Backgr.und: Inter.r.tation of par.thyr.id hor.one (PTH) measur.ments r.quir.s an under.tanding of its str.ctur.l confor.ation. PTH r.flects calcium intake and vitamin D status but is commonly measur.d in the fasted state using 2nd gener.tion assays, known to cr.ss-r.act with PTH 7-84 fr.gments, which is not obser.ed with 3r. gener.tion assays. The objective was to deter.ine if plasma PTH in the fed and non-fed state differ.while contr.lling for.diur.al r.ythm and the gener.tion of PTH assay. Methods: Blood was sampled, 2 d apar., fr.m Spr.gue-Dawley r.ts (30 males and 30 females) in both fed and non-fed states at 20 wk of age (weight: 470.2 ± 23.2 g) for.measur.ment of ionized calcium (iCa), total intact (INT) and bioactive 1-84 (BIO) PTH. Differ.nces between gr.ups wer. tested using a GLIMMIX model with sex and feeding state as fixed effects and individual r.ts as a r.ndom effect. r.sults: Females had a lower.iCa than males (F: 1.43 ± 0.01 vs M: 1.46 ± 0.01 mmol/L, P = 0.03). In males and females, ther. was no differ.nce between fed and non-fed gr.ups when PTH was assessed using the INT PTH assay (M Fed: 21.6 ± 1.3 vs M Non-fed: 22.6 ± 2.3 pmol/L, P = 0.59;F Fed: 19.3 ± 1.5 vs F Non-fed: 22.8 ± 2.9 pmol/L, P = 0.39). However. in females only, PTH measur.d using the BIO PTH, was significantly lower.in the fed gr.up ver.us the non-fed (Fed: 8.4 ± 0.8 vs Non-fed: 16.7 ± 3.4 pmol/L, P = 0.05). Conclusions:

Backgr.und: Amazonian r.ver.ide communities consume lar.e quantities of fr.shwater.fish, compar.ble to mar.ne fish consumption of Inuit, Scandinavian and Japanese populations. Few studies have consider.d the r.lation of high fr.shwater.fish consumption and intake of omega-3 fatty acids (FA). Objective: The objective of the pr.sent study was to deter.ine the pr.file of the concentr.tions of plasma phospholipid FAs and its r.lation with fr.shwater.fish intake in 12 r.ver.ide communities in the Tapajós r.ver.basin (State of Par., Br.zilian Amazon). Design: This cr.ss-sectional study included 333 adults (15 - 86 year. old). Fish meal fr.quency was deter.ined using a 7-day inter.iew-administer.d dietar. r.call questionnair.. Fish wer. categor.zed as piscivor.us and non-piscivor.us fish on tr.phic level. Plasma phospholipid FAs wer. measur.d by capillar. gas-liquid chr.matogr.phy. r.sults: Par.icipants consumed 5 to 6 fish meals a week, with twice as many non-piscivor.us fish than piscivor.us fish (4.4 fish/week vs 2.2 fish/week, r.spectively). The omega-3 FA levels in plasma phospholipids wer. low (EPA + DHA = 31.21 mg/L;%EPA + DHA = 2.59%). Omega-3 FAs incr.ased with fr.quency of fish intake, and par.icular.y with piscivor.us fish, contr.lling for.sociodemogr.phic, anthr.pometr.c and lifestyle char.cter.stics. DHA levels wer. gr.ater.in women than in men. Conclusions: Omega-3 FA in this Amazonian population incr.ased with fish consumption, but although they consumed fr.shwater.fish almost daily, the concentr.tions of omega-3 FA wer. r.latively low and compar.ble to fish-eater.communities for.which fish is not a dietar. mainstay. It is possible that nutr.ents pr.sent in mar.ne, but not in cer.ain fr.shwater.fish species, may facilitate absor.tion of omega-3 FA. Sex and/or.gender.differ.nces must be taken into account when assessing the r.lationship between fish consumption and plasma phospholipid omega-3 FA levels.
