AIM To establish minimum clinically important difference(MCID) for measurements in an orthopaedic patient population with joint *** Adult patients aged 18 years and older seeking care for joint conditions at an orthopaedic clinic took the Patient-Reported Outcomes Measurement Information System Physical Function(PROMIS~? PF) computerized adaptive test(CAT), hip disability and osteoarthritis outcome score for joint reconstruction(HOOS JR), and the knee injury and osteoarthritis outcome score for joint reconstruction(KOOS JR) from February 2014 to April 2017. MCIDs were calculated using anchorbased and distribution-based methods. Patient reports of meaningful change in function since their first clinic encounter were used as an *** There were 2226 patients who participated with a mean age of 61.16(SD = 12.84) years, 41.6% male, and 89.7% Caucasian. Mean change ranged from 7.29 to 8.41 for the PROMIS~? PF CAT, from 14.81 to 19.68 for the HOOS JR, and from 14.51 to 18.85 for the KOOS JR. ROC cut-offs ranged from 1.97-8.18 for the PF CAT, 6.33-43.36 for the HOOS JR, and 2.21-8.16 for the KOOS JR. Distribution-based methods estimated MCID values ranging from 2.45 to 21.55 for the PROMIS~? PF CAT; from 3.90 to 43.61 for the HOOS JR, and from 3.98 to 40.67 for the KOOS JR. The median MCID value in the range was similar to the mean change score for each measure and was 7.9 for the PF CAT, 18.0 for the HOOS JR, and 15.1 for the KOOS *** This is the first comprehensive study providing a wide range of MCIDs for the PROMIS? Preparation of a recombinant adeno-associated viral vector with a mutation of human factor-Ⅸin large scale and its expression in vitro and in vivo

A series of adeno-associated viral vectors containing a mutation of human factor Ⅸ (hFⅨR338A) with different regulation elements were constructed and used to transduce cell lines. The plasmids and the stable transduction cell clones with high expression level of hFⅨR338A were obtained by selecting and optimizing, and then, the recombinant adeno-associated viral vector with hFⅨR338A was prepared via novel rHSV/AAV hybrid virus packaging system on a large scale, which contained the capsid protein genes. A method for producing rAAV-hFⅨR338A viral stocks on a large scale and higher titer was established, which can be used for industrial purpose. The titer of rAAV-hFⅨR338A was more than 1.25?012 particle/ml, and then, a mammalian cell line, C2C12 and the factor Ⅸ knock-out mice were transfected with the rAAV-hFⅨR338A in vitroand in vivo. The results show that the high-level expression of rAAV-hFⅨR338A was achieved in cell line and hemophilia B mice. It reached at (2551.32±92.14) ng·(106 cells)-1·(24h)-1 in C2C12 cell in vitro and had a peak concentration of 463.28 ng/ml in mice treated with rAAV-hFⅨR338A, which was as high as the expression of rAAV-hFⅨ-wt(2565.76?4.36) ng·(106 cells) -1·(24 h)-1 in C2C12 and453.92 ng/ml in the mice treated with rAAV-hFⅨ-wt) in vitro and in vivo, there is no any difference between two groups, but the clotting activity of hFⅨR338A is about 2.46 times higher than that of hFⅨ-wt. It was first reported that a mutation of human factor Ⅸ was used into gene therapyresearch for hemophilia B, meanwhile, a novel packagingsystem, rAAV/HSV was used for preparation of rAAV-hFⅨR338A on a large scale, which laid the foundation ofindustrial production for applying rAAV viral stocks to gene therapy clinical trial for hemophilia B mediated withrAAV-hFⅨ.

Decades of research has explored the epigenetic control of gene expression and the impact of histone post-translational modifications （PTMs）, such as acetylation, on chromatin remodel- ing. Indeed, the writers, readers, and erasers of lysine acetylation are increasingly well understood. Recent studies have added crotonylation, butyrylation, and propionylation to the types of acylations by which histones are modified, and identified the YEATS protein domain as a critical reader of crotonylation. Now, Haitao li, David Allis, and their col- leagues expand the scope of protein domains capable of read- ing crotonyl-lysine （Kcr） to include double PHD finger （DPF） domains. Importantly, the mechanism through which these domains recognize Kcr is quite distinct from their recognition by the YEATS domain [1]. In this highlight, we discuss recognition of acylated histones by the bromodomain （BRD）, the YEATS domain, and PHD fingers. We contrast the structural basis for their recognition of histones modified by acetylation and more recently discovered histone crotonylation [2-6].

