Morphometry and subpopulation structure of Holstein bull spermatozoa： variations in ejaculates and cryopreservation straws

Sperm quality is evaluated for the calculation of sperm dosage in artificial reproductive programs. The most common parameter used is motility, but morphology has a higher potential as a predictor of genetic quality. Morphometry calculations from CASA-Morph technology improve morphological evaluation and allow mathematical approaches to the problem. Semen from 28 Holstein bulls was collected by artificial vagina, and several ejaculates were studied. After general evaluation, samples were diluted, packaged in 0.25 ml straws, and stored in liquid nitrogen. Two straws per sample were thawed, and slides were processed and stained with Diff-Quik. Samples were analyzed by a CASA-Morph system for eight morphometric parameters. In addition to the ＂classical＂ statistical approach, based on variance analysis （revealing differences between animals, ejaculates, and straws）, principal component （PC） analysis showed that the variables were grouped into PC1, related to size, and PC2 to shape. Subpopulation structure analysis showed four groups, namely, big, small, short, and narrow from their dominant characteristics, representing 31.0%, 27.3%, 24.1%, and 17.7% of the total population, respectively. The distributions varied between animals and ejaculates, but between straws, there were no differences in only four animals. This modern approach of considering an ejaculate sperm population as divided into subpopulations reflecting quantifiable parameters generated by CASA-Morph systems technology opens a new view on sperm function. This is the first study applying this approach to evaluate different ejaculates and straws from the same individual. More work must be done to improve seminal dose calculations in assisted reproductive programs.

Non-technical approach to the challenges of ecological architecture： Learning from Van der Laan

Up to now, ecology has a strong influence on the development of technical and instrumental aspects of architecture, such as renewable and efficient of resources and energy, COz emissions, air quality, water reuse, some social and economical aspects. These concepts define the physical keys and codes of the current ＇sustainable＇ architecture, normally instrumental but rarely and insufficiently theorised. But is not there another way of bringing us to nature？ We need a theoretical referent. This is where we place the Van der Laan＇s thoughts： he considers that art completes nature and he builds his theoretical discourse on it, trying to better understand many aspects of architecture. From a conceptual point of view, we find in his works sense of timelessness, universality, special attention on the ＇locus＇ and a strict sense of proportions and use of materials according to nature. Could these concepts complement our current sustainable architecture？ How did Laan apply the current codes of ecology in his architecture？ His work may help us to get a theoretical interpretation of nature and not only physical. This paper develops this idea through the comparison of thoughts and works of Laan with the current technical approach to ＇sustainable＇ architecture.

The shortfin mako shark(Isurus oxyrinchus)is one of the fastest marine fishes,reaching speeds of up to 70 km·h^-*** speed is related to the skin surface design composed of dermal *** vary in size and shape according to placement on the body and minimize turbulence around the *** objective of this study is to analyze the interaction between seawater flow and denticles on the dorsal ***-resolution microscopy(scanning electron microscopy and confocal microscopy)were used to measure defined parts of the dermal *** measurements,along with ratios based on length-to-width define three morphologies(rounded,semi-rounded,long)that were 3D *** fluid dynamics simulated fluid passage over reconstructed denticles and describe hy-drodynamic efficiency under different *** increase in angle of inclination produced a relevant increase in the drag coefficient,especially for high velocity *** lowest drag coefficient values were found in long and semi-rounded,followed by rounded *** hydrodynamic behavior of shark skin demonstrates a relation to the morphological characteristics of dermal denticles on the dorsal *** is concluded that the best hydroefficiency relies on the rounded morphology and may serve to design hydrodynamically efficient surfaces or manmade assemblies.

