Genetic interactions between polycystin-1 and Wwtr1 in osteoblasts define a novel mechanosensing mechanism regulating bone formation in mice

Molecular mechanisms transducing physical forces in the bone microenvironment to regulate bone mass are poorly ***,we used mouse genetics,mechanical loading,and pharmacological approaches to test the possibility that polycystin-1 and Wwtr1 have interdependent mechanosensing functions in *** created and compared the skeletal phenotypes of control Pkd1^(flox/)+;Wwtr1^(flox/)+,Pkd1^(Oc-cKO),Wwtr1^(Oc-cKO),and Pkd1/Wwtr1^(Oc-cKO)mice to investigate genetic *** with an interaction between polycystins and Wwtr1 in bone in vivo,Pkd1/Wwtr1^(Oc-cKO)mice exhibited greater reductions of BMd and periosteal MAR than either Wwtr1Oc-cKOor Pkd1^(Oc-cKO)***-CT 3d image analysis indicated that the reduction in bone mass was due to greater loss in both trabecular bone volume and cortical bone thickness in Pkd1/Wwtr1Oc-cKO mice compared to either Pkd1Oc-cKOor Wwtr1^(Oc-cKO)***1/Wwtr1^(Oc-cKO)mice also displayed additive reductions in mechanosensing and osteogenic gene expression profiles in bone compared to Pkd1Oc-cKOor Wwtr1^(Oc-cKO)***,we found that Pkd1/Wwtr1^(Oc-cKO)mice exhibited impaired responses to tibia mechanical loading in vivo and attenuation of loadinduced mechanosensing gene expression compared to control ***,control mice treated with a small molecule mechanomimetic,MS2 that activates the polycystin complex resulted in marked increases in femoral BMd and periosteal MAR compared to vehicle *** contrast,Pkd1/Wwtr1^(Oc-cKO)mice were resistant to the anabolic effects of *** findings suggest that PC1 and Wwtr1 form an anabolic mechanotransduction signaling complex that mediates mechanical loading responses and serves as a potential novel therapeutic target for treating osteoporosis.

An individual’s morphology is shaped by the environmental pressures it experiences,and the resulting morphological response is the culmination of both genetic factors and environmental(non-genetic)conditions experienced early in its life(*** plasticity).The role that phenotypic plasticity plays in shaping phenotypes is important,but evidence for its influence is often *** exposed female neonate diamond-backed watersnakes(Nerodia rhombifer)from populations experiencing different prey-size regimes to different feeding treatments to test the influence of phenotypic plasticity in shaping trophic *** found that snakes in a large-prey treatment from a population frequently encountering large prey exhibited a higher growth rate in body size than individuals in a small-prey treatment from the same *** pattern was not observed in snakes from a population that regularly encounters small *** also found that regardless of treatment,snakes from the smallprey population were smaller at birth than snakes from the large-prey population and remained so throughout the *** results suggest that the ability to plastically respond to environmental pressures may be population*** results also indicate a genetic predisposition towards larger body sizes in a population where large prey items are more common.

Background: There is evidence that sow heat stress(HS) during gestation affects fetal development with implications for impaired muscle growth. We have previously demonstrated that maternal HS during early to midgestation compromised muscle fibre hyperplasia in developing fetal pigs. Thus, we hypothesised these phenotypic changes are associated with a change in expression of genes regulating fetal skeletal muscle development and metabolism. To test this, at d 60 of gestation, RNA sequencing and immunohistochemistry were performed on fetal longissimus dorsi(Ld) muscle biopsies collected from pregnant gilts that had experienced either thermoneutral control(CON, 20 ℃, n = 7 gilts, 18 Ld samples) or controlled HS(cyclic 28 to 33 ℃, n = 8 gilts, 23 Ld samples)conditions for 3 ***: A total of 282 genes were differentially expressed between the HS and CON groups in female Ld muscles(false discovery rate(FdR) ≤ 0.05), whereas no differentially expressed genes were detected in male Ld muscles between the two groups(FdR > 0.05). Gestational HS increased the expression of genes associated with transcription corepressor activity, adipogenesis cascades, negative regulation of angiogenesis and pro-inflammatory signalling in female Ld muscles. Immunohistochemical analyses revealed a decreased muscle vascularity density in fetuses from HS group for both sexes compared to those from the CON group(P = 0.004).Conclusions: These results reveal gilt HS during early to mid-gestation altered gene expression profiles in fetal Ld muscles in a sexually dimorphic manner. The molecular responses, including transcription and angiogenesis repressions and enhanced adipogenesis cascades, were exclusively observed in females. However, the associated reductions in muscle vascularity were observed independently of sexes. Collectively this may indicate female fetal pigs are more adaptive to gestational HS in terms of gene expression changes, and/or there may be sexually dimorphic differences