As femtosecond(fs)laser machining advances from micro/nanoscale to macroscale,approaches capable of machining macroscale geometries that sustain micro/nanoscale precisions are in great *** this research,an fs laser sharp shaping approach was developed to address two key challenges in macroscale machining(*** on edges and tapered sidewalls).The evolution of edge sharpness(edge transition width)and sidewall tapers were systematically investigated through which the dilemma of simultaneously achieving sharp edges and vertical sidewalls were *** decreasing the angle of incidence(AOI)from 0◦to−5◦,the edge transition width could be reduced to below 10µm but at the cost of increased sidewall ***,by analyzing lateral and vertical ablation behaviors,a parameter-compensation strategy was developed by gradually decreasing the scanning diameters along depth and using optimal laser powers to produce non-tapered *** fs laser ablation behaviors were precisely controlled and coordinated to optimize the parameter compensations in general manufacturing *** AOI control together with the parameter compensation provides a versatile solution to simultaneously achieve vertical sidewalls as well as sharp edges of entrances and exits for geometries of different shapes and *** mm-scale diameters and depths were realized with dimensional precisions below 10µm and surface roughness below 1µ*** research establishes a novel strategy to finely control the fs laser machining process,enabling the fs laser applications in macroscale machining with micro/nanoscale precisions.

The writings of Soren Kierkegaard provide a theological and philosophical foundation as experienced in the life of a tragic hero and a knight of faith. It will be shown that the ethical dimensions as experienced by Abraham in ＂Fear and Trembling＂ provide a core ontology in the lives of Jan Patocka and Viktor Frankl. Jan Parocka developed a phenomenological, Kierkegaardian, existential approach in his writings and lectures that enabled him to undergo his interrogations by the Czech communist secret police. The World War II concentration camp experiences of Viktor Frankl led him to develop a Kierkegaardian, existential, noetic approach to discover meaning under inhuman living conditions. We, as humans, are searching for a spiritual core to transcendence, as seen in the writings of Kierkeggard to reach beyond ourselves in the fully human dimension of the good, the true, and the beautiful in faith and in love. Special attention will be given to the lives of Patocka, Frankl, and how Kierkegaard＇s theology and philosophy contribute a transhistorical, phenomenological, existential, meaning-centered approach to understanding trauma, especially in the lives of returning veterans and their families.

我们在斑块化的景观中实验测定了犬尾田鼠(Microtus canicaudus)的生境喜好,验证下列假说:在一斑块生境中,与边缘区域相比,雌性田鼠喜好内部区域。在低密度和高密度时成年雌性的巢区在生境内部区域分别占100%和76%。高密度时在边缘区域雌性的捕获率较低,这种差异在低密度时更突出。在高密度时通过选择性去除生境内部和边缘区域的一些雌体,边缘区域13只雌体中8只(占62%)的巢区发生从边缘到内部区域的转移,内部区域20只雌体中只有3只(占15%)的巢区向边缘区域转移。动物的繁殖率、生存和体重在两个区域之间没有差异。但是,边缘区域个体的巢区比内部区域的要小。这些结果支持一些关于其他啮齿动物的研究观察结果(如鼠平类和田鼠类),但与草原田鼠不同。因此在一斑块生境中,与边缘区域相比,长尾田鼠更喜好内部区域,边缘区域与内部区域的比率可潜在影响动物的生境选择,也可能会影响斑块化生境中的种群统计学特征。

Dear Editor,Three polyamines,putrescine,spermidine,and spermine,are tightly regulated and are synthesized from arginine,ornithine,and methionine(Miller-Fleming et al.,2015;Figure 1A).Polyamine levels are increased in the red blood cells of amyotrophic lateral sclerosis and Parkinson’s disease(PD)patients(Miller-Fleming et al.,2015).Sulfur amino acid catabolism regulated by the molybdopterin synthase associating complex prevents onset of early events in PD and Alzheimer’s disease(Suganuma et al.,2018).MPT synthase,a heterodimer of MOCS2A and MOCS2B encoded by MOCS2,is essential for the biosynthesis of molybdenum cofactor(Moco).