This work provides information on the blue fox ejaculated sperm quality needed for seminal dose calculations. Twenty semen samples, obtained by masturbation, were analyzed for kinematic and morphometric parameters by using CASA-Mot and CASA-Morph system and principal component （PC） analysis. For motility, eight kinematic parameters were evaluated, which were reduced to PC1, related to linear variables, and PC2, related to oscillatory movement. The whole population was divided into three independent subpopulations： SP1, fast cells with linear movement; SP2, slow cells and nonoscillatory motility; and SP3, medium speed cells and oscillatory movement. In almost all cases, the subpopulation distribution by animal was significantly different. Head morphology analysis generated four size and four shape parameters, which were reduced to PC 1, related to size, and PC2, related to shape of the cells. Three morphometric subpopulations existed. SPI： large oval cells; SP2： medium size elongated cells; and SP3. small and short cells. The subpopulation distribution differed between animals. Combining the kinematic and morphometric datasets produced PC1, related to morphometric parameters, and PC2, related to kinematics, which generated four sperm subpopulations - SPI.. high oscillatory motility, large and short heads; SP2; medium velocity with small and short heads; SP3. slow motion small and elongated cells; and SP4. high linear speed and large elongated cells. Subpopulation distribution was different in all animals. The establishment of sperm subpopulations from kinematic, morphometric, and combined variables not only improves the well-defined fox semen characteristics and offers a good conceptual basis for fertility and sperm preservation techniques in this species, but also opens the door to use this approach in other species, included humans.

Dogs have been under strong artificial selection as a consequence of their relationship with man. Differences between breeds are evident that could be reflected in seminal characteristics. The present study was to evaluate differences in sperm head morphometry between seven well-defined breeds of dog： the British Bulldog, Chihuahua, German Shepherd, Labrador Retriever, Spanish Mastiff, Staffordshire Terrier, and Valencian Rat Hunting dog. Semen samples were obtained by masturbation and smears stained with Diff-Quik. Morphometric analysis （CASA-Morph） produced four size and four shape parameters. Length, Ellipticity, and Elongation showed higher differences between breeds. MANOVA revealed differences among all breeds. Considering the whole dataset, principal component analysis （PCA） showed that PC1 was related to head shape and PC2 to size. Procluster analysis showed the British Bulldog to be the most isolated breed, followed by the German Shepherd. The PCA breed by breed showed the Chihuahua, Labrador Retriever, Spanish Mastiff, and Staffordshire Terrier to have PC1 related to shape and PC2 to size, whereas the British Bulldog, Valencia Rat Hunting dog, and German Shepherd had PC1 related to size and PC2 to shape. The dendrogram for cluster groupings and the distance between them showed the British Bulldog to be separated from the rest of the breeds. Future work on dog semen must take into account the large differences in the breeds＇ sperm characteristics. The results provide a base for future work on phylogenetic and evolutionary studies of dogs, based on their seminal characteristics.

Milk Yield of Holstein Cows Induced into Lactation Twice Consecutively and Lactation Curve Models Fitted to Artificial Lactations

Nineteen multiparous barren Holstein cows were subjected to an induction of lactation protocol for 21 d administering estradiol cypionate （2 mg kg-1 of body weight （BW） d-1, on day 1 to 14）, progesterone （0.10 mg kg-1 of BW, on day 1 to 7）, lfumethasone （0.03 mg kg-1 of BW, on day 18 to 20） and recombinant bovine somatotropin （rbST;500 mg per cow, on day 1, 6, 16 and 21）. At the end of lactation and with a minimum of a 2-mon dry period, the same cows were again hormonally induced into lactation. Cows in both lactations were not artiifcially inseminated, they were milked 3 times daily and received rbST throughout lactation. Mean accumulated milk yield at 305 d in milk （DIM） did not differ between the ifrst and second induced lactations （（9 710 ＆#177;1 728） vs. （9 309＆#177;2 150） kg;mean＆#177;SD）. Total milk yield （（12 707＆#177;3 406） vs. （12 306＆#177;4 218） kg;mean＆#177;SD） and lactation length （（405＆#177;100） vs. （410＆#177;91） d;mean＆#177;SD） were not different between the ifrst and second induced lactations. In a second study, 15 empirical models including exponential, power law, yield-density, sigmoidal and miscellaneous models were compared for their suitability by modeling 12-mon （n=334）, 18-mon （n=164） and 29-mon （n=22） lactation cycles of Holsteins cows induced into lactation and treated with rbST throughout the lactation. Hoerl （Y=ab1/xxc）, Wood （Y=axb exp（cx）） and Dhanoa （Y=ax（bc）exp（cx）） models were equally suitable to describe 12-mon lactations. An exponential model with ifve parameters （Y=exp（a＋bx＋cd2＋e/x）） showed the best ift for milk yield for 18-mon lactations. The rational model （Y=a＋bx/1＋cx＋dx2） was found to produce the closest ift for 29-mon lactations. It was concluded that, with the protocol used in the present study, multiparous cows respond favorably to a second cycle of induced lactation, with milk yield similar to that experienced during the ifrst cycle. Z-line examination by the PillCam^(TM) SB:Prospective comparison of three ingestion protocols

