Functional Tregs play a key role in tumor development and progression,representing a major barrier to anticancer *** mechanisms by which Tregs are generated in cancer and the influence of the tumor microenvironment on these processes remain incompletely ***,by using NMR,chemoenzymatic structural assays and a plethora of in vitro and in vivo functional analyses,we demonstrate that the tumoral carbohydrate A10(Ca10),a cell-surface carbohydrate derived from Ehrlich’s tumor(ET)cells,is a heparan sulfate-related proteoglycan that enhances glycolysis and promotes the development of tolerogenic features in human ***10-stimulated human DCs generate highly suppressive Tregs by mechanisms partially dependent on metabolic reprogramming,PD-L1,IL-10,and ***10 also reprograms the differentiation of human monocytes into DCs with tolerogenic *** solid ET-bearing mice,we found positive correlations between Ca10 serum levels,tumor size and splenic Treg *** of isolated Ca10 also increases the proportion of splenic Tregs in tumor-free ***,we provide evidence supporting the presence of a circulating human Ca10 counterpart(Ca10H)and show,for the first time,that serum levels of Ca10H are increased in patients suffering from different cancer types compared to healthy *** note,these levels are higher in prostate cancer patients with bone metastases than in prostate cancer patients without ***,we reveal novel molecular mechanisms by which heparan sulfate-related structures associated with tumor cells promote the generation of functional Tregs in *** discovery of this novel structural-functional relationship may open new avenues of research with important clinical implications in cancer treatment.

On the Hyperferritinemia and Hereditary Cataract syndrome

Introduction. Mutations in the promoter region of ferritin light gene can induce an uncontrolled over expression of this protein. Consequently, ferritin is found in serum at very high levels (~1000 ng/mL) and it accumulates in the crystalline lens, generating cataracts. This entity is known as hyperferritin and hereditary cataract syndrome (HHCs) which is inherited in an autosomal dominant manner. Case Presentation. We describe a family affected by HHCs. The proband was identified among subjects submitted to a biological screening for hemochromatosis. He had very high levels of serum ferritin (~900 ng/mL) with normal transferrin saturation (Ts). The proband has a single H63D HFE-gene mutation and normal HAMP-gene. He was submitted to periodical phlebotomies that induced anemia and a decrease in Ts but no changes on serum ferritin levels. Analyses of promoter region of ferritin-light chain gene showed a 39 C > T mutation, responsible for HHCs. The proband’s sister carried also this mutation. Both subjects had developed cataracts. Discussion. similar to the first family described carrying this syndrome and to other cases reported, the proband was erroneously submitted to phlebotomies. Clinical consequences are illustrated in this report. HHCs is an infrequent entity which has to be correctly identified. The unique therapeutic approach to this syndrome must be cataract surgery.

Age structure of a lizard along an elevational gradient reveals nonlinear lifespan patterns with altitude

Lifespan is one of the main components of life *** lifespans can be expected in marginal ***,in the case of ectotherms,lifespan typically increases with altitude,even though temperature-one of the main factors to determine ectotherms'life history-declines with *** pattern can be explained by the fact that a shorter activity time favors *** this study,we analyzed how lifespan and other life-history traits of the lizard Psammodromus algirus vary along a 2,200 m elevational gradient in sierra Nevada(sE spain).Populations at inter-mediate altitudes(1,200-1,700 m),corresponding to the optimal habitat for this species,had the shortest lifespans,whereas populations inhabiting marginal habitats(at both low and at high altitudes)lived ***,this lizard did not follow the typical pattern of ectotherms,as it also lived longer at the lower limit of its distribution,nor did it show a longer lifespan in areas with optimal *** results might be explained by a complex combination of different gradients along the mountain,namely that activity time decreases with altitude whereas food availability *** could explain why lifespan was maximum at both high(limited activity time)and low(limited food availability)altitudes,resulting in similar lifespans in areas with contrasting environmental *** findings also indicated that reproductive investment and body condition increase with elevation,suggesting that alpine populations are locally adapted.

