Graphitic carbon nitride(g-C_(3)N_(4))nanosheets have attracted widespread interest in the construction of advanced separation ***,dense stacking and a single functionality have limited the membrane ***,an advanced two-/three-dimensional(2D/3D)g-C_(3)N_(4)/TiO_(2)@MnO_(2) membrane is constructed by intercalating 3D TiO_(2)@MnO_(2) nanostructures into g-C_(3)N_(4) *** 3D flower-like nanostructures broaden the transport channels of the composite *** membrane can effectively separate five oil-in-water(O/W)emulsions,with a maximum flux of 3265.67±15.01 L·m^(-2)·h^(-1)·bar^(-1) and a maximum efficiency of 99.69%±0.45%for toluene-in-water emulsion(T/W).Meanwhile,the TiO_(2)@MnO_(2) acts as an excellent electron acceptor and provides positive spatial separation of electrons–holes(e^(-)–h^(+)).The formation of 2D/3D heterojunctions allows the material with wider light absorption and smaller bandgap(2.10 eV).These photoelectric properties give the membrane good degradation of three different pollutants,with about 100%degradation for methylene blue(MB)and malachite green(MG).The photocatalytic antibacterial efficiency of the membrane is also about 100%.After cyclic experiment,the membrane maintains its original separation and photocatalytic *** remarkable multifunctional and self-cleaning properties of the g-C_(3)N_(4) based membrane represent its potential value for complex wastewater treatment.

Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is a monogenic small vessel disease caused by mutations in the NOTCH3 gene. However, the pathogenesis of CADASIL remains unclear, and patients have limited treatment options. Here, we use human induced pluripotent stem cells (hiPSCs) generated from the peripheral blood mononuclear cells of a patient with CADASIL carrying a heterozygous NOTCH3 mutation (c.1261C>T, p.R421C) to develop a disease model. The correction efficiency of different adenine base editors (ABEs) is tested using the HEK293T-NOTCH3 reporter cell line. ABEmax is selected based on its higher efficiency and minimization of predicted off-target effects. Vascular smooth muscle cells (VSMCs) differentiated from CADASIL hiPSCs show NOTCH3 deposition and abnormal actin cytoskeleton structure, and the abnormalities are recovered in corrected hiPSC-derived VSMCs. Furthermore, CADASIL blood vessel organoids generated for in vivo modeling show altered expression of genes related to disease phenotypes, including the downregulation of cell adhesion, extracellular matrix organization, and vessel development. The dual adeno-associated virus (AAV) split-ABEmax system is applied to the genome editing of vascular organoids with an average editing efficiency of 8.82%. Collectively, we present potential genetic therapeutic strategies for patients with CADASIL using blood vessel organoids and the dual AAV split-ABEmax system.

Influencing Factors and Prediction of Risk of Returning to Ecological Poverty in Liupan Mountain Region,China

China has resolved its overall regional poverty in 2020 by attaining moderate societal *** country has entered a new development stage designed to achieve its second centenary ***,ecological fragility and risk susceptibility have increased the risk of returning to ecological *** this paper,the Liupan Mountain Region of China was used as a case study,and the counties were used as the scale to reveal the spatiotempora differentiation and influcing factors of the risk of returning to poverty in study *** indicator data for returning to ecological poverty from 2011-2020 were collected and summarized in three dimensions:ecological,economic and *** autoregressive integrated moving average model(ARIMA)time series and exponential smoothing method(ES)were used to predict the multidimensional indicators of returning to ecological poverty for 61 counties(districts)in the Liupan Mountain Region for *** back propagation neural network(BPNN)and geographic information system(GIS)were used to generate the spatial distribution and time variation for the index of the risk of returning to ecological poverty(RREP index).The results show that 1)ecological factors were the main factors in the risk of returning to ecological poverty in Liupan Mountain Region.2)The RREP index for the 61 counties(districts)exhibited a downward trend from *** RREP index declined more in medium-and high-risk areas than in low-risk *** 2021 to 2025,the RREP index exhibited a slight downward *** 2026 to2030,the RREP index was expected to decline faster,especially from 2029-2030.3)Based on the RREP index,it can be roughly divided into three types,namely,the high-risk areas,the medium-risk areas,and the low-risk *** natural resource conditions in lowrisk areas of returning to ecological poverty,were better than those in medium-and high-risk areas.