AIM:To evaluate the Z-line visualization by the PillCamTM SB2 using three different ingestion protocols. METHODS:Ninety consecutive patients undergoing small bowel capsule endoscopy(SBCE)between January and May 2008 were included in the study. They swallowed the capsule in the standing(Group A= 30),supine(Group B=30)and right supine positions (Group C=30).baseline patient characteristics, difficulties in capsule ingestion,esophageal transit times(ETT)and Z-line visualization were noted. RESULTS:No significant differences were found between the groups with regard to baseline patient characteristics,ingestion difficulties and complete SB examinations(P>0.05).At least 1 frame of the Z-line was detected in 15.8%,46.7%and 90%of patients in groups A,B and C,respectively(P<0.001).The average number of Z-line images was 0.21±0.53,3.23 ±6.59 and 5.53±7.55 and the mean%of the Z-linedetected was 71.3,25.1 and 8.3,in groups A,B and C, respectively(both P<0.001).ETT times were longer in the supine group followed by the right supine and the standing groups(median of 237 s vs 64 s and 39 s, respectively;P<0.001). CONCLUSION:Z-line visualization in patients undergoing SBCE can be accurately achieved in most cases when the capsule is swallowed in the right supine position.

Episcleral brachytherapy as an effective alternative in vasoproliferative tumors

The aim of this study is to assess the results of episcleral brachytherapy as treatment of retinal vasoproliferative tumors(RVTs) in a referral Intraocular Tumors Unit(ITU). A retrospective review of all patients diagnosed with vasoproliferative tumors of the retina and treated with episcleral brachytherapy in the ITU, University Hospital of Valladolid between 2009 and 2015 was done. Five patients accomplished the inclusion criteria. All of them presented associated exudation and secondary retinal detachments(RD). Four patients had received prior treatments. Decreased tumor size and exudation regression was found in all cases after treatment. Visual acuity remained stable or increased in all patients. No recurrences have been found after twelve-months follow up. The results of the present study suggest that episcleral brachytherapy is an efficient and safe option in the management of vasoproliferative tumors, especially when large tumor or extensive subretinal fluid is present. In these cases episcleral brachytherapy could be considered as a first line treatment.

Enhanced history matching process by incorporation of saturation logs as model selection criteria

This paper proposes a methodology for an alternative history matching process enhanced by the incorporation of a simplified binary interpretation of reservoir saturation logs(RST) as objective function. Incorporating fluids saturation logs during the history matching phase unlocks the possibility to adjust or select models that better represent the near wellbore waterfront movement, which is particularly important for uncertainty mitigation during future well interference assessments in water driven reservoirs. For the purposes of this study, a semi-synthetic open-source reservoir model was used as base case to evaluate the proposed methodology. The reservoir model represents a water driven, highly heterogenous sandstone reservoir from Namorado field in Brazil. To effectively compare the proposed methodology against the conventional methods, a commercial reservoir simulator was used in combination with a state-of-the-art benchmarking workflow based on the Big LoopTMapproach. A well-known group of binary metrics were evaluated to be used as the objective function, and the Matthew correlation coefficient(MCC) has been proved to offer the best results when using binary data from water saturation logs. History matching results obtained with the proposed methodology allowed the selection of a more reliable group of reservoir models,especially for cases with high heterogeneity. The methodology also offers additional information and understanding of sweep behaviour behind the well casing at specific production zones, thus revealing full model potential to define new wells and reservoir development opportunities.
