Differences in characteristics of patients with and without known risk factors for hepatocellular carcinoma in the United States

AIM： To examine the clinical characteristics of a subgroup of patients with hepatocellular carcinoma （hCC） and compare them to those with known risk factors. METhODS： We used the hCC database of 306 patients seen at our institution from January 1, 1995 to December 31, 2001. Of the 306 patients, 63 （20%, group 1） had no known risk factors （hepatitis C virus, hepatitis B virus, alcohol, hemochromatosis or cirrhosis from any cause） and 243 （group 2） had one or more risk factors. RESULTS： The median age was similar in both groups, but there were disproportionate numbers of younger （〈 30 years old）, older （〉 80 years） patients, women （33% vs 18%）, and Caucasians （81% vs 52%） in group 1 as compared to group 2. There were fewer Asians （2% vs 11%） and African Americans （13% vs 27%） in group 1. Abdominal pain （70% vs 37%） was more common while gastrointestinal bleeding （0% vs 11%） and ascites （4% vs17%） were less common in group i compared to group 2. Group 1 had larger tumor burden （median size 9.4 cm vs 5.7 cm） at the time of presentation, but there were no differences in the site （right, left or bilateral lesions）, or number of tumors between the two groups. CONCLUSION： hCC patients without identifiable risk factors have different characteristics and clinical presentation compared to those with known risk *** of cirrhosis and larger tumor burden may explain the differences in the presenting symptoms.

The purpose is to evaluate the feasibility of imaging mouse brain with high resolution ultrasound (hiRes US), and generation of mouse brain tumor (glioma) model under hiRes US guided implantation of glioma cells. Normal mouse brain was imaged with 30 Mhz hiRes US in six pups and ten adult nude mice. Glioma model was developed by injecting human glioma cells (5 × 105), U-87MG-luc, under hiRes US guidance, in three pups and five adult mice. Bioluminescence imaging, magnetic resonance imaging, and hiRes US were used for in vivo tumor imaging. In addition, brain imaging ex vivo with hiRes US were also performed in three tumor bearing and five normal brains. The brain parenchyma was seen as a homogeneous low echo on hiRes US without locoregional echo differences. An inverted U shaped linear echo structure (fissure) differentiated the telencephalon from the diencephalon. Bilaterally budding structure at the base of the skull represented the trigeminal nerve. The inserted needle, used to implant the glioma cells, was seen as a high linear echogenic reflection. Brain tumor on ex vivo hiRes US was well demarcated, homogeneous and hyperechoic compared to the surrounding healthy brain. In conclusion, imaging the brain with hiRes US in small animal model like mouse is possible and convenient. Real-time guiding is possible to perform any intervention from tumor implantation to percutaneous therapeutic procedures. Ex vivo, hiRes US is extremely useful to study the detailed anatomical features.

Many aphid species exhibit a variation in reproductive mode which is influenced by winter climate regimes,with cyclical parthenogenetic (CP)lines dominating in cold winter areas (because they produce cold-resistant eggs)and obligate parthenogenetic (OP) ones in mild winter regions (because of their parthenogenetic overwintering).Genetic studies on several aphid species have shown that the OP trait can be transmitted during sexual events involving the 2 types of *** genetic system could be considered as a local safeguarding mechanism for OP alleles in case severe frost would have killed all parthenogenetically overwintering ***,this strategy would only be efficient in restoring local polymorphism in breeding systems if the newly hatched OP recombinants remain competitive over their CP *** this study we compared egg hatching sequences of CP and OP F1 clones from several crosses obtained for 2 cereal aphid species,Sitobion avenae (constant 5℃,8 h of light)and Rhopalosiphum padi (winter outdoor conditions).For ***,we obtained F1 offspring from 6 crosses, involving 4 clones while in *** F1 were obtained from 11 crosses involving 14 clones. We showed that in both species proportions of OP clones were higher in the first half of the progeny relative to the second *** addition,F1 OP clones hatched in the mean about a week earlier than their CP sibs,which gives them a demographic advantage at the start of the growth *** then discussed the consequences of this fitness advantage for the maintenance and spread of the OP trait in aphid populations.

