Chronic exposure to excess iron promotes EMT and cancer via p53 loss in pancreatic cancer

Based on the evidence that hemochromatosis, an iron-overload disease, drives hepatocellular carcinoma, we hypothesized that chronic exposure to excess iron, either due to genetic or environmental causes, predisposes an individual to cancer. Using pancreatic cancer as our primary focus, we employed cell culture studies to interrogate the connection between excess iron and cancer, and combined in vitro and in vivo studies to explore the connection further. Ferric ammonium citrate was used as an exogenous iron source. Chronic exposure to excess iron induced epithelial-mesenchymal transition(EMT) in normal and cancer cell lines, loss of p53, and suppression of p53 transcriptional activity evidenced from decreased expression of p53 target genes(p21, cyclin D1, Bax, SLC7A11). To further extrapolate our cell culture data, we generated EL-KrasG12D( EL-Kras) mouse(pancreatic neoplastic mouse model) expressing Hfe+/+ and Hfe-/- genetic background. p53 target gene expression decreased in EL-Kras/Hfe-/- mouse pancreas compared to EL-Kras/Hfe+/+ mouse pancreas. Interestingly, the incidence of acinar-to-ductal metaplasia and cystic pancreatic neoplasms(CpN) decreased in EL-Kras/Hfe-/- mice, but the CpNs that did develop were larger in these mice than in EL-Kras/Hfe+/+ mice. In conclusion, these in vitro and in vivo studies support a potential role for chronic exposure to excess iron as a promoter of more aggressive disease via p53 loss and SLC7A11 upregulation within pancreatic epithelial cells.

Due to the dynamic nature and node mobility,assuring the security of Mobile Ad-hoc Networks(MANET)is one of the difficult and challenging tasks *** MANET,the Intrusion Detection System(IDS)is crucial because it aids in the identification and detection of malicious attacks that impair the network’s regular *** machine learning and deep learning methodologies are used for this purpose in the conventional works to ensure increased security of ***,it still has significant flaws,including increased algorithmic complexity,lower system performance,and a higher rate of ***,the goal of this paper is to create an intelligent IDS framework for significantly enhancing MANET security through the use of deep learning ***,the min-max normalization model is applied to preprocess the given cyber-attack datasets for normalizing the attributes or fields,which increases the overall intrusion detection performance of ***,a novel Adaptive Marine predator Optimization Algorithm(AOMA)is implemented to choose the optimal features for improving the speed and intrusion detection performance of ***,the Deep Supervise Learning Classification(DSLC)mechanism is utilized to predict and categorize the type of intrusion based on proper learning and training *** evaluation,the performance and results of the proposed AOMA-DSLC based IDS methodology is validated and compared using various performance measures and benchmarking datasets.

printed circuit boards(pCBs)are representative composite materials,and their high-quality drilling machining remains a persistent challenge in the *** finishing of the cutting edge of a microdrill is crucial to drill performance in machining fine-quality holes with a prolonged tool *** miniature size involving submicron scale geometric dimensions,a complex flute shape,and low fracture toughness makes the cutting edge of microdrills susceptible to breakage and has been the primary limiting factor in edge preparation for *** this study,a newly developed cutting edge preparation method for microdrills was tested experimentally on electronic printed circuit *** proposed method,namely,shear thickening polishing,limited the cutting edge burrs and chipping on the cutting edge,and this in turn transformed the cutting edge’s radius from being sharp to ***,the edge–edge radius could be regulated by adjusting the processing *** drilling experiments were conducted to investigate the influence of different cutting edge radii on wear,hole position accuracy,nail head value,and hole wall *** proposed approach showed 20%enhancement in hole position accuracy,33%reduction in the nail head value,and 19%reduction in hole wall roughness compared with the original ***,a threshold is needed;without it,excessive shear thickening polishing will result in a blunt edge,which may accelerate the wear of the *** was identified as the primary factor that reduced hole *** study indicates that in printed circuit board machining,microdrills should effectively eliminate grinding defects and maintain the sharpness of the cutting edge as much as possible to obtain excellent drilling ***,shear thickening polishing is a promising method for cutting edge preparation of *** research and optimization can lead to additional improvements in microdrill performance and contribute to the conti

Enhanced entropy generation and heat transfer characteristics of magnetic nano-encapsulated phase change materials in latent heat thermal energy storage systems

The objective of the current study is to investigate the importance of entropy generation and thermal radiation on the patterns of velocity,isentropic lines,and temperature contours within a thermal energy storage device filled with magnetic nanoencapsulated phase change materials(NEpCMs).The versatile finite element method(FEM)is implemented to numerically solve the governing *** effects of various parameters,including the viscosity parameter,ranging from 1 to 3,the thermal conductivity parameter,ranging from 1 to 3,the Rayleigh parameter,ranging from 102 to 3×10^(2),the radiation number,ranging from 0.1 to 0.5,the fusion temperature,ranging from 1.0 to 1.2,the volume fraction of NEpCMs,ranging from 2%to 6%,the Stefan number,ranging from 1 to 5,the magnetic number,ranging from 0.1 to 0.5,and the irreversibility parameter,ranging from 0.1 to 0.5,are examined in detail on the temperature contours,isentropic lines,heat capacity ratio,and velocity ***,the heat transfer rates at both the cold and hot walls are analyzed,and the findings are presented *** results indicate that the time taken by the NEpCMs to transition from solid to liquid is prolonged inside the chamber region as the fusion temperatureθf ***,the contours of the heat capacity ratio Cr decrease with the increase in the Stefan number Ste.