Electrochemical treatment is an alternative modality for tumor treatment based on the application of a low intensity direct electric current to the tumor tissue through two or more platinum electrodes placed within the tumor zone or in the surrounding areas. This treatment is noted for its great effectiveness, minimal invasiveness and local effect. several studies have been conducted worldwide to evaluate the anfitumoral effect of this therapy. In all these studies a variety of biochemical and physiological responses of tumors to the applied treatment have been obtained. By this reason, researchers have suggested various mechanisms to explain how direct electric current destroys tumor ceils. Although, it is generally accepted this treatment induces electrolysis, electroosmosis and electroporation in tumoral tissues. However, action mechanism of this alternative modality on the tumor tissue is not well understood. Although the principle of Electrochemical treatment is simple, a standardized method is not yet available. The mechanism by which Electrochemical treatment affects tumor growth and survival may represent more complex process. The present work analyzes the latest and most important research done on the electrochemical treatment of tumors. We conclude with our point of view about the destruction mechanism features of this alternative therapy. Also, we suggest some mechanisms and strategies from the thermodynamic point of view for this therapy. In the area of Electrochemical treatment of cancer this tool has been exploited very little and much work remains to be done. Electrochemical treatment constitutes a good therapeutic option for patients that have failed the conventional oncology methods.

Prey availability, prey selection, and trophic niche width in the lizard Psammodromus algirus along an elevational gradient

Mountains imply enormous environmental variation, with alpine habitats entailing harsh environ- ments, especially for ectotherms such as lizards. This environmental variability also may imply variation in prey availability. However, little is known about how lizard trophic ecology varies with elevation. In this study, we analyze diet, prey availability, prey selection, and trophic niche width in the lacertid lizard Psammodromus algirus along a 2,200-m elevational gradient in the sierra Nevada （sE spain）. The analysis of fecal samples has shown that Orthoptera, Formicidae, Hemiptera, Coleoptera, and Araneae are the main prey, although, according to their abundance in pitfall traps, Formicidae and Coleoptera are rejected by the lizard whereas Orthoptera, Hemiptera, and Araneae are preferred. Prey abundance and diversity increase with elevation and diet subtly varies along with the elevational gradient. The consumption of Coleoptera increases with elevation probably as a consequence of the lizard foraging more in open areas while basking. The electivity for Araneae increases with elevation. Araneae are rejected in the lowlands--where they are rela- tively abundant--whereas, at high elevation, this lizard positively selects them, despite they being less abundant. The lizard trophic niche width expands with elevation due to concomitant greater prey diversity and hence this lizard feeds on more prey types in highlands. Although no sex difference in diet has been found, the trophic niche is broader in females than males. As a whole, alpine lizards show a trophic niche simitar to that found at lower elevat（ons, suggesting that P. algirus is well adapted to the harsh environment found in alpine areas.

studying the causes of parasite geographic distribution is relevant to understand ecological and evolutionary processes that affect host populations as well as for species conservation. Temperature is one of the most important environmental variables affecting parasite distribution, as raising temperatures positively affect development, reproduction, and rate of transmission of both endo- and ectoparasites. In this context, it is generally accepted that, in mountains, parasite abundance decreases with elevation. However, empirical evidence on this topic is limited. In the present study, we analyzed the elevational variation of hemoparasites and ectoparasites of a lizard, Psammodromus algirus, along a 2,200-m elevational gradient in sierra Nevada （sE spain）. As pre- dicted, ectoparasite （mites, ticks, mosquitoes, and sandflies） abundance decreased with elevation. However, hemoparasite prevalence and intensity in the lizard augmented with altitude, showing a pattern contrary to their vectors （mites）. We suggest that tolerance to hemoparasites may increase with elevation as a consequence of lizards at high altitudes taking advantage of increased body condition and food availability, and reduced oxidative stress. Moreover, lizards could have been selected for higher resistance against hemoparasites at lowlands （where higher rates of replication are expected）, thus reducing hemoparasite prevalence and load. Our findings imply that, in a scen- ario of climate warming, populations of lizards at high elevation may face increased abundance of ectoparasites, accompanied with strong negative effects.

Monitoring by 1H Nuclear Magnetic Resonance of the Glycerin Purification by Vacuum Distillation

Given the growth in biodiesel production and other biofuels, it involves large volumes of byproducts which aren"t suitable for commercial use. We studied the glycerol purification by vacuum distillation of the glycerol rich phase in the mixture reaction at the end of the biodiesel production process. since glycerin is one of those byproducts, useful and plentiful, therefore the options to purify this one and achieve its better trading benefit are interesting. The process was monitored by 1 H NMR and showed that by vacuum fractional distillation of the glycerin rich phase of the biodiesel production it was possible to obtain pure glycerin between 97.4% and 98.9% on dry basis with 49.0% yield. Vacuum distillation without fractionation was inconvenient because it generates thermal degradation of the material and causes the formation of foam which induces low yield of glycerin and hinders the operation progress.
