AIM: To analyze the prevalence of germline MLH1 and MSH2 gene mutations and evaluate the clinical characteristics of Hungarian hereditary non-polyposis colorectal cancer (HNPCC) families. METHODS: Thirty-six kindreds were tested for mutations using conformation sensitive gel electrophoreses, direct sequencing and also screening for genomic rearrangements applying multiplex ligation-dependent probe amplifi cation (MLPA). RESULTS: Eighteen germline mutations (50%) were identifi ed, 9 in MLH1 and 9 in MSH2. Sixteen of these sequence alterations were considered pathogenic, the remaining two were non-conservative missense alterations occurring at highly conserved functional motifs. The majority of the defi nite pathogenic mutations (81%, 13/16) were found in families fulfilling the stringent Amsterdam Ⅰ/Ⅱ criteria, including three rearrangements revealed by MLPA (two in MSH2 and one in MLH1). However, in three out of sixteen HNPCC-suspected families (19%), a disease-causing alteration could be revealed. Furthermore, nine mutations described here are novel, and none of the sequence changes were found in more than one ***: Our study describes for the f irst time the prevalence and spectrum of germline mismatch repair gene mutations in Hungarian HNPCC and suspected-HNPCC families. The results presented here suggest that clinical selection criteria should be relaxed and detection of genomic rearrangements should be included in genetic screening in this population.

Numerical modeling of salt tectonics is a rapidly evolving field; however, the constitutive equations to model long-term rock salt rheology in nature still remain controversial. Firstly, we built a database about the strain rate versus the differential stress through collecting the data from salt creep experiments at a range of temperatures（20–200 ℃） in laboratories. The aim is to collect data about salt deformation in nature, and the flow properties can be extracted from the data in laboratory ***, as an important preparation for salt tectonics modeling, a numerical model based on creep experiments of rock salt was developed in order to verify the specific model using the Abaqus package. Finally, under the condition of low differential stresses, the deformation mechanism would be extrapolated and discussed according to microstructure research. Since the studies of salt deformation in nature are the reliable extrapolation of laboratory data, we simplified the rock salt rheology to dislocation creep corresponding to power law creep（n = 5） with the appropriate material parameters in the salt tectonic modeling.

Ga-Al-MFI samples were synthesized in hydrothermal conditions from gels of composition 1.08CH3NH2- 0.134TPABr-1SiO2-xAl2O3-yGa2O3-40H2O at 175 ℃ for 7 days, with x = 0.005 and 0.0025, y = 0.005, 0,010 and 0.020. The samples were characterized by XRD, BET measurements, thermal analysis （TGA-DTA） atomic absorption and high resolution solid state MAS 27Al and 71Ga NMR measurements. The aromatization of propane was studied as catalytic test. The activity and selectivity of the catalysts were determined for benzene, toluene and xylenes on the one hand and for methane and ethane on the other hand. The most active sample was obtained with the highest Ga/AI ratio. For this sample, the BTX selectivity obtained by aromatization was always higher than the hydrocracking selectivity leading to methane and ethane. The relative amount of toluene was higher than that of benzene and ofxylenes. The samples were deactivated by coke formation that was revealed more severe for the most active sample,

AIM:To assess the combined effect of disease phenotype, smoking and medical therapy [steroid, azathioprine(AZA), AZA/biological therapy] on the probability of disease behavior change in a Caucasian cohort of patients with Crohn's disease(CD).METHODS:Three hundred and forty well-characterized, unrelated, consecutive CD patients were analyzed(M/F:155/185, duration:9.4 ± 7.5 years) with a complete clinical *** records including disease phenotype according to the Montreal classification, extraintestinal manifestations, use of medications and surgical events were analyzed *** were interviewed on their smoking habits at the time of diagnosis and during the regular follow-up ***:A change in disease behavior was observed in 30.8% of patients with an initially non-stricturing, non-penetrating disease behavior after a mean diseaseduration of 9.0 ± 7.2 *** a logistic regression analysis corrected for disease duration, perianal disease, smoking, steroid use, early AZA or AZA/ biological therapy use were independent predictors of disease behavior *** a subsequent Kaplan-Meier survival analysis and a proportional Cox regression analysis, disease location(P = 0.001), presence of perianal disease(P < 0.001), prior steroid use(P = 0.006), early AZA(P = 0.005) or AZA/biological therapy(P = 0.002), or smoking(P = 0.032) were independent predictors of disease behavior ***:Our data suggest that perianal disease, small bowel disease, smoking, prior steroid use, early AZA or AZA/biological therapy are all predictors of disease behavior change in CD patients.

The pathogenesis of inflammatory bowel disease （IBD） is only partially understood. Various environmental and host （e.g. genetic-, epithelial-, immune and nonimmune） factors are involved. It is a multifactorial polygenic disease with probable genetic heterogeneity. Some genes are associated with IBD itself, while others increase the risk of ulcerative colitis （UC） or Crohn＇ s disease （CD） or are associated with disease location and/or behaviour. This review addresses recent advances in the genetics of IBD. The article discusses the current information on the crosstalk between microbial and genetic factors （e.g. NOD2/CARD15, SLC22A46A5 and DLG5）. The genetic data acquired in recent years help in understanding the pathogenesis of IBD and can identify a number of potential targets for therapeutic intervention. In the future, genetics may help more accurately diagnose and predict disease course in IBD.

