Effects of Indole-3-Carbinol and Flavonoids Administered Separately and in Combination on Nitric Oxide Production and iNOS Expression in Rats

Beneficial effects of natural compounds are often attributed to modulation of NO production;however effects produced by plant extracts do not correlate with effects of purified components. The goal of our work was to study ability of flavonoids and indole-3-carbinol, as well as their combinations to modify NO production, iNOS gene and protein expression in rat tissues. Baicalein and luteolin decreased NO concentration in both intact and LPS-treated animals. Baicalein decreased iNOS gene expression. Luteolin decreased NO production in the liver and heart and number of iNOS-positive cells in the liver of LPS-treated animals. Combination of the two substances did not decrease the NO synthesis triggered by LPS, although it decreased iNOS gene expression. Quercetin decreased NO production in the heart, kidneys and blood of intact rats, but enhanced the LPS effect in testes, spleen and blood on NO production and iNOS protein expression in the liver. Indole-3-carbinol decreased NO concentration in the cerebellum, blood, lungs and skeletal muscles. The drug enhanced the LPS-triggered increase of NO production in all rat organs. It increased iNOS protein expression in intact liver;however it decreased the LPS-triggered outburst of the enzyme biosynthesis. Combination of indole3-carbinol with quercetin decreased NO production in LPS-treated animals however it slightly increased iNOS gene expression. Taken together our results suggest that modifications of NO level in tissues by a natural compound cannot be predicted from data about its effects on NOS expression or activity. Combination of substances can produce an effect differing from that of individual substances. This stresses importance of direct measurements of NO in the tissues.

AIM: To study short ds RNA oligonucleotides(si RNA)as a potent tool for artificially modulating gene expression of N-Myc down regulated gene 1(NDRG1) gene induced under different physiological conditions(Normoxia and hypoxia) modulating NDRG1 transcription, m RNA stability and translation. METHODS: A cell line established from a patient with glioblastoma multiforme. Plasmid DNA for transfections was prepared with the Endofree Plasmid Maxi kit. From plates containing 5 × 107 cells, nuclear extracts were prepared according to previous protocols. The p SUPERNDRG1 vectors were designed, two sequences were selected from the human NDRG1 c DNA(5'-GCATTATTGGCATGGGAAC-3' and 5'-ATGCAGAGTAACGTGGAAG-3'. reverse transcription polymerase chain reaction was performed using primers designed using published information on β-actin and hypoxia-inducible factor(HIF)-1α m RNA sequences in Gen Bank. NDRG1 m RNA and protein level expression results under different conditions of hypoxia or reoxygenation were compared to aerobic control conditions using the Mann-Whitney U test. Reoxygenation values were also compared to the NDRG1 levels after 24 h of hypoxia(P < 0.05 was considered significant).RESULTS: si RNA- and iodoacetate(IAA)-mediated downregulation of NDRG1 m RNA and protein expression in vitro in human glioblastoma cell lines showed a nearly complete inhibition of NDRG1 expression when compared to the results obtained due to the inhibitory role of glycolysis inhibitor IAA. Hypoxia responsive elements bound by nuclear HIF-1 in human glioblastoma cells in vitro under different oxygenation conditions and the clearly enhanced binding of nuclear extracts from glioblastoma cell samples exposed to extreme hypoxic conditions confirmed the HIF-1 Western blotting results. CONCLUSION: NDRG1 represents an additional diagnostic marker for brain tumor detection, due to the role of hypoxia in regulating this gene, and it canrepresent a potential target for tumor treatment in human glioblastoma. Over the last two centuries,the impact of the Human System has grown dramatically,becoming strongly dominant within the Earth System in many diferent ***,inequality,and population have increased extremely fast,especially since about 1950,threatening to overwhelm the many critical functions and ecosystems of the Earth *** in the Earth System,in turn,have important feedback efects on the Human System,with costly and potentially serious ***,current models do not incorporate these critical *** argue that in order to understand the dynamics of either system,Earth System Models must be coupled with Human System Models through bidirectional couplings representing the positive,negative,and delayed feedbacks that exist in the real *** particular,key Human System variables,such as demographics,inequality,economic growth,and migration,are not coupled with the Earth System but are instead driven by exogenous estimates,such as United Nations population *** makes current models likely to miss important feedbacks in the real Earth–Human system,especially those that may result in unexpected or counterintuitive outcomes,and thus requiring diferent policy interventions from current *** importance and imminence of sustainability challenges,the dominant role of the Human System in the Earth System,and the essential roles the Earth System plays for the Human System,all call for collaboration of natural scientists,social scientists,and engineers in multidisciplinary research and modeling to develop coupled Earth–Human system models for devising efective science-based policies and measures to beneit current and future generations.