Droplet digital PCR for quantification of ITGA6 in a stool mRNA assay for the detection of colorectal cancers

AIM To investigate the use of droplet digital polymerase chain reaction(dd PCR) for detecting host m RNA markers in stools as a non-invasive test for colorectal cancer *** dd PCR and quantitative PCR were compared side by side for their performance in the detection of ITGA6 and ITGA6 A transcripts in stool samples obtained from patients with various types of colorectal lesions(advanced adenomas and stage Ⅱ-Ⅳ colorectal cancers) and control(patients displaying no pathological findings) using duplex Taq Man reactions for both methods. ITGA6 and ITGA6 A were chosen for this proof-of-concept study based on their relative medium and low abundance in stool samples, respectively, as established in a previous *** We found that the dd PCR and q PCR methods per-formed equally well in this Taq Man duplex assay for the detection of ITGA6 and ITGA6 A transcripts in stools of patients with colorectal lesions. For ITGA6, receiver operating characteristic(ROC) curve analysis showed comparable areas under the curve of 0.91(P he prediction of advanced adenomas and colorectal cancers, respectively. ITGA6 A, which was detected at very low levels in control patients, was found to be significantly elevated(over 40 times) in stage Ⅱ and Ⅲ colorectal cancers(P he two sets of data revealed a strong correlation of the copy numbers obtained by dd PCR and q PCR for both ITGA6 and ITGA6 *** We found that ITGA6 and ITGA6 A detection in stools of patients with colorectal cancers with dd PCR is comparable to that of q PCR using Taq Man assays.

Iron （Fe） homeostasis is integrated with the production of reactive oxygen species （ROS）, and distribution at the root tip participates in the control of root growth. Excess Fe increases ferritin abundance, enabling the storage of Fe, which contributes to protection of plants against Fe-induced oxidative stress. AtFerl and AtFer3 are the two ferritin genes expressed in the meristematic zone, pericycle and endodermis of theAra- bidopsis thaliana root, and it is in these regions that we observe Fe stained dots. This staining disappears in the triple ferl-3-4 ferritin mutant. Fe excess decreases primary root length in the same way in wild-type and in ferl-3-4 mutant. In contrast, the Fe-mediated decrease of lateral root （LR） length and density is enhanced in ferl-3-4 plants due to a defect in LR emergence. We observe that this interaction between excess Fe, ferritin, and root system architecture （RSA） is in part mediated by the h_2O_2/O2·^- balance between the root cell proliferation and differentiation zones regulated by the UPB1 transcription factor. Meristem size is also decreased in response to Fe excess in ferritin mutant plants, implicating cell cycle arrest mediated by the ROS-activated SMR5/SMR7 cyclin-dependent kinase inhibitors pathway in the interaction between Fe and RSA.

The SMYD(SET and MYND domain)family of lysine methyltransferases(KMTs)plays pivotal roles in various cellular processes,including gene expression regulation and DNA damage *** identified as genuine histone methyltransferases,specific members of this family have recently been shown to methylate non-histone proteins such as p53,VEGFR,and the retinoblastoma tumor suppressor(pRb).To gain further functional insights into this family of KMTs,we generated the protein interaction network for three different human SMYD proteins(SMYD2,SMYD3,and SMYD5).Characterization of each SMYD protein network revealed that they associate with both shared and unique sets of *** those,we found that hSP90 and several of its co-chaperones interact specifically with the tetratrico peptide repeat(TPR)-containing SMYD2 and ***,using proteomic and biochemical techniques,we provide evidence that SMYD2 methylates K209 and K615 on hSP90 nucleotide-binding and dimerization domains,*** addition,we found that each methylation site displays unique reactivity in regard to the presence of hSP90 co-chaperones,ph,and demethylation by the lysine amine oxidase LSD1,suggesting that alternative mechanisms control hSP90 methylation by ***,this study highlights the ability of SMYD proteins to form unique protein complexes that may underlie their various biological functions and the SMYD2-mediated methylation of the key molecular chaperone hSP90.