Whole-genome duplications(WGDs)are regarded as important drivers of macroevolution in plants(Clark and Donoghue,2018).However,there are few studies that test a connection between WGDs and evolutionary *** proper study of evolutionary patterns related to WGDs needs a well-resolved phylogeny,accurate dates of the WGD events,and historical reconstructions of the evolutionary novelties(Clark and Donoghue,2018).The advent of exhaustive phylogenomic studies is becoming a promising way to solve these issues,as wholegenome data can help clarify obscure phylogenetic relationships,evaluate patterns associated with WGDs,and track down the evolution of life-history traits that shaped plant lineages(Bellot et al.,2020).

Regularization of the level-set(LS)field is a critical part of LS-based topology optimization(TO)*** this is achieved by advancing the LS field through the solution of a Hamilton-Jacobi equation combined with a reinitialization *** approach,however,may limit the maximum step size and introduces discontinuities in the design ***,energy functionals and intermediate LS value penalizations have been *** paper introduces a novel LS regularization approach based on a signed distance field(SDF)which is applicable to explicit LSbased *** SDF is obtained using the heat method(HM)and is reconstructed for every design in the optimization *** governing equations of the HM,as well as the ones describing the physical response of the system of interest,are discretized by the extended finite element method(XFEM).Numerical examples for pro?blems modeled by linear elasticity,nonlinear hyperelasticity and the incompressible Navier-Stokes equations in two and three dimensions are presented to show the applicability of the proposed scheme to a broad range of design optimization problems.

Research on macroinvertebrate community structure in Andean rivers has been oriented towards describing patterns of alpha and gamma diversity by taking into account environmental predictors at local spatial scales(e.g.,micro-and mesohabitats).However,the patterns of beta diversity and the importance of landscape-scale variables have been evaluated to a lesser *** objective of this study was to describe the patterns of alpha and beta diversity of benthic macroinvertebrates in the Andean rivers of the Orinoco basin and their relationship with local and landscape environmental variation.A stratified random sampling of macroinvertebrates was carried out at 40 sites(comprising an altitudinal range of between 500 and 2900 m.a.s.l.),local and landscape variables were *** results showed that the variation of alpha diversity was influenced by local and landscape variables,which are directly and indirectly related to the contribution of sediments,substrate composition and flow velocity,providing a heterogeneity of *** beta diversity was explained by the combined effect of local and landscape *** the beta diversity phenomena,turnover was predominant while nestedness presented a minor contribution and both were explained in greater proportion by local descriptors and some landscape variables,specifically those of a geomorphological *** results concur with the view of an environmental and spatial hierarchy within the river habitat and highlight the influence of multiple scales on macroinvertebrate *** above suggests that both local and landscape scales must necessarily be considered for environmental management and the conservation of Andean lotic ecosystems.

AIM： To establish whether there are fundamental differences in the biochemistries of adenocarcinomas of the gastroesophageal junction （GEJ） and the squamous cell carcinomas of the lower third of the esophagus （LTE）. METHODS： Between February 1, 1997 and February 1, 2000, we obtained tissue samples at the moment of resection from 54 patients for biochemical analysis. The full set of data could be comprehensively analyzed in 47 of 54 patients＇ samples （81%）. Of these, 29 were adenocarcinomas of the GEJ Siewert type Ⅰ （n = 8）, type Ⅱ （n = 12）, type Ⅲ （n = 9）, and 18 presented as squamous cell carcinomas of the LTE. We evaluated the mean values of 11-lysosomal enzyme and 1-cytosol protease activities of the tumorous and surrounding mucosae as well as their relative activities, measured as the ratio of activity in tumor and normal tissues from the same patient. These data were further analyzed to establish the correlation with tumor localization, TNM stage （lymph-node involvement）, histological type （papillary, signet-ring cell, tubular）, state of differentiation （good, moderate, poor）, and survival （≤24 or ≥24 mo）. RESULTS： In adenocarcinomas, the activity of α-mannosidase （AMAN）, cathepsin B （CB） and dipeptidyl-peptidase Ⅰ （DPP Ⅰ） increased significantly as compared to the normal gastric mucosa. In squamous cell carcinomas of the esophagus, we also found a significant difference in the activity of cathepsin L and tripeptidyl-peptidase Ⅰ in addition to these three. There was a statistical correlation of AMAN, CB, and DPP Ⅰ activity between the level of differentiation of adenocarcinomas of the GEJ and lymph node involvement,because tumors with no lymph node metastases histologically confirmed as well-differentiated, showed a significantly lower activity. Many attempts on optimization of sorghum[Sorghum bicolor(L.)Moench]tissue culture induction media have been made,but the culture system remains with some bottlenecks compared to that of other *** study aimed at assessing the suitability of various induction media to produce embryogenic callus(yellow and friable)with high induction rates and reduced phenolic *** six culture medium modifications:3 based on Murashige and Skoog(MS)medium and one each based on Chu N6,Gamborg B5 and 190-2 media respectively were applied in the culture of mature embryos from 10 sorghum *** there was a genotype influence on the attainment of a yellow callus,friability of the callus was determined to be dependent on the culture medium and not the *** strength MS medium with 0.2 mg/l 2,4-D with 2.8 g/l Gelrite®as the gelling agent modified with 1.0 g/l KH_(2)PO_(4),1.0 g/l L-proline,1.0 g/l L-asparagine and 0.16 mg/l CuSO_(4)·5H_(2)O(type E)was found to be the most effective resulting in about 60%yellow coloured callus induction with 25%*** of CuSO_(4)·5H_(2)O,KH_(2)PO_(4),L-proline and L-asparagine significantly reduced the phenolic *** strength MS medium was observed to contribute to quality callus production when compared to full strength MS media modified with the *** half strength MS medium was also observed to suppress phenolic *** 190-2 produced the highest regeneration frequency(40%)among the 3-regeneration media *** results provide information on a suitable sorghum callus induction medium necessary for embryogenesis.