Background:Bitter taste receptors(Tas2rs)are generally considered to sense various bitter compounds to escape the intake of toxic *** taste receptors have been found to widely express in extraoral tissues and have important physiological functions outside the gustatory system in ***:To investigate the physiological functions of the bitter taste receptor cluster Tas2r106/Tas2r104/Tas2r105/Tas2r114 in lingual and extraoral tissues,multiple Tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for Tas2rs and Gnat3 gene was microinjected into the cytoplasm of the ***,T7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder *** real-time polymerase chain reaction(qRT-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste *** to taste substance was also studied using twobottle preference ***:We successfully produced several Tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 *** results showed that the expression of GNAT3 and PLCB2 was not altered in Tas2rs mutant *** qRT-PCR results revealed the changed expression profile of m Tas2rs gene in taste buds of these mutant *** two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of *** addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB ***:These findings implied that these mutant mice would be a valuable means to understand the biological functions of TAS2Rs in extraoral tissues and investigate bitter compound-induced responses mediated by these TAS2Rs in many extraoral tissues.

Response of global agricultural productivity anomalies to drought stress in irrigated and rainfed agriculture

The response of agricultural productivity anomalies to drought stress plays a crucial role in the carbon cycle within terrestrial ecosystems and in ensuring food security. However, detailed analysis of how global agricultural productivity anomalies response to drought stress, particularly within irrigated and rainfed agricultural systems, remains insufficient. In this study, the impact of drought stress on agricultural productivity anomalies during the growing season（zcNDVIS）, across both irrigated and rainfed agriculture, were analyzed using a suite of hydro-climatic variables. Specifically, the investigation utilized the multi-scalar Standardized Precipitation Evapotranspiration Index（SPEI）, the Multivariate ENSO Index（MEI）, and the Madden-Julian Oscillation（MJO）. Meanwhile, the relationships between hydroclimatic variables and zcNDVISwere analyzed at one, two, three and four months before the ending of growing season（EOS）. Results showed that（1） the percentages of significant（p<0.1） drying trends varied across the globe from 8.30% to 13.42%, 6.50% to 14.63%, 6.52% to 14.23%, and 6.47%to 14.95% at one-, two-, three-, and four-month lead times before EOS, respectively, during 2001–2020, which represented by the multiscalar SPEI. This observation highlights that most regions across the globe tend to be arid, which could significantly impact agricultural productivity;（2） the global mean correlation coefficients（rmax） for SPEI-1, SPEI-3, SPEI-6, SPEI-12（indicating SPEI at 1-, 3-, 6-, and 12-month lags）, MEI, and MJO with zcNDVISranged between 0.24–0.25, 0.27–0.28, 0.25–0.26, 0.21–0.22, –0.02–0.01 and 0.06–0.11, respectively, across both irrigated and rainfed agriculture system from 2001 to *** productivity anomalies demonstrated a significant correlation with drought stress. Among steganalysis techniques,detection against MV(motion vector)domain-based video steganography in the HEVC(High Efficiency Video Coding)standard remains a challenging *** the purpose of improving the detection performance,this paper proposes a steganalysis method that can perfectly detectMV-based steganography in ***,we define the local optimality of MVP(Motion Vector Prediction)based on the technology of AMVP(Advanced Motion Vector Prediction).Secondly,we analyze that in HEVC video,message embedding either usingMVP index orMVD(Motion Vector Difference)may destroy the above optimality of *** then,we define the optimal rate of MVP as a steganalysis ***,we conduct steganalysis detection experiments on two general datasets for three popular steganographymethods and compare the performance with four state-ofthe-art steganalysis *** experimental results demonstrate the effectiveness of the proposed feature ***,our method stands out for its practical applicability,requiring no model training and exhibiting low computational complexity,making it a viable solution for real-world scenarios.
