBV markers in HBV related decompensated liver cirrhosis after liver transplantation. METHODS: Twenty-five recipients were divided into three groups: HBV active replication group (15 recipients), HBV inactive replication group (7), and control group (3). 100 mg/d lamivudine was administered preoperatively except in the control group. The HBV markers of serial sera and liver biopsy samples of the 25 recipients were evaluated re- gularly with enzyme-linked radioimmunoassay, HBV DNA fluorecent quantitative assay, immunohistochemical stain- ing , labelled streptavidin biotin ( LSAB) and digoxin la- belled HBV DNA hybridization in situ. The dynamic alter- nation of HBV markers under lamivudine prophylaxis was observed. RESULTS: In the HBV active replication group who had received lamivudine 2 weeks before liver transplantation, serum HBV DNA positive converted to negative by 80%. HBsAg of all recipients disappeared after liver transplanta- tion , but corresponding antibodies of HBV appeared within one week after the operation. HBsAb 9/15, HBcAb 13/15 and HBeAb 11/15 appeared and subsided gradually within 24 weeks. HBV DNA in sera was kept negative; HBsAg, HBcAg and HBV DNA hybridization in situ of liver biopsy samples remained negative after use of lamivudine. Ten of the 15 recipients showed clearance of HBV, and per se HBV markers were undetectable both in serum and liver bi- opsy samples between 12 to 44 weeks (24 weeks on ave- rage). Clinical study on safety of adult-to-adult living donor liver transplantation in both donors and recipients

AIM： TO investigate the safety of adult-to-adult living donor liver transplantation （A-A LDLT） in both donors and recipients. METHODS： From January 2002 to July 2006, 50 cases of A-A LDLT were performed at West China Hospital, Sichuan University, consisting of 47 cases using right lobe graft without middle hepatic vein （HHV）, and 3 cases using dual grafts （one case using two left lobe, 2 using one right lobe and one left lobe）. The most common diagnoses were hepatitis B liver cirrosis, 30 （60%） cases; and hepatocellular carcinoma, 15 （30%） cases in adult recipients. Among them, 10 cases had the model of end-stage liver disease （HELD） with a score of more than 25. Donor screening consisted of reconstruction of the hepatic blood vessels and biliary system with 3-dimension computed tomography and volumetry of whole liver and right liver volume. Various improved surgical techniques were adopted in the procedures for both donors and recipients. RESULTS： Forty-nine right lobes and 3 left lobes （2 left lobe grafts for 1 recipient, 1 left lobe graft for 1 recipient who had received right lobe graft donated by relative living donor） were obtained from 52 living donors. The 49 right lobe grafts, without HHV, weighed 400 g-850 g （media 550 g）, and the ratio of graft volume to recipient standard liver volume （GV/SLV） ranged from 31.74% to 71.68% （mean 45.35%）. All donors＇ remnant liver volume was over 35% of the whole liver volume. There was no donor mortality. With a follow- up of 2-52 mo （media 9 too）, among 50 adult recipients, complications occurred in 13 （26%） cases and 4 （8%） died postoperatively within 3 mo. Their 1-year actual survival rate was 92%.CONCLUSION： When preoperative CT volumetry shows volume of remnant liver is more than 350, the ratio of right lobe graft to recipients standard liver volume exceeding 40%, A-A LDLT using right lobe graft without MHV should be a very safe procedure for both donors and recipients, otherwise dual grafts liver transplantation