AIM: To study the expression of carbonic anhydrase(CA) 9 in human hepatocellular carcinoma(HCC) ***: We studied CA9 protein, CA9 m RNA and hypoxia-inducible factor-1 alpha(HIF-1α) protein levels in Hep3 B cells exposed in different parallel approaches. In one of these approaches, HCC cells were exposed to extreme in vitro hypoxia(24 h 0.1% O2) without or with interleukin(IL)-1, IL-6, tumor necrosis factoralpha(TNF-α) and transforming growth factor-beta(TGF-β) stimulation for the same hypoxic exposure time or exposed to normoxic oxygenation conditions without or with cytokine ***: The tumour cell line analysed showed a strong hypoxic CA9 m RNA expression pattern in response to prolonged severe hypoxia with cell-line specific patterns and a marked induction of CA9 protein in response to severe hypoxia. These results were paralleled by the results for HIF-1α protein under identical oxygenation conditions with a similar expression tendency to that displayed during the CA9 protein expression experimental series. Continuous stimulation with the cytokines, IL-1, IL-6, TNF-α and TGF-β, under normoxic conditions significantly increased the carbonic anhydrase 9 expression level at both the protein and m RNA level, almost doubling the CA9 m RNA and CA9 and HIF-1α protein expression levels found under hypoxia. The findings from these experiments indicated that hypoxia is a positive regulator of CA9 expression in HCC, and the four signal transduction pathways, IL-1, IL-6, TNF-α and TGF-β, positively influence CA9 expression under both normoxic and hypoxic ***: These findings may potentially be considered in the design of anti- cancer therapeutic approaches involving hypoxia-induced or cytokine stimulatory effects on expression. Retroperitoneal hemorrhage is a possible complication that has been noticed in COVID-19 patients on anticoagulant therapy. The data related to treatment and outcomes in this condition are scarce. We present a case series of four COVID-19 patients with lumbar artery bleeding, who were treated with transcatheter arterial embolization using embolization particles. The procedure was executed without any complications, and hemostasis was achieved in all four patients. Despite the patients survived the procedure, one patient died due to respiratory failure three weeks after the procedure, and one died due to hypovolemic shock two days later. Considering the ongoing state of COVID-19 pandemic, it is essential to be aware of transcatheter arterial embolization as a safe and effective procedure for treating retroperitoneal hemorrhage in COVID-19 patients.

在现代信息技术中，应用数据库进行交流存在着一个极为重要的问题，即用户的观点差异问题。为了能够应用信息系统进行交流，传统的系统要求用户具有专门的训练。这种专门训练往往是长期的，甚至必须具有良好的计算机基础知识和勤奋好学的性格。因此，无论这些信息系统多么先进和有益，上述原因足以使一些人望而却步，放弃使用计算机信息资源。基于这一问题，贝尔格莱德大学矿业和地质学院开发了一个连接软件，它可弥补上述传统用户交流手段的不足。本文介绍了这一软件的结构，即沉积学数据的录入方法和应用数据库进行图解式交流的连接软件的原理。

In this study, the chemical composition, antioxidant, and hypoglycemic activity of Rosa arvensis Huds. leaves and fruits collected from Zlatibor, Željin, Veliki Krš mountains and Petnica village (Serbia) were analysed. Phenolic and vitamin profiles, as well as in vitro antioxidant potential and enzyme inhibitory activity on α-amylase and α-glucosidase, were studied. Phytochemical analysis showed higher phenolic content in leaf extracts than in those obtained from fruits. Gallic, chlorogenic and syringic acids were the most abundant phenolic acids, while catechin was the major flavonoid compound. Furthermore, the highest level of α-tocopherol (1.94 mg/100 g fw) in achenes and ascorbic acid (9.60 mg/100 g fw) in hypanthium were quantified in the fruits sampled on the Veliki Krš Mt. High antioxidant capacity was observed for the leaf extracts. It was highly correlated with their chemical composition and primarily referred to the catechin content (r > 0.95, p −0.97, p 0.80, p 0.49, p < 0.01). Among the tested R. arvensis samples, those from Zlatibor Mt. contained the highest amount of target compounds and expressed the highest biological activity. The obtained results suggest that R. arvensis extracts present a new source of bioactive compounds that could be implemented as novel food ingredients.
