Pinea.ple is the first fruit crop cultiva.ed in south Benin tha. grea.ly contributes for food a.d nutritiona. security a.d fa.mers’ income. But the la.k of homogenous pla.ting ma.eria. constitutes the ma.or constra.nt for improving pinea.ple yield. In vitro micropropa.a.ion is now used in the production of homogenous a.d free disea.e pla.ting ma.eria.s of pinea.ple. However, the a.clima.iza.ion to na.ura. condition of pinea.ple pla.tlets is a. importa.t step in pla.ting ma.eria. production of this crop. Here, we determined the intrinsic a.d extrinsic fa.tors which influence the beha.ior of pla.tlets during the a.clima.iza.ion process. For this purpose, pla.tlets from different ca.egories were selected, trimmed a.d pla.ted on a.horticultura. substra.e ma.e up of potting soil, white sa.dust a.d compost previously sterilized. The pla.tlets were then incuba.ed in under a.clima.iza.ion greenhouse with a.era.e tempera.ure of 29˚C a.d 70.2% of humidity. a.ba.ch of pla.tlets wa. subjected to two different wa.ering solutions: Shive a.d Robbins solution a.d NPK 14-6-5 folia. fertilizer. The results obta.ned initia.ly showed high ra.e (100%) of surviva. a.d growth of the pla.tlets wa.ered with Shive a.d Robbins solution a.a.nst 50% of the pla.tlets wa.ered with the folia. fertilizer solution. In a.dition, the pla.tlets with sprea. pores exhibiting the cha.a.teristics of which the number of lea.es va.ies between 9 a.d 11, the weight between 1.2 a.d 1.5 g, the size of 4.5 to 5.5 cm, a.d a.good junction between the a.ria. pa.t a.d the root system were those which were successfully grown in a.clima.iza.ion pha.e under greenhouse, unlike pla.tlets with erected pores ha.ing lower success ra.e. This study goes a.long wa. in providing good procedures of a.clima.iza.ion of homogenous a.d free disea.e pla.ting ma.eria. of pinea.ple to the fa.ers.

Pla.t Regenera.ion through Indirect Orga.ogenesis in Two Cultiva.s of Pinea.ple (a.a.a. comosus L.)

Una.a.la.ility of performa.t pla.ting ma.eria. of pinea.ple constitutes a.ma.or problem of its cultiva.ion in a.rica. For this purpose, indirect orga.ogenesis technique is used to eva.ua.e the in vitro responses of two cultiva.s of pinea.ple during the expla.t’s regenera.ion. Ca.li were induced from crown lea. a.d pla.tlets lea. of “Smooth Ca.enne” a.d “Suga.loa. cultiva.s”. Mura.hige a.d Skoog medium with vita.ins B5 supplemented with different growth regula.ors combina.ions were used. Ba. a.d/or 2,4-D ha.e been a.ded to ba.e medium for ca.li cells’ differentia.ion while Ba. a.d Ga. ha.e been a.ded for pla.t elonga.ion. The results indica.ed tha. expla.ts from regenera.ed pla.tlets lea.es cultiva.ed on MS supplemented with copper (II) sulpha.e 5-hydra.e concentra.ions incorpora.ed ha. significa.t (p a.lus induction in pinea.ple cultiva.s. Likewise, MS medium with Na. (0.5 mg/l) + Ba. (1 mg/l) ha. a.highly significa.t influence with 8.8 differentia.ed Ca.li. a.so, MS medium supplemented with Ba. (3 mg/l) + Ga. (2 mg/l) for the “smooth Ca.enne” ha. significa.tly influenced (p a.li regenera.ion with a.high ra.e of 55.25% pla.tlets. MS medium conta.ning 0.5 mg/l of Na. + 0 mg/l IBa.produced a.high number of roots in Suga.loa. wherea. the medium conta.ning 1.5 mg/l Na. + 0.5 mg/l (IBa. produced high number of roots in smooth Ca.enne. We ha.e esta.lished a. efficient a.d reproducible protocol for ma.s propa.a.ion a.d genetic tra.sforma.ion of pinea.ple though indirect orga.ogenesis. This protocol ma. be used in genetics engineering studies for pinea.ple breeding.

The a.a.ta.ion of ca.sa.a.pla.tlets to na.ura. conditions rema.ns the ma.n obsta.le in the process of free virus pla.tlets production. In this study, ca.sa.a.pla.tlets a.clima.iza.ion wa. a.sessed by pre-a.clima.iza.ion, height of a.clima.ed pla.tlets, substra.e, a.d nutrient wa.ering solution. The results revea.ed tha. pre-a.clima.iza.ion ga.e a.high surviva. ra.e (61.9%) wherea. the direct tunnel a.clima.iza.ion wa. conducted to low ra.e (27.31%) of pla.tlets. Furthermore, the height of pla.tlets influenced significa.tly (p a. ra.e with best surviva. ra.e observed on pla.tlet height from 1 cm to 5 cm. There wa. no significa.t difference between the substra.es used a.d wa.ering solutions. However, the Sa.dust substra.e ga.e a.high surviva. ra.e (46.67%) a.d better phylogenesis (1.8) in the va.iety RB 89509. Likewise, pla.tlets trea.ed with NPK nutrient wa.ering solutions (20-10-10) ha. a.better surviva. ra.e (48.57%), better phylogenesis (2.125) with the va.iety RB 89509 a.d better growth in height (1.18 cm) with the va.iety BF92/0267. These findings constitute a.da.a.a.e for the esta.lishment of the technica. itinera.y for the a.clima.iza.ion of ca.sa.a.pla.tlets.
