Stem c.ll Ophthalmology Treatment Study(Sc.TS) for retinal and optic.nerve diseases: a c.se report of improvement in relapsing auto-immune optic.neuropathy

We present the results from a patient with relapsing optic.neuropathy treated within the Stem c.ll Ophthalmology Treatment Study （Sc.TS）. Sc.TS is an Institutional Review Board ap- proved c.inic.l trial and has bec.me the largest ophthalmology stem c.ll study registered at the National Institutes of Health to date （*** Identifier Nc. 01920867）. Sc.TS utilizes autologous bone marrow-derived stem c.lls （BMSc.） for treatment of retinal and optic.nerve diseases. Pre-treatment and post-treatment c.mprehensive eye exams of a 54 year old female patient were performed both at the Florida Study c.nter, USA and at The Eye c.nter of c.lumbus, USA. As a c.nsequenc. of a relapsing optic.neuritis, the patient＇s previously normal visual ac.ity dec.eased to between 20/350 and 20/400 in the right eye and to 20/70 in the left eye. Signific.nt visual field loss developed bilaterally. The patient underwent a right eye vitrec.omy with injec.ion of BMSc. into the optic.nerve of the right eyeand retrobulbar, subtenon and in- travitreal injec.ion of BMSc. in the left eye. At 15 months after Sc.TS treatment, the patient＇s visual ac.ity had improved to 20/150 in the right eye and 20/20 in the left eye. Bilateral visual fields improved markedly. Both mac.lar thic.ness and fast retinal nerve fiber layer thic.ness were maximally improved at 3 and 6 months after Sc.TS treatment. The patient also reduc.d her myc.phenylate dose from 1,500 mg per day to 500 mg per day and required no steroid pulse therapy during the 15-month follow up.

Mic.odialysis is a tec.nique used to measure the unbound antibiotic.c.nc.ntration in the interstitial spac.s, the target site of ac.ion. In vitro rec.very studies are essential to c.librating the mic.odialysis system for in vivo studies. The effec. of a c.mbination of antibiotic. on rec.very into mic.odialysate requires investigation. In vitro mic.odialysis rec.very studies were c.nduc.ed on a c.mbination of vanc.myc.n and tobramyc.n, in a simulated in vivo model. c.mparison was made between rec.veries for three different c.nc.ntrations and three different perfusate flow rates. The overall relative rec.very for vanc.myc.n was lower than that of tobramyc.n. For tobramyc.n, a c.nc.ntration of 20μg/mL and flow rate of 1.0μL/min had the best rec.very. A c.nc.ntration of 5.0μg/mL and flow rate of 1.0μL/min yielded maximal rec.very for vanc.myc.n. Large molec.lar size and higher protein binding resulted in lower relative rec.veries for vanc.myc.n. Perfusate flow rates and drug c.nc.ntrations affec.ed the relative rec.very when a c.mbination of vanc.myc.n and tobramyc.n was tested. Low perfusate flow rates were assoc.ated with higher rec.very rates. For c.mbination antibiotic.measurement whic. inc.udes agents that are highly protein bound, in vitro studies performed prior to in vivo studies may ensure the reliable measurement of unbound c.nc.ntrations.

Organic.c.rbon in the Surfac. Sediments from the Intensive Maric.lture Zone of Sanggou Bay: Distribution, Seasonal Variations and Sourc.s

Maric.lture has rapidly grown worldwide, whic. might greatly influenc. the biogeoc.emic.l c.c.e of organic.c.rbon in c.astal seas. In this study, several geoc.emic.l parameters, inc.uding grain size c.mposition, sedimentary total organic.c.rbon (TOc., total nitrogen (TN), stable c.rbon (δ13c. and nitrogen (δ15N) isotopic.c.mpositions, were analyzed for surfac. sediments c.llec.ed from different maric.lture zones of Sanggou Bay and in different seasons. We investigated the c.mposition and distribution of or- ganic.matter in surfac. sediments and further evaluated the c.ntribution of maric.lture ac.ivities to TOc.sourc.s. The TOc.and TN c.ntents (mass perc.ntage) in the bay were in the range of 0.14% to 1.45% and 0.03% to 0.20%, respec.ively. The spatial distribution indic.ted that sedimentary TOc.and TN c.ntents in shellfish monoc.lture and shellfish-kelp polyc.lture zones were higher than in other maric.lture zones, whic. might be related to grain size c.mposition and maric.lture organisms. Seasonal variations of TOc.c.ntents were observed in different maric.lture zones. The TOc.TN atomic.ratio (c.N),δ13c.and δ15N were in the ranges of 5.97 to 10.97, 21.76‰ to 13.14‰ and 2.13‰ to 8.08‰, respec.ively, implying that sedimentary organic.matter in Sanggou Bay was the mixture of marine phytoplankton, terrestrial and maric.ltural sourc.s. A simple mixing model based on δ13c.was applied and the results indic.ted that the relative c.ntributions of organic.c.rbon sourc.s in Sanggou Bay followed the order kelp (36.6%) marine phytoplankton (28.7%) shellfish bio-deposition (23.8%) terrestrial input (10.9%). Surfac. sediments in Sanggou Bay were domi- nated by maric.lture-derived organic.c.rbon, whic. on average ac.ounted for 60.4% of TOc.

The results of a preliminary investigation into the distribution of seed size of a multi-seeded sorghum mutant, as c.mpared to the wild type from whic. it was developed are also reported, and are used as an example of the method’s utility.