A Hybrid Optimization Approach of Single point Incremental Sheet Forming of AISI 316L Stainless Steel Using Grey Relation Analysis Coupled with principal Component Analysiss

We investigated the parametric optimization on incremental sheet forming of stainless steel using Grey Relational Analysis(GRA) coupled with principal Component Analysis(pCA). AISI 316L stainless steel sheets were used to develop double wall angle pyramid with aid of tungsten carbide tool. GRA coupled with pCA was used to plan the experiment conditions. Control factors such as Tool Diameter(TD), Step Depth(SD), Bottom Wall Angle(BWA), Feed Rate(FR) and Spindle Speed(SS) on Top Wall Angle(TWA) and Top Wall Angle Surface Roughness(TWASR) have been studied. Wall angle increases with increasing tool diameter due to large contact area between tool and workpiece. As the step depth, feed rate and spindle speed increase,TWASR decreases with increasing tool diameter. As the step depth increasing, the hydrostatic stress is raised causing severe cracks in the deformed surface. Hence it was concluded that the proposed hybrid method was suitable for optimizing the factors and response.

Use of magnesium is the need of the hour due to its low density as well as its high strength-to-weight and stiffness-to-weight ratio *** study focuses on the effectiveness of liquid nitrogen(LN_(2))assisted cryogenic machining on the surface integrity(SI)characteristics of AZ91 magnesium *** milling using uncoated carbide inserts have been performed under liquid nitrogen(LN_(2))assisted cryogenic condition and compared with conventional(dry)*** are performed using machining parameters in terms of cutting speeds of 325,475,625 m/min,feed rates of 0.05,0.1,0.15 mm/teeth and depth of cuts of 0.5,1,1.5 mm *** significant surface integrity characteristics such as surface roughness,microhardness,microstructure,and residual stresses have been *** of SI characteristics with respect to milling parameters have been identified using statistical technique such as ANOVA and signal-to-noise(S/N)ratio ***,the multi criteria decision making(MCDM)techniques such as additive ratio assessment method(ARAS)and complex proportional assessment(COpRAS)have been utilized to identify the optimal conditions for milling AZ91 magnesium alloy under both dry and cryogenic *** of LN_(2)during machining,resulted in reduction in machining temperature by upto 29%with a temperature drop from 251.2℃under dry condition to 178.5℃in cryogenic *** showed the advantage of performing cryogenic milling in improving the surface integrity to a significant *** machining considerably minimized the roughness by upto 28%and maximised the microhardness by upto 23%,when compared to dry *** speed has caused significant impact on surface roughness(95.33%-dry,92.92%-cryogenic)and surface microhardness(80.33%-dry,82.15%-cryogenic).Due to the reduction in machining temperature,cryogenic condition resulted in compressive residual stresses(maximumσ║=-113 Mpa)on the alloy *** indicate no

Background:Hard mineral and acid salt deposits that clump together in concentrated urine are known as kidney stones.A complicated chain of physicochemical events,including hyper saturation,nucleation,growth,aggrega-tion,and retention in the urinary tract,leads to urolithiasis,the third most prevalent urinary tract *** traditional medications and therapies are less successful in treating urolithiasis and frequently cause ***:The present study is focused on evaluating the anti-urolithiasis activities of the polyherbal formulation prepared from Spinacia oleracea *** Coriandrum sativum ***:To evaluating the anti-urolithiasis activities of the polyherbal formulation prepared from *** and *** seeds against oxalate crystals by employing an in vitro inhibition,aggregation,and nucleation assay,employing a spectrophotometer followed by microscopic observation,and further monitored by egg semiperme-able membrane model,using cystone as a standard ***:In the present work,the alkaloids,glycosides,flavonoids,glycosides,phenols,saponins,steroids,tannins,terpenoids,and triterpenoids were found in the extracts of *** and *** in vitro assays in the present study clearly indicated that all the mixture extracts readily prevented crystal nucleation,growth and *** aqueous extract of the mixture inhibited significant inhibitory potential against the crystallization ***:It can be said that the phyto-constituents enriched homemade preparation possibly will be used as a medication for the management of kidney stones by dissolving the oxalate stone in the kidney.

On the functions of astrocyte-mediated neuronal slow inward currents