Objective:Functional endoscopic sinus surgery is a commonly performed otolaryngologic procedure that often uses the microdebrider device for tissue *** the ubiquitous nature of the instrument,we sought to better define the patterns of device failure using the postmarket surveillance openFdA ***:The openFdA database was queried for all microdebrider‐related adverse events from January 1,2000 to November 1,*** information on the nature of device failure and any associated patient injury was *** not directly related to device failure were excluded from the ***:A total of 641 events were included in the *** most common device failure was overheating(n=348,54.3%),followed by material separation(n=173,27%),and inconsistent device activation(n=52,8.1%).Of the reported events,the vast majority did not result in patient harm(n=579,90.3%).On review of the remaining cases,only 24 events(3.7%)resulted in true harm to the patient,defined as a temporary or permanent injury or>30 min of additional anesthesia *** these cases,the need to reschedule surgical cases(n=5,0.8%),retained foreign body(n=5,0.8%),and thermal tissue injury(n=3,0.5%)were the most *** patients suffered an injury due to surgeon error unrelated to device malfunction(n=5,0.8%).Conclusions:Microdebrider device failures are extremely *** they do occur,less than 10%result in patient *** cases of patient harm related to microdebrider failure,preoperative testing of the device before use could prevent many of the reported malfunctions.

Most vascular plants interact with arbuscular mycorrhizal fungi （AMF） and are thereby provided significant advantages in nutrient acquisition, especially phosphate. The widespread retention of this symbiosis in vascular plants is testament to its importance and ancient origins. This highlight focuses on the reports of two of the first genes identified through for- ward genetic screens for AMF symbiotic mutants： Required for Abuscular Mycorrhization 1 （RAM1） and RAM2, which respectively encode a GRAS transcription factor and a glyc- erol-3-phosphate acyl-transferase （GPAT） that are required for the formation of fungal entry structures （hyphopodia） on the root surface.

Risk of colorectal cancer in patients with diverticular disease

Colorectal cancer constitutes an important burden on the healthcare system. Screening at-risk populations to reduce colorectal cancer-related morbidity and mortality has become part of good clinical practice. However, recommendations regarding subgroups of patients with diverticular disease are subject to controversy. Herein, we review the most recent literature regarding the prevalence of colorectal cancer in patients with diverticular disease, diverticulitis and uncomplicated diverticulitis. The recent literature does not identify diverticular disease as a long-term risk factor for colorectal cancer. However, the risk of colorectal cancer is increased in the short-term period after hospitalization related to diverticular disease. According to a recent systematic review and meta-analysis, the prevalence of colorectal cancer is 1.6% in patients with acute diverticulitis who underwent colonoscopy. The risk of having colorectal cancer after an episode of acute diverticulitis is 44-fold higher than that of an age-and gender-adjusted reference population. despite lower among patients with uncomplicated episode, the risk of colorectal cancer remains 40-fold higher in that subpopulation than that in the reference population. To conclude, the recent literature describes an increased risk of colorectal cancer among patients with acute diverticulitis compared to the reference population. Colonoscopy is therefore recommended in patients with diverticulitis to exclude colorectal cancer.

Strong, lightweight, and highly conductive CNT/Au/Cu wires from sputtering and electroplating methods

In this study, we present a 2-step deposition method via sputtering and electroplating that uses carbon nanotube(CNT) wires synthesized from a wet-spinning technique to produce high-performance CNT/Au/Cu composite wires. After the Au sputtering pre-treatment, the deposition of Cu on the CNT wires was found to be much more homogeneous due to improved wettability and reactivity of the wire surface. At different electrodeposition time, the mechanical strength of the CNT/Au/Cu composite wires could be as high as 0.74 GPa(~2 times stronger than metal wires) while their electrical conductivity could reach 4.65 × 10~5 S/cm(~80% of that for copper). More importantly, the CNT/Au/Cu composite wires with high CNT volume fraction are expected to be lightweight(up to 42% lower than Cu mass density), suggesting that our high-performance composite wires are a promising candidate to substitute conventional heavy metal wires in the future applications.