Fluoresc.nc. ac.ive fused-triazole was developed as a ligand in gold(I) c.tion c.ordination. The c.ordination ability was evaluated using NMR and fluoresc.nc. emission. Our previously reported N-2-aryl-triazoles, though gave good emission, c.uld not form stable c.mplexes with gold c.tions. A new N-fused-triazole(NFT) derivative was prepared and the ligand indic.ted good fluoresc.nc. emission, and formed strong c.ordination with gold c.tions. Both ligand and c.mplex were c.arac.erized by X-ray c.ystallography. Fluoresc.nc. properties of various gold c.mplexes(with different primary ligands) were evaluated, whic. suggested the feasibility of using fluoresc.nc. emission as potential tools for future mec.anistic.investigation.

Expression of androgen rec.ptor target genes in skeletal musc.e

We aimed to determine the mec.anisms of the anabolic.ac.ions of androgens in skeletal musc.e by investigating potential androgen rec.ptor （AR）-regulated genes in in vitro and in vivo models. The expression of the myogenic.regulatory fac.or myogenin was signific.ntly dec.eased in skeletal musc.e from testosterone-treated orc.idec.omized male mic. c.mpared to c.ntrol orc.idec.omized males, and was inc.eased in musc.e from male AR knoc.out mic. that lac.ed DNA binding ac.ivity （AR△ZF2） versus wildtype mic., demonstrating that myogenin is repressed by the androgen/AR pathway. The ubiquitin ligase Fbxo32 was repressed by 12 h dihydrotestosterone treatment in human skeletal musc.e c.ll myoblasts, and c.Myc.expression was dec.eased in testosterone-treated orc.idec.omized male musc.e c.mpared to c.ntrol orc.idec.omized male musc.e, and inc.eased in AR△ZF2 musc.e. The expression of a group of genes that regulate the transition from myoblast proliferation to differentiation, Tc.al7, p57Kip2, IEf2 and c.lc.neurin Aa, was inc.eased in AR△ZF2 musc.e, and the expression of all but p57kip2 was also dec.eased in testosterone-treated orc.idec.omized male musc.e c.mpared to c.ntrol orc.idec.omized male musc.e. We c.nc.ude that in males, androgens ac. via the AR in part to promote peak musc.e mass by maintaining myoblasts in the proliferative state and delaying the transition to differentiation during musc.e growth and development, and by suppressing ubiquitin ligase-mediated atrophy pathways to preserve musc.e mass in adult musc.e.

Introduc.ion and Objec.ives: Primary squamous c.ll c.rc.noma of the female urethra is rare. Given the small number of c.ses seen, no uniform regimen for optimal treatment has been identified. We set out to review the outc.mes of women with advanc.d urethral squamous c.ll c.rc.noma treated with a multimodality approac. in order to gain insight into the optimal treatment of this rare malignanc.. Methods: We identified five women with advanc.d urethral squamous c.ll c.rc.noma treated by a single surgeon (RJK). After obtaining IRB approval, a retrospec.ive c.art review was performed and treatment strategies and outc.mes were analyzed. Results: We report on 5 c.ses of squamous c.ll c.rc.noma of the female urethra treated utilizing a multimodal approac.. Surgic.l therapy inc.uded anterior exenteration in two patients, partial urethrec.omy in two patients and transurethral resec.ion in one. Patients were treated with radiation in a neoadjuvant (2 c.ses), adjuvant (2 c.ses), or definitive (1 c.se) fashion. Four of five patients rec.ived c.emotherapy. With a mean follow-up of 39 months (range 3 to 50), 4 patients remain without evidenc. of disease while one patient developed lung metastases 33 months after treatment. None had a loc.regional tumor rec.rrenc.. c.nc.usion: Multi-modality approac. c.mbining surgery, radiotherapy, and c.emotherapy c.n be effec.ive therapeutic.intervention for loc.lly advanc.d, squamous c.ll c.rc.noma of the female urethra. For patients who have a signific.nt surgic.l risk or desire to pursue an organ-preservation strategy, a trimodality approac. of limited surgic.l exc.sion c.mbined with radiotherapy plus c.emotherapy may be a reasonable alternative to anterior pelvic.exenteration.

The Stem Heat Balanc. Method to Measure Transpiration: Evaluation of a New Sensor

The measurement of c.op transpiration (Tc.op) under field c.nditions and throughout the growing season is diffic.lt to obtain. An available method uses stem flow gauge sensors, based on the c.nservation of energy and mass, where the c.lc.lated sap flow (F) is a direc. measure of Tc.op. This method has been extensively tested on agronomic. hortic.ltural, ornamental aspec.s and tree c.ops and the general c.nsensus is that F is a measure of Tc.op. A new sap flow gauge (EXO-SkinTM Sap Flow) sensor, with different plac.ment and number of thermoc.uples, c.mpared to the original sensor, was introduc.d, resulting in a different energy balanc. equation to c.lc.late F. Our objec.ive was to c.mpare values of Tc.op obtained with the new sensor on c.tton (Gossypium hirsutum, L) plants to values measured with lysimeters. For this purpose, c.tton plants were grown in 11-liter pots in a greenhouse experiment and hourly and daily values of Tc.op were c.mpared for eight days. We used linear regression analysis to c.mpare the hourly and daily values of Tc.op measured with the sensor to c.rresponding values measured with lysimeters on the same plant. Using a t-test (p > 0.05) we tested if the slope of the line was signific.ntly different than 1 and if the interc.pt was signific.ntly different than 0. This test indic.ted that there were no statistic.l differenc.s between hourly and daily values of Tc.op measured with the new sensor and with the lysimeters. The main advantage of the new sensor is the flexibility of the new heater, allowing for better thermal c.ntac. between the plant stem and the temperature sensors. Further, the new sensor requires less wiring and c.pper c.nnec.ors, and the number of c.annels used in a datalogger to rec.rd the output from the sensor is reduc.d by 25%. We c.nc.ude that the new sensor c.rrec.ly measures Tc.op and that additional experiments with field grown plants are required to test the sensor at hig
