In this paper, the authors present an analysis of the magnitude of the temporal and spatial acceleration (inertial) terms in the surface-water flow equations and determine the conditions under which these inertial terms have sufficient magnitude to be required in the computations. data from two South Florida field sites are examined and the relative magnitudes of temporal acceleration, spatial acceleration, and the gravity and friction terms are compared. Parameters are derived by using dimensionless numbers and applied to quantify the significance of the hydrodynamic effects. The time series of the ratio of the inertial and gravity terms from field sites are presented and compared with both a simplified indicator parameter and a more complex parameter called the Hydrodynamic Significance Number (HSN). Two test-case models were developed by using the SWIFT2d hydrodynamic simulator to examine flow behavior with and without the inertial terms and compute the HSN. The first model represented one of the previously-mentioned field sites during gate operations of a structure-managed coastal canal. The second model was a synthetic test case illustrating the drainage of water down a sloped surface from an initial stage while under constant flow. The analyses indicate that the times of substantial hydrodynamic effects are sporadic but significant. The simplified indicator parameter correlates much better with the hydrodynamic effect magnitude for a constant width channel such as Miami Canal than at the non-uniform North River. Higher HSN values indicate flow situations where the inertial terms are large and need to be taken into account.

A martensitic stainless steel AISI420 fabricated by selective laser melting(SLM)and post-processed by austenitizing and tempering heat treatment was investigated in this *** as-fabricated SLMed AISI420 showed a strong mechanical property anisotropy and low ductility in the longitudinal *** microstructural characterization revealed the presence of austenite and a relatively sharp solidification texture in the as-fabricated state,while the sharp texture was considered as the cause for the mechanical property *** contrast,a fully martensitic microstructure with a very weak texture was achieved after the austenitizing and tempering heat *** mechanical property anisotropy was also fully eliminated,witnessing a significant improvement in the ductility,and thus,comparable mechanical performance with the wrought product was achieved in this study.

The Coastal Wetlands Planning, Protection and Restoration Act (CWPPRA) funded construction of the West Bay Sediment diversion Project (WBSd) on the west bank of the Mississippi River for the purpose of coastal restoration. A multi-tiered sediment study for the WBSd was conducted to determine impacts to the adjacent navigation channel and to the Pilottown Anchorage Area (PAA). One tier of the study is the implementation of HEC-6T, a one-dimensional (1-d) sediment model, to evaluate the regional impacts of the WBSd. The HEC-6T model results shows the long-term channel changes associated with the WBSd to be increasing shoaling in the adjacent areas on the order of 10% - 20%, as compared to the no WBSd condition. However, it is extremely difficult to isolate the impacts associated with a single diversion due to multiple diversions in the region. From a holistic vantage point, the 1-d model shows the necessity to examine, on a regional scale, the lower Mississippi River as a single congruent system on a regional scale.

Last year, the first attempt to genetically modify human embryos in the United States was reported and sparked a huge debate (Ma et al., 2017). Although the first human germline modification was only performed two years ago, the study showed that rapid adva nces in tech no logy has allowed the rate of off-target effects and mosaicism to be reduced considerably (Liang et al., 2015). Recently, Vertex and CRISPR therapeutics collaborated and developed CTX001, the first CRISPR/Cas9-based therapy, targeting patients with P-thalassemia and have begun phase 1/2 clinical trials. With policies and technologies regarding genome editing both developing rapidly, explorations into the possibility of clinical gene editing for hundreds of hereditary diseases are starting to become achievable. Here, we address the progress of huma n embryo editi ng tech no logies so far and its promise and risks in advancing therapy for hereditary diseases.

Previous results indicated that in grapevine （Vitis vinifera）, regulation of the flavonoid pathway genes by MYB transcription factors depends on their interaction with basic helix-loop-helix proteins （bHLHs）. The present study describes the first functional characterization of a bHLH factor from grapevine named VvMYC1. This transcription factor is phylogenetically related to Arabidopsis bHLH proteins, which participate in the control of flavonoid biosynthesis and epidermal cell fate. Transient promoter and yeast two-hybrid assays demonstrated that VvMYC1 physically interacts with MYB5a, MYB5b, MYBA1/A2, and MYBPA1 to induce promoters of flavonoid pathway genes involved in anthocyanin and/ or proanthocyanidin （PA） synthesis. Additionally, transient promoter assays revealed that VvMYC1 is involved in feedback regulation of its own expression. Transcript levels of VvMYC1 during berry development correlate with the synthesis of anthocyanins and PAs in skins and seeds of berries, suggesting that VvMYC1 is involved in the regulation of anthocyanins and PA synthesis in these organs. Likewise, transient expression of VvMYC1 and VvMYBA1 induces anthocyanin synthesis in grapevine suspension cells. These results suggest that VvMYCl is part of the transcriptional cascade controlling antho- cyanin and PA biosynthesis in grapevine.

Co-evolution of host-parasitoid interactions is determined by the costs of host resistance, which received empirical evidence, and the costs ofparasitoid virulence, which have been mostly hypothesized. Asobara tabida is a parasitoid, which mainly parasitizes drosophila melanogaster and d. subobscura, the first species being able to resist to the parasitoid development while the second species is not. To parasitize resistant hosts, including d. melanogaster, A. tabida develops sticky eggs, which prevent encapsulation, but this virulence mechanism may be costly. Interindividual and interpopulation variation in the proportion of sticky eggs respectively allowed us to （i） artificially select and compare life-history traits of a virulent and a nonvirulent laboratory strain, and （ii） compare a virulent and a nonvirulent field strain, to investigate the hypothetical costs of virulence. We observed strong differences between the 2 laboratory strains. The nonvirulent strain invested fewer resources in reproduction and walked less than the virulent one but lived longer. Concerning the field strains, we observed that the nonvirulent strain had larger wings while the virulent one walked more and faster. All together, our results suggest that virulence may not always be costly, but rather that different life histories associated with different levels of virulence may coexist at both intra- and interdodulation levels.
